HETEROLOGOUS EXPRESSION AND CHARACTERIZATION OF MANNITOL-1-PHOSPHATE DEHYDROGENASE FROM THE BASIODIOMYCETE PHOLIOTA NAMEKO
ABSTRACT The gene encoding mannitol-1-phosphate dehydrogenase, mpd, which has similar function with alcohol dehydrogenase, has been sequenced and characterized from the basiodiomycete Pholiota nameko. The coding region of mpd was composed of 2992 bp and found to encode a polypeptide of 359 amino acids that has similarity with Laccaria bicolor. To evaluate the expression level of mannitol-1-phosphate from P. nameko, mpd cDNA was inserted into pCold shock vector and expressed in host BL21 (DE3) by 24 h induction with 1 mM isopropyl 1-thio-β-Dgalactopyranoside at 15 O C after 2 h growth at 37 O C. The cross-reactivity of the enterovirus 71 to human brain tissue and identification of the cross-reactivity related fragments
<p>Abstract</p> <p>Background</p> <p>EV71 occasionally cause a series of severe neurological symptoms, including aseptic meningitis, encephalitis, and poliomyelitis-like paralysis. However, the neurological destruction mechanism was remained to be clarified. This study described the cross reaction between EV71 induced IgG and human brain tissue.</p> <p>Results</p> <p>Cross reaction of the IgG from 30 EV71 infected patients' sera to human tissues of cerebra was observed, which suggested that some EV71 antigens could induce IgG cross-reactivity to human cerebra. To identify the regions of EV71 virus that containing above antigens, the polypeptide of virus was divided into 19 peptides by expression in prokaryotes cell. Mouse anti-sera of these peptides was prepared and applied in immunohistochemical staining with human adult and fetus brain tissue, respectively. The result indicated the 19 peptides can be classified into three groups: strong cross-reactivity, weak cross-reactivity and no cross-reactivity with human brain tissue according the cross reaction activity. Then, the increased Blood Brain Barrier (BBB) permeability and permits IgG entry in neonatal mice after EV71 infection was determined.</p> <p>Conclusion</p> <p>EV71 induced IgG could enter BBB and cross-reacted with brain tissue in EV71 infected neonatal mice, and then the peptides of EV71 that could induce cross-reactivity with brain tissue were identified, which should be avoided in future vaccine designing.</p
Post-transplantation cyclophosphamide combined with tacrolimus and low-dose post-engraftment anti-thymoglobulin as GVHD prophylaxis for patients undergoing peripheral blood stem cell transplantation from haploidentical family donor: A single center analysis
IntroductionPost-transplantation cyclophosphamide (PT-Cy) use is a recent graft-versus-host disease (GVHD) prophylaxis strategy for patients undergoing allogeneic stem cell transplantation (allo-HSCT). PT-Cy combined with two immunosuppressants is now widely used after haplo-identical (haplo) and HLA-matched peripheral blood stem cell (PBSC) transplantations with promising GVHD and relapsefree survival (GRFS) probabilities. Although appealing, these results may benefit from improvement notably outside matched sibling donor transplantation, and should be investigated in various ethnic populations.MethodsTherefore, we report our experience of GVHD prophylaxis regimen combining PT-Cy and tacrolimus with addition of post-engraftment low-dose anti-thymocyte globulin (ATG) in allogeneic stem cell transplantation from haplo-identical donors (Haplo). Sixtyseven patients were included in the analysis. All patients received myeloablative or intensified sequential conditioning regimen.ResultsThe median follow-up was 521 (range, 10~991) days. The cumulative incidences of 100-day grade II-IV acute GVHD was 14.9±4.4%, and no case of grade III-IV acute GVHD was documented. The cumulative incidences of 2-yearchronic GVHD and moderate-to-severe chronic GVHD were 25.4±5.4% and 11.9±4%, respectively. The non-relapse mortality at day+100 and 2year were 7.5±3.2% and 9.0±3.5%, respectively. The cumulative incidence of relapse at 2year was 16±6.4%. The 2-year probability of DFS and OS were 73.8% (95%CI, 61.5~88.4%) and 72.5% (95% CI, 57.1~92.1%), respectively. The 2-year GRFS was estimated as 63.6% (95%CI, 50.6~80%).DiscussionOur results suggested that a combination of PT-Cy, tacrolimus, and low-dose post-engraftment ATG was a promising GVHD prophylaxis with low incidence of acute GVHD in the haplo-transplantation setting

Superconducting Diode Effect and Large Magnetochiral Anisotropy in T-MoTe Thin Film
In the absence of time-reversal invariance, metals without inversion symmetry
may exhibit nonreciprocal charge transport -- a magnetochiral anisotropy that
manifests as unequal electrical resistance for opposite current flow
directions. If superconductivity also sets in, the charge transmission may
become dissipationless in one direction while remaining dissipative in the
opposite, thereby realizing a superconducting diode. Through both
direct-current and alternating-current measurements, we study the nonreciprocal
effects in thin films of the noncentrosymmetric superconductor
T-MoTe\textsubscript{2} with disorders. We observe nonreciprocal
superconducting critical currents with a diode efficiency close to 20\%~, and a
large magnetochiral anisotropy coefficient up to
\SI{5.9e8}{\per\tesla\per\ampere}, under weak out-of-plane magnetic field in
the millitesla range. The great enhancement of rectification efficiency under
out-of-plane magnetic field is likely abscribed to the vortex ratchet effect,
which naturally appears in the noncentrosymmetric superconductor with
disorders. Intriguingly, unlike the finding in Rashba systems, the strongest
in-plane nonreciprocal effect does not occur when the field is perpendicular to
the current flow direction. We develop a phenomenological theory to demonstrate
that this peculiar behavior can be attributed to the asymmetric structure of
spin-orbit coupling in T-MoTe\textsubscript{2}. Our study highlights how
the crystallographic symmetry critically impacts the nonreciprocal transport,
and would further advance the research for designing the superconducting diode

Observation of a ppb mass threshoud enhancement in \psi^\prime\to\pi^+\pi^-J/\psi(J/\psi\to\gamma p\bar{p}) decay
The decay channel
is studied using a sample of events collected
by the BESIII experiment at BEPCII. A strong enhancement at threshold is
observed in the invariant mass spectrum. The enhancement can be fit
with an -wave Breit-Wigner resonance function with a resulting peak mass of
and a
narrow width that is at the 90% confidence level.
These results are consistent with published BESII results. These mass and width

are observed for the first time using
\psip events accumulated at the BESIII detector at the BEPCII
collider. The branching fractions are measured to be , , and , for , , and ,
respectively. The observation of decays into a pair of vector
mesons , and indicates that the hadron
helicity selection rule is significantly violated in decays. In
addition, the measurement of gives the rate of doubly
OZI-suppressed decay. Branching fractions for and

First Observation of the Decays chi_{cJ} -> pi^0 pi^0 pi^0 pi^0
We present a study of the P-wave spin -triplet charmonium chi_{cJ} decays
(J=0,1,2) into pi^0 pi^0 pi^0 pi^0. The analysis is based on 106 million
\psiprime decays recorded with the BESIII detector at the BEPCII electron
positron collider. The decay into the pi^0 pi^0 pi^0 pi^0 hadronic final state
is observed for the first time. We measure the branching fractions B(chi_{c0}
-> pi^0 pi^0 pi^0 pi^0)=(3.34 +- 0.06 +- 0.44)*10^{-3}, B(chi_{c1} -> pi^0 pi^0
pi^0 pi^0)=(0.57 +- 0.03 +- 0.08)*10^{-3}, and B(chi_{c2} -> pi^0 pi^0 pi^0
pi^0)=(1.21 +- 0.05 +- 0.16)*10^{-3}, where the uncertainties are statistical

Study of radiative decays into a vector meson
The decays () are studied with
a sample of radiative \psip\to\gamma\chi_{cJ} events in a sample of
(1.06\pm0.04)\times 10^{8} \psip events collected with the BESIII detector.
The branching fractions are determined to be: , , and . The decay is observed for the first time. Upper limits at the 90% confidence
level on the branching fractions for and \chict decays into these
final states are determined. In addition, the fractions of the transverse
polarization component of the vector meson in decays
are measured to be for , for , and for , respectively. The first errors are statistical and the second
