    Reconstruction of compressed spectral imaging based on global structure and spectral correlation

    In this paper, a convolution sparse coding method based on global structure characteristics and spectral correlation is proposed for the reconstruction of compressive spectral images. The proposed method uses the convolution kernel to operate the global image, which can better preserve image structure information in the spatial dimension. To take full exploration of the constraints between spectra, the coefficients corresponding to the convolution kernel are constrained by the norm to improve spectral accuracy. And, to solve the problem that convolutional sparse coding is insensitive to low frequency, the global total-variation (TV) constraint is added to estimate the low-frequency components. It not only ensures the effective estimation of the low-frequency but also transforms the convolutional sparse coding into a de-noising process, which makes the reconstructing process simpler. Simulations show that compared with the current mainstream optimization methods (DeSCI and Gap-TV), the proposed method improves the reconstruction quality by up to 7 dB in PSNR and 10% in SSIM, and has a great improvement in the details of the reconstructed image

    Infection status and etiological characteristics of diarrheogenic Escherichia coli among diarrhea patients in sentinel hospitals of Fujian Province in 2019

    Objective To investigate the infectious status, virulence gene, molecular typing and antibiotic resistance of diarrheogenic Escherichia coli (DEC) in sentinel hospitals of Fujian Province in 2019. Methods Fluorescent polymerase chain reaction (PCR) was used to identify 210 fecal samples of diarrhea patients after isolation of E. coli according to GB 4789. 6-2016, and then pulsed field gel electrophoresis (PFGE) molecular traceability and antibiotic resistance test were conducted on the isolated DEC. Results Thirty two strains of bacteria were detected, with a detection rate of 15.2% (32/210). Among them, enteropathogenic E. coli (EPEC) accounted for 37.5% (12/32), enteroaggregative E. coli (EAEC) for 37.5% (12/32), and enterotoxigenic E. coli (ETEC) for 25.0% (8/32). The results of antibiotic resistance test showed that these 32 strains of bacteria were most resistant to ampicillin, with a resistance rate of 78.1% (25/32), followed by tetracycline and trimethoprim/sulfamethoxazole, with resistance rates of 62.5% (20/32) and 59.4% (19/32), respectively. The multiple antibiotic resistance rate was 50.0% (16/32). The results of PFGE showed that 32 strains of bacteria causing diarrhea were divided into 28 PFGE banding patterns. Among them, 12 strains of EPEC and 12 strains of EAEC were divided into 10 PFGE banding patterns, respectively, and 8 strains of ETEC were divided into 8 PFGE banding patterns. The results of cluster analysis showed that two groups of EPEC strains had 100.0% similar banding patterns, one group of EAEC strains had 100.0% similar banding patterns, and ETEC strains did not have completely consistent banding pattern. Conclusion EPEC and EAEC were the main pathogens of diarrhea in surveillance points of Fujian Province in 2019. The genetic diversity of the strains showed that the genetic relationship between them was relatively distant. The antibiotic resistance of DEC was severe, and the rate of multiple antibiotic resistance was high

    Validation of the digital health literacy assessment among the university students in China

    PurposeWith the development of the internet, digital health literacy (DHL) has become increasingly important for managing health. Consequently, various digital health literacy scales have been created for different groups. The purpose of this study was to verify the reliability and validity of the simplified Chinese version of the Digital Health Literacy Assessment (DHLA) scale among university students in China.MethodSnowball sampling was used to recruit the participants via an online platform (Wenjuan.com), and finally 304 university students were included in the survey. Demographic information and the status of DHL were collected through the online questionnaire. Cronbach’s alpha and split-half reliability were used to test the internal consistency of the scale, while the structural validity was verified by exploratory factor analysis and confirmatory factor analysis. Additionally, the convergence of the scale was tested by composite reliability (CR) and average variance extracted (AVE).ResultTwo dimensions were generated from 10 entries in the scale, named Self-rated Digital Health Literacy and Trust Degree of Online Health Information, respectively. The Cronbach’s alpha and split-half reliability of the total scale were 0.912 and 0.828, while the Cronbach’s alpha of the two dimensions were 0.913 and 0.830, respectively. The structural validity-related indexes of the scale met the standards (RMSEA = 0.079, GFI = 0.943, AGFI = 0.902, CFI = 0.971). In each dimension, the CR and AVE also reached critical values (CR > 0.7 and AVE > 0.5).ConclusionThe scale had high reliability and validity, indicating the simplified Chinese DHLA scale could be used to evaluate the DHL of university students in China

    The objective of the present study was to investigate the optimal nutritional requirements for mycelial growth and sporulation of entomopathogenic fungus Aschersonia aleyrodis Webber by orthogonal layout methods. Herein the order of effects of nutrient components on mycelial growth was tryptone > Ca2+ > soluble starch > folacin, corresponding to the following optimal concentrations: 1.58% Soluble Starch, 3.16% Tryptone, 0.2 mmol l-1 Ca2+ and 0.005% Folacin. The optimal concentration of each factors for sporulation was 1.16% lactose, 0.394% tryptone, 0.4 mmol l-1 Fe2+ and 0.00125% V B1, and the effects of medium components on sporulation were found to be in the order lactose > V B1 > Fe2+ > tryptone. Under the optimal culture conditions, the maximum production of mycelial growth achieved 20.05 g l-1 after 7 days of fermentation, while the maximum spore yield reached 5.23 ×10(10) spores l-1 after 22 days of cultivation. This is the first report on optimization of nutritional requirements and design of simplified semi-synthetic media for mycelial growth and sporulation of A. aleyrodis.<br>O objetivo deste estudo foi investigar as exigências nutricionais ótimas para o crescimento micelial e esporulação do fungo entomopatogênico Aschersonia aleyrodis Webber. A ordem dos efeitos dos nutrientes na multiplicação micelial foi triptona>Ca2+>amido solúvel>folacina, com as seguintes concentrações ótimas: amido solúvel 1,58%, tritona 3,16%, Ca2+ 0,2mmol.l-1 e folacina 0,005%. Para a esporulação, a concentração ótima de cada fator foi: lactose 1,16%, triptona 0,394%, Fe2+ 0,4mmol.l-1 e V B1 0,00125%, na seguinte ordem: lactose> V B1> Fe2+>tritona. Em condições ótimas de cultura, a produção máxima de micélio foi 20,05g.l-1 após 7 dias de fermentação, enquanto o rendimento máximo de esporos foi 5,23 x 10(10) esporos.l-1 após 22 dias de cultivo.     Entomopathogenic fungus Verticillium lecanii is a promising whitefly and aphid control agent. Chitinases secreted by this insect pathogen have considerable importance in the biological control of some insect pests. An endochitinase gene Vlchit1 from the fungus was cloned and overexpressed in Escherichia coli. The Vlchit1 gene not only contains an open reading frame (ORF) which encodes a protein of 423 amino acids (aa), but also is interrupted by three short introns. A homology modelling of Vlchit1 protein showed that the chitinase Vlchit1 has a (&#945;/&#946;)8 TIM barrel structure. Overexpression test and Enzymatic activity assay indicated that the Vlchit1 is a functional enzyme that can hydrolyze the chitin substrate, so the Vlchit1 gene can service as a useful gene source for genetic manipulation leading to strain improvement of entomopathogenic fungi or constructing new transgenic plants with resistance to various fungal and insects pests.<br>O fungo entomopatogênico Verticillium lecanii é um agente promissor no controle da mosca-branca e do pulgão. As quitinases secretadas por esse patógeno de insetos têm uma grande importância no controle biológico de doenças causadas por insetos. Um gene de endoquitinase Vlchit1 desse fungo foi clonado e expresso em Escherichia coli. O gene Vlchit contém não apenas um ORF que codifica uma proteína de 423 aminoácidos, mas também é interrompido por três pequenos introns. A modelagem de homologia da proteína Vlchit1indicou que a quitinase Vlchit1 tem uma estrutura (&#945;/&#946;) 8 TIM barrel. Testes de expressão e de atividade enzimática indicaram que Vlchit1 é uma enzima funcional que hidroliza quitina, portanto o gene Vlchit pode ser um gene útil para manipulação genética para melhoramento de cepas de fungos entomopatogênicos ou para a construção de novas plantas transgênicas com resistência a várias doenças causadas por fungos e insetos

    Risk Analysis of Viral Diseases in Infected Pig Farms during the Lockdown Period in China, January to May 2020

    Biosecurity plays a critical role in preventing and controlling the introduction and spread of infectious diseases. The COVID-19 pandemic in China triggered a nationwide lockdown policy which reduced most of the daily activities of people, but the pig industry was encouraged to ensure the pork supply. An investigation of biosecurity practices in intensive pig farms across several provinces in China was conducted in June 2020 via questionnaire to evaluate the factors that may pose viral diseases risk to the farms during the lockdown period from January to May 2020. A total of 50 farms in 12 provinces of China were engaged. Fourteen of them were classified as positive farms since at least one viral disease was presented during this period, including porcine reproductive and respiratory syndrome (seven farms), porcine epidemic diarrhea (three farms), and pseudorabies (one farm). The other three farms only reported their disease positive status but refused to release disease names. The overall farm level prevalence of viral disease was 28.0% (95%CI: 16.3&ndash;42.5%). A logistic regression model was built to identify risk/protective factors for farm positivity. In the multivariable logistic regression model, the risk factor of dead pig &lsquo;removal by the others&rsquo; (OR = 8.0, 95%CI: 1.5, 43.5) was found to be significantly associated with viral disease positivity. On-farm incineration pits are highly recommended to be the administered for the harmless treatment of dead pigs. This is not only crucial for controlling the transmission of viral diseases but also plays a key role in reducing activity in the illegal dead meat business. According to previous studies, factors such as adapting an all-in-all-out system, on-farm incineration pits, and requiring workers to wash their hands regularly would reduce the risk of virus transmission, even though these factors did not show significance in our study. The results of our study could help to design better surveillance strategies in China and other countries

    Characterization of cry1Cb3 and cry1Fb7 from Bacillus thuringiensis subsp. galleriae

    Two cry1-type genes encoding insecticidal crystal proteins (ICPs) were detected by PCR-RFLP and cloned from Bacillus thuringiensis subsp. galleriae 87. The nucleotide sequences were deposited in GenBank with accession numbers EU679501 and EU679502, and designated as cry1Fb7 and cry1Cb3 respectively by B. thuringiensis Delta- Endotoxin Nomenclature Committee. cry1Cb3 shared 99% homology with other cry1Cb genes. The existence of two additional stop codons indicated cry1Cb3 was a silent gene. The cry1Cb3 was 3531 bp with 38.98% G+C content and its first open reading frame (ORF) encoded a protein of 213 amino acid residues with a calculated molecular weight of 23.8 kDa and a predicted pI value of 4.63. Five amino acid sequence blocks (block 1, block 2, block 3, block 4 and block 5) were found in Cry1Cb3. Translation of cry1Fb7 revealed an ORF of 3525 bp with 39.12% G+C content and a protein with a calculated molecular weight of 133.2 kDa and a predicted pI value of 5.18. Cry1Fb7 had five amino acid sequence blocks (blocks 1, 2, 3, 4 and 5) and three domains (I, II and III), which consisted of 218 residues (Leu34 to Ala252), 197 residues (Thr257 to Asp454), and 138 residues (Ile464 to Glu600), respectively

    Characteristics of the MicroRNA Expression Profile of Exosomes Released by Vero Cells Infected with Porcine Epidemic Diarrhea Virus

    Exosomes are nanoscale vesicles actively secreted by a variety of cells. They contain regulated microRNA (miRNA), allowing them to function in intercellular communication. In the present study, the role of exosomal miRNAs in porcine epidemic diarrhea virus (PEDV) infection was investigated using exosomes isolated from Vero cells infected with PEDV. The results of transmission electron microscopy observation showed that the exosomes are spherical in shape, uniform in size, and negatively stained in the membrane. Nanoparticle tracking analysis showed that the average exosome particle size is 130.5 nm. The results of miRNA sequencing showed that, compared with the control group, a total of 115 miRNAs are abnormally expressed in the exosomes of infected cells. Of these, 80 miRNAs are significantly upregulated and 35 miRNAs are significantly downregulated. Functional annotation analysis showed that the differentially expressed miRNAs are associated with PEDV infection through interaction with the cAMP, Hippo, TGF-beta, HIF-1, FoxO, MAPK, and Ras signaling pathways. Thus, our findings provide important information about the effects of PEDV infection on exosomal miRNA expression and will aid the search for potential anti-PEDV drug candidates

    Association between dyslipidemia and risk of type 2 diabetes mellitus in middle-aged and older Chinese adults: a secondary analysis of a nationwide cohort

    Aims To evaluate the type 2 diabetes mellitus (T2DM) risk of individuals with different types of dyslipidaemia and compare the predictive value of distinct lipid parameters in predicting T2DM.Methods We conducted a secondary analysis of data from the China Health and Retirement Longitudinal Study (CHARLS). 17 708 individuals over 45 years old were interviewed, and 11 847 blood samples were collected at the baseline survey (2011–2012). Outcome of T2DM was confirmed during two follow-up surveys (2013–2014 and 2015–2016). The HRs and 95% CI of T2DM associated with dyslipidaemia were estimated by Cox proportional hazards regressions model. The discriminatory value of eight lipid parameters were compared by the area under the receiver operating characteristic (ROC) curve (AUC).Results A total of 7329 participants were enrolled in our analysis; during the mean follow-up time of 3.4 years, 387 (5.28%) participants developed new-onset diabetes. Compared with participants in normal lipid levels, the T2DM risk of those with hypercholesterolaemia, hypertriglyceridaemia and low high-density lipoprotein cholesterol (HDL-C) were significantly increased (HRs (95% CI) were 1.48 (1.11 to 1.96), 1.92 (1.49 to 2.46) and 1.67 (1.35 to 2.07), respectively). The AUCs of non-HDL-C (0.685, 95% CI 0.659 to 0.711), triglyceride (TG) (0.684, 95% CI 0.658 to 0.710), total cholesterol (TC)/HDL-C (0.685, 95% CI 0.659 to 0.712) and TG/HDL-C (0.680, 95% CI 0.654 to 0.706) were significantly (p&lt;0.005) larger than that of other lipid parameters.Conclusion Middle-aged and elderly adults with hypertriglyceridaemia, hypercholesterolaemia and low HDL-C were at higher risk for developing diabetes. Non-HDL-C, TG, TC/HDL and TG/HDL have greater performance than other lipid parameters in predicting T2DM incidence
