Selenium intake help prevent age-related cataract formation: Evidence from NHANES 2001–2008
IntroductionCataract is one of the leading causes of blindness and visual impairment, about 16 million people around the world. Trace elements play an important role in a variety of the processes in human body. This study aimed to investigate the association between daily dietary intake of trace elements and age-related cataract incidence based on data from the National Health and Nutrition Examination Survey (NHANES) 2001–2008.MethodsIron, zinc, copper, and selenium were conducted in this study among subjects aged 50 years and older for African Americans and 55 and older in US adults. Multivariate logistic regression analysis was used in different models to investigate the association of trace elements intake and cataract.ResultsAfter screening, 7,525 subjects were ultimately included in this study. A significant negative association was found between selenium intake and cataract incidence in adjusted models using multivariate logistic regression analysis (model 1: OR = 0.998, 95% CI = 0.997–1.000; model 2: OR = 0.997, 95% CI = 0.995–1.000; and model 3: OR = 0.998, 95% CI = 0.995–1.000). After dividing selenium intake into quintiles, significant negative associations between selenium intake and cataract were observed in the first quintile of model 3, the fourth and fifth quintiles of all models. In subgroup analyses adjusted for age and sex, a significant negative association was observed only in women aged 65–74 years.DiscussionOur study points out that maintaining daily dietary selenium intake at higher levels is helpful for cataract prevention, and that increasing daily dietary selenium intake in American women aged 65–74 years may contribute to the prevention of age-related cataract. The intakes of iron, zinc, copper may not be associated with age-related cataract
Relationship between high dose intake of vitamin B12 and glaucoma: Evidence from NHANES 2005–2008 among United States adults
ObjectiveGlaucoma has currently become the second leading cause of blindness in the world. Serum vitamin B12 level has been found to be involved in the development and progression of glaucoma. We performed the present study to confirm this association.MethodsThis cross-sectional study included 594 participants aged 40 years and older in the National Health and Nutrition Examination Survey (NHANES) from 2005 to 2008. Retinal imaging was performed using the Ophthalmic Digital Imaging system (Retinography) to assess the retina for the presence of features of glaucomatous lesions. Logistic regression models were used to assess the association between dietary vitamin intake and glaucoma.ResultsAfter screening, 594 subjects were finally included. Among all vitamin intakes, we observed significant differences between the two groups for vitamin B12 intake (5.93 vs. 4.77 mg, p = 0.033). According to the logistic regression results, the intake of vitamin B12 was significantly positively associated with glaucoma (model 1: OR = 1.078, 95% CI = 1.019–1.141; model 2: OR = 1.092, 95% CI = 1.031–1.158; model 3: OR = 1.092, 95% CI = 1.029–1.158). After performing a quantile regression, we observed a significant positive association between vitamin B12 intake and incident glaucoma in the fourth quartile (model 1: OR = 1.133, 95% CI = 1.060–1.210; model 2: OR = 1.141, 95% CI = 1.072–1.215; model 3: OR = 1.146, 95% CI = 1.071–1.226).ConclusionsTherefore, the above results, high-dose intake of vitamin B12 may promote the development of glaucoma
MD-ALL: an integrative platform for molecular diagnosis of B-acute lymphoblastic leukemia
B-acute lymphoblastic leukemia (B-ALL) consists of dozens of subtypes defined by distinct gene expression profiles (GEP) and various genetic lesions. With the application of transcriptome sequencing (RNA sequencing [RNA-seq]), multiple novel subtypes have been identified, which lead to an advanced B-ALL classification and risk-stratification system. However, the complexity of analyzing RNA-seq data for B-ALL classification hinders the implementation of the new B-ALL taxonomy. Here, we introduce Molecular Diagnosis of Acute Lymphoblastic Leukemia (MD-ALL), an integrative platform featuring sensitive and accurate B-ALL classification based on GEP and sentinel genetic alterations from RNA-seq data. In this study, we systematically analyzed 2,955 B-ALL RNA-seq samples and generated a reference dataset representing all the reported B-ALL subtypes. Using multiple machine learning algorithms, we identified the feature genes and then established highly sensitive and accurate models for B-ALL classification using either bulk or single-cell RNA-seq data. Importantly, this platform integrates multiple aspects of key genetic lesions acquired from RNA-seq data, which include sequence mutations, large-scale copy number variations, and gene rearrangements, to perform comprehensive and definitive B-ALL classification. Through validation in a hold-out cohort of 974 samples, our models demonstrated superior performance for B-ALL classification compared with alternative tools. Moreover, to ensure accessibility and user-friendly navigation even for users with limited or no programming background, we developed an interactive graphical user interface for this MD-ALL platform, using the R Shiny package. In summary, MD-ALL is a user-friendly B-ALL classification platform designed to enable integrative, accurate, and comprehensive B-ALL subtype classification. 3% diquafosol sodium eye drops in Chinese patients with dry eye: a phase IV study
IntroductionThe efficacy and safety of 3% diquafosol sodium eye drops in Chinese patients with dry eye in the real-world setting remains unclear.Methods3099 patients with dry eye symptoms were screened according to Asia Dry Eye Society latest recommendation. Among them, 3000 patients were enrolled for a phase IV study. We followed up with multiple clinical characteristics including corneal fluorescein staining, tear break up time, Schirmer’s tests, visual acuity, intraocular pressure, and others. The follow ups were performed at baseline, 2 weeks and 4 weeks after treatment.ResultsBased on the results of corneal fluorescein staining and tear break up time, all age and gender subgroups exhibited obvious alleviation of the symptoms among the patients with dry eye, and the data in elderly group showed the most significant alleviation. All the adverse drug reactions (ADRs, 6.17%) were recorded, among which 6% local ocular ADRs were included. Meanwhile, mild ADRs (91.8%) accounted for the most. Most of the ADRs (89.75%) got a quick and full recovery, with an average time at 15.6 days. 1.37% of patients dropped out of the study due to ADRs.DiscussionThe use of 3% diquafosol sodium eye drop is effective and safe in the treatment of dry eye, with a low incidence of ADRs showing mild symptoms. Research of Improved FP-Growth Algorithm in Association Rules Mining
Association rules mining is an important technology in data mining. FP-Growth (frequent-pattern growth) algorithm is a classical algorithm in association rules mining. But the FP-Growth algorithm in mining needs two times to scan database, which reduces the efficiency of algorithm. Through the study of association rules mining and FP-Growth algorithm, we worked out improved algorithms of FP-Growth algorithm—Painting-Growth algorithm and N (not) Painting-Growth algorithm (removes the painting steps, and uses another way to achieve). We compared two kinds of improved algorithms
with FP-Growth algorithm. Experimental results show that Painting-Growth algorithm is more than 1050 and N Painting-Growth algorithm is less than 10000 in data volume; the performance of the two kinds of improved algorithms is better than that of FP-Growth algorithm
Early Expression of Tet1 and Tet2 in Mouse Zygotes Altered DNA Methylation Status and Affected Embryonic Development
Ten-eleven translocation (Tet) dioxygenases can induce DNA demethylation by catalyzing 5-methylcytosine(5mC) to 5-hydroxymethylcytosine(5hmC), and play important roles during mammalian development. In mouse, Tet1 and Tet2 are not expressed in pronucleus-staged embryos and are not involved in the genomic demethylation of early zygotes. Here, we investigated the influence of Tet1 and Tet2 on methylation of parental genomes by ectopically expressing Tet1 and Tet2 in zygotes. Immunofluorescence staining showed a marked 5hmC increase in the maternal pronucleus after injection of Tet1 or Tet2 mRNA into zygotes. Whole-genome bisulfite sequencing further revealed that Tet2 greatly enhanced the global demethylation of both parental genomes, while Tet1 only promoted the paternal demethylation. Tet1 and Tet2 overexpression altered the DNA methylation across genomes, including various genic elements and germline-specific differently methylated regions. Tet2 exhibited overall stronger demethylation activity than Tet1. Either Tet1 or Tet2 overexpression impaired preimplantation embryonic development. These results demonstrated that early expression of Tet1 and Tet2 could substantially alter the zygotic methylation landscape and damage embryonic development. These findings provide new insights into understanding the function of Tet dioxygenases and the mechanism of DNA methylation in relation to embryogenesis
Influence of Uncomplicated Phacoemulsification on Central Macular Thickness in Diabetic Patients: A Meta-Analysis
<div><p>Objective</p><p>To evaluate the effect of uncomplicated phacoemulsification on central macular thickness (CMT) and best corrected visual acuity (BCVA) in both diabetic patients without diabetic retinopathy (DR) and diabetic patients with mild to moderate non-proliferative diabetic retinopathy (NPDR).</p><p>Methods</p><p>Potential prospective observational studies were searched through PubMed and EMBASE. Standardized mean difference (SMD) and 95% confidence interval (CI) for changes in CMT and BCVA were evaluated at postoperative 1, 3 and 6 months. The pooled effect estimates were calculated in the use of a random-effects model.</p><p>Results</p><p>A total of 10 studies involving 190 eyes of diabetic patients without diabetic retinopathy and 143 eyes of diabetic patients with NPDR were identified. CMT values demonstrated a statistically significant increase after uncomplicated phacoemulsification at 1 month (SMD, -0.814; 95%CI, -1.230 to -0.399), 3 months (SMD, -0.565; 95%CI, -0.927 to -0.202) and 6 months (SMD, -0.458; 95%CI, -0.739 to -0.177) in diabetic patients with NPDR. There was no statistical difference in CMT values at postoperative 1 month (SMD, -1.206; 95%CI, -2.433 to 0.021)and no statistically significant increase in CMT values at postoperative3 months (SMD, -0.535; 95%CI, -1.252 to 0.182) and 6 months (SMD, -1.181; 95%CI, -2.625 to 0.263) in diabetic patients without DR.BCVA was significantly increased at postoperative 1 month (SMD, 1.149; 95%CI, 0.251 to 2.047; and SMD,1.349; 95%CI, 0.264 to 2.434, respectively) and 6 months (SMD, 1.295; 95%CI, 0.494 to 2.096; and SMD, 2.146; 95%CI, 0.172 to 4.120, respectively) in both diabetic patients without DR and diabetic patients with NPDR. Sensitivity analysis showed that the results were relatively stable and reliable.</p><p>Conclusion</p><p>Uncomplicated phacoemulsification in diabetic patients with mild to moderate NPDR seemed to influence significantly the subclinical thickening of the macular zones at postoperative 1, 3 and 6 months compared with diabetic patients without DR. BCVA was significantly improved in both diabetic patients without DR and diabetic patients with mild to moderate NPDR.</p></div
Contrast Sensitivity and Spherical Aberration in Eyes Implanted with AcrySof IQ and AcrySof Natural Intraocular Lens: the Results of a Meta-Analysis
<div><p>Background</p><p>To systematically evaluate the visual performance of aspheric AcrySof IQ and spherical AcrySof Natural intraocular lens (IOL) after cataract surgery.</p> <p>Methodology/Principal Findings</p><p>Potential randomized controlled trials (RCTs) that involved implanting AcrySof IQ and AcrySof Natural were searched from PubMed, Web of science, EMBASE, Chinese Science and Technology Periodicals Databases and Cochrane Central Register of Controlled Trials. The methodological quality of the studies was assessed by the Jadad method. Standardized mean differences (SMDs) with 95% confidence intervals (CIs) of best-corrected visual acuity (BCVA), contrast sensitivity and spherical aberration were pooled using a random-effects model. Seven studies were identified and analyzed to compare AcrySof IQ (236 eyes) with AcrySof Natural (232 eyes) after phacoemulsification. There was no significant difference in postoperative BCVA between AcrySof IQ and AcrySof Natural (<i>p</i> =0.137) after a follow up of 3 months. For contrast sensitivity, these differences reached statistical significance under photopic conditions at two spatial frequencies (3 cycles per degree (cpd), 6 cpd, 12 cpd, and 18 cpd; <i>p</i> =0.022, <i>p</i> =0.017, <i>p</i> = 0.065, and <i>p</i>=0.191, respectively) and under mesopic conditions at three spatial frequencies (3 cpd, 6 cpd, 12 cpd, and 18 cpd; <i>p</i> =0.007, <i>p</i> =0.033, <i>p</i> =0.030, and <i>p</i> =0.080, respectively). Eyes with AcrySof IQ also had statistically significant less spherical aberration than eyes with AcrySof Natural (<i>p</i><0.001). Sensitivity analysis showed that the results were relatively stable and reliable.</p> <p>Conclusions/Significance</p><p>The overall findings indicate that AcrySof IQ with a modified aspheric surface induced significantly less spherical aberration than AcrySof Natural. Contrast sensitivity in eyes with AcrySof IQ is better than that in eyes with AcrySof Natural, especially under mesopic conditions.</p> </div
