    Study of imbibition in various geometries using phase field method

    Phase field method has been widely utilized to study multiphase flow problems, but has seldom been applied to the study of imbibition. Previous methods used to simulate imbibition, such as moving mesh method, need to specify capillary pressure as a boundary condition a priori, whereas phase field method can calculate capillary pressure automatically for various geometries. Therefore, phase field method would be a versatile tool for the study of imbibition in various geometries. In this paper, phase field method is employed to solve dynamical imbibition problem in various geometries, including straight tube, conical tube and structures in which the topology changes. The variation of the imbibition height with respect to time from phase field simulation is verified with theoretical predictions from Lucas-Washburn law in a straight capillary tube with three gravitational scenarios. In addition, the capillary pressure and velocity field are found to be consistent with Laplace-Young equation and Hagen-Poiseuille equation in various geometries.     Regulation of Ferredoxin-NADP+ Oxidoreductase to Cyclic Electron Transport in High Salinity Stressed Pyropia yezoensis

    Pyropia yezoensis can survive the severe water loss that occurs during low tide, making it an ideal species to investigate the acclimation mechanism of intertidal seaweed to special extreme environments. In this study, we determined the effects of high salinity on photosynthesis using increasing salinity around algal tissues. Both electron transport rates, ETR (I) and ETR (II), showed continuous decreases as the salinity increased. However, the difference between these factors remained relatively stable, similar to the control. Inhibitor experiments illustrated that there were at least three different cyclic electron transport pathways. Under conditions of severe salinity, NAD(P)H could be exploited as an endogenous electron donor to reduce the plastoquinone pool in Py. yezoensis. Based on these findings, we next examined how these different cyclic electron transport (CETs) pathways were coordinated by cloning the gene (HM370553) for ferredoxin-NADP+ oxidoreductase (FNR). A phylogenetic tree was constructed, and the evolutionary relationships among different FNRs were evaluated. The results indicated that the Py. yezoensis FNR showed a closer relationship with cyanobacterial FNR. The results of both real-time polymerase chain reaction and western blotting showed that the enzyme was upregulated under 90–120‰ salinity. Due to the structure-function correlations in organism, Py. yezoensis FNR was proposed to be involved in NAD(P)H-dependent Fd+ reduction under severe salinity conditions. Thus, through the connection between different donors bridged by FNR, electrons were channeled toward distinct routes according to the different metabolic demands. This was expected to make the electron transfer in the chloroplasts become more flexible and to contribute greatly to acclimation of Py. yezoensis to the extreme variable environments in the intertidal zone

    Abstract: Molecular imaging allows direct visualization of targets and characterization of cellular pathways, as long as a high signal/background ratio can be achieved, which requires a sufficient amount of probes to accumulate in the imaging region. The Asn-Gly-Arg (NGR) tripeptide selected by phage display can specifically target tumor vasculature. Recognizing the aminopeptidase N (APN or CD13) receptor on the membrane of tumor cells, the peptide can be further internalized into cytoplasma by the endosomal pathway. Hence NGR can serve as an ideal candidate for tumor imaging, once it is conjugated with fluorescent or radiolabeled imaging probes. Herein, we highlight some recent developments of NGR peptide based imaging of tumors. Although still in the preliminary stage, some NGR probes have shown potential as promising agents in future clinical applications

    Baicalin Weakens Staphylococcus aureus Pathogenicity by Targeting Sortase B

    Staphylococcus aureus (S. aureus) is a human and other animal pathogen that contributes to the primary etiology of nosocomial pneumonia, a disease with high mortality rates and costs. Treatment of multidrug-resistant S. aureus infection is extremely challenging, and new therapeutic strategies beyond antibiotic treatment are needed. Anti-virulence agents that specifically target the molecular determinants of virulence may be a novel method for treating drug-resistant nosocomial infections. Sortase B (SrtB) is a crucial virulence factor in S. aureus and plays an important role during infection. In this study, we find that baicalin suppresses the activity of SrtB. Minimum inhibitory concentration and growth curve assays confirmed that baicalin has no anti-S. aureus properties. We performed live/dead, lactate dehydrogenase (LDH), adherence, and enzyme-linked immunosorbent assays to confirm that baicalin reduced human alveolar epithelial A549 cell injury caused by S. aureus, reduced the adherence of S. aureus to A549 cells, and significantly attenuated the inflammatory response of mouse macrophage J774 cells to S. aureus. Additionally, we were able to elucidate the binding mechanics and identify the interacting sites of baicalin and SrtB via a molecular dynamics simulation, site-directed mutagenesis, and fluorescence spectroscopy quenching. Finally, we confirmed that baicalin directly binds to the active center of SrtB, and the residues Asn92 and Tyr128 perform an important function in the interaction of SrtB and baicalin. Taken together, these data indicate that baicalin is a promising candidate to combat S. aureus infections

    Sustainable and scalable in-situ synthesis of hydrochar-wrapped Ti3AlC2-derived nanofibers as adsorbents to remove heavy metals

    To ensure a sustainable future, it is imperative to efficiently utilize abundant biomass to produce such as platform chemicals, transport fuels, and other raw materials; hydrochar is one of the promising candidates derived by hydrothermal carbonization of biomass in pressurized hot water. The synthesis of “hydrochar-wrapped Ti3AlC2-derived nanofibers” was successfully achieved by a facile one-pot hydrothermal reaction using glucose as the hydrochar precursor. Meanwhile, cellulose and pinewood sawdust as raw materials were also investigated. Products were characterized by XRD, N2 adsorption-desorption isotherms, SEM, TEM and FT-IR to investigate their crystal structures, textural properties, morphologies, and surface species. In the adsorption test to remove Cd(II) and Cu(II) in aqueous solution, hydrochar-wrapped nanofibers outperformed pure nanofibers derived from Ti3AlC2, hydrothermal carbon derived from glucose and commercial activated carbon. Finally, the regeneration, sorption kinetics, and possible adsorption mechanism were also explored

    A Hybrid Algorithm of Traffic Accident Data Mining on Cause Analysis

    Road traffic accident databases provide the basis for road traffic accident analysis, the data inside which usually has a radial, multidimensional, and multilayered structure. Traditional data mining algorithms such as association rules, when applied alone, often yield uncertain and unreliable results. An improved association rule algorithm based on Particle Swarm Optimization (PSO) put forward by this paper can be used to analyze the correlation between accident attributes and causes. The new algorithm focuses on characteristics of the hyperstereo structure of road traffic accident data, and the association rules of accident causes can be calculated more accurately and in higher rates. A new concept of Association Entropy is also defined to help compare the importance between different accident attributes. T-test model and Delphi method were deployed to test and verify the accuracy of the improved algorithm, the result of which was a ten times faster speed for random traffic accident data sampling analyses on average. In the paper, the algorithms were tested on a sample database of more than twenty thousand items, each with 56 accident attributes. And the final result proves that the improved algorithm was accurate and stable

    Phloretin Attenuates Listeria monocytogenes Virulence Both In vitro and In vivo by Simultaneously Targeting Listeriolysin O and Sortase A

    The critical roles of sortase A (SrtA) and listeriolysin O (LLO) in Listeria monocytogenes pathogenicity render these two virulence factors as ideal targets for the development of anti-virulence agents against L. monocytogenes infection. Additionally, the structures of SrtA and LLO are highly conserved among the members of sortase enzyme family and cholesterol dependent toxin family. Here, phloretin, a natural polyphenolic compound derived from apples and pears that has little anti-L. monocytogenes activity, was identified to simultaneously inhibit LLO expression and neutralize SrtA catalytic activity. Phloretin neutralized SrtA activity by causing a conformational change in the protein's active pocket, which prevented engagement with its substrate. Treatment with phloretin simultaneously reduced L. monocytogenes invasion into host cells and blocked the escape of vacuole-entrapped L. monocytogenes into cytoplasm. Further, L. monocytogenes-infected mice that received phloretin showed lower mortality, decreased bacterial burden and reduced pathological injury. Our results demonstrate that phloretin is a promising anti-infective therapeutic for infections caused by L. monocytogenes due to its simultaneous targeting of SrtA and LLO, which may result in fewer side effects than those caused by other antibiotics
