    Relationship between adherence to anti-diabetic medication and depression among patients with diabetes mellitus in three selected Chinese hospitals

    Purpose: To determine the relationship between adherence to anti-diabetic medication and depression among patients with diabetes mellitus in three hospitals in Chinese.Methods: This research utilized a quantitative and descriptive design, and included 200 diabetic patients who fulfilled the inclusion criteria for recruitment through a convenient sampling technique. The study applied Beckā€™s depression inventory II scale for assessment of depression, and a questionnaire for adherence to anti-diabetic medication.Results: A total of 64 (32 %) participants had diabetes for 6 to 10 years. There was a high level of adherence to anti-diabetic medication in 96 patients (48 %); 74 participants (37 %) had moderate adherence to anti-diabetic medication, while 30 patients (15 %) had low adherence. A majority of the patients (181, 90.5 %) had no depression. Six (6) patients (3 %) had mild mood disturbance, 2 patients (1 %) had borderline clinical depression, while 11patients (5.5 %) experienced moderate depression.Adherence to anti-diabetic medication was not associated with depression (p = 0.068). However, depression was associated with age Ėƒ 50 years (p = 0.041), female sex (p = 0.043), long duration of illness (> 5-years) (p = 0.048), and presence of one or more comorbidities (p = 0.049).Conclusion: There was no association between adherence to anti-diabetic medication and depression among diabetic patients.     Double suicide genes selectively kill human umbilical vein endothelial cells

    <p>Abstract</p> <p>Background</p> <p>To construct a recombinant adenovirus containing CDglyTK double suicide genes and evaluate the killing effect of the double suicide genes driven by kinase domain insert containing receptor (KDR) promoter on human umbilical vein endothelial cells.</p> <p>Methods</p> <p>Human KDR promoter, <it>Escherichia coli </it>(<it>E. coli</it>) cytosine deaminase (CD) gene and the herpes simplex virus-thymidine kinase (TK) gene were cloned using polymerase chain reaction (PCR). Plasmid pKDR-CDglyTK was constructed with the KDR promoter and CDglyTK genes. A recombinant adenoviral plasmid AdKDR-CDglyTK was then constructed and transfected into 293 packaging cells to grow and harvest adenoviruses. KDR-expressing human umbilical vein endothelial cells (ECV304) and KDR-negative liver cancer cell line (HepG2) were infected with the recombinant adenoviruses at different multiplicity of infection (MOI). The infection rate was measured by green fluorescent protein (GFP) expression. The infected cells were cultured in culture media containing different concentrations of prodrugs ganciclovir (GCV) and/or 5-fluorocytosine (5-FC). The killing effects were measured using two different methods, i.e. annexin V-FITC staining and terminal transferase-mediated dUTP nick end-labeling (TUNEL) staining.</p> <p>Results</p> <p>Recombinant adenoviruses AdKDR-CDglyTK were successfully constructed and they infected ECV304 and HepG2 cells efficiently. The infection rate was dependent on MOI of recombinant adenoviruses. ECV304 cells infected with AdKDR-CDglyTK were highly sensitive to GCV and 5-FC. The cell survival rate was dependent on both the concentration of the prodrugs and the MOI of recombinant adenoviruses. In contrast, there were no killing effects in the HepG2 cells. The combination of two prodrugs was much more effective in killing ECV304 cells than GCV or 5-FC alone. The growth of transgenic ECV304 cells was suppressed in the presence of prodrugs.</p> <p>Conclusion</p> <p>AdKDR-CDglyTK/double prodrog system may be a useful method for suppressing tumor angiogenesis.</p

    Phylogeny of the genus Morus (Urticales: Moraceae) inferred from ITS and trnL-F sequences

    Both nuclear ribosomal ITS and chloroplast trnL-F sequences were acquired from 13 mulberry genotypes belonging to nine species and three varieties, and one paper mulberry. The later belongs to genus B. papyrifera, designed as outgroup, and were analyzed. Within the genus Morus, the sequence diversity of ITS was much higher than that of trnL-F. The results of phylogenetic analyses based on these data (separately or combined) show that the genus Morus is monophyletic group. Strict consensus tree obtained through the Neighbor-joining method can be divided into five major clades in the genus Morus, according to combined sequence data. M. bombycis, M. alba var. venose formed clades A and B, respectively. Clade C comprises of 5 species; M. rotundiloba, M. atropurpurea, M. mongolica, M. australi, and M. mongolica var. diabolica. Clade D comprises of 3 species; M. wittiorum, M. laevigata, and M. alba. Clade E comprises of 2 species; M. multicaulis, and M.alba var. macrophylla.     Deviation between willingness and actual behavior regarding community participation in protected areas: A case study in Shengjin lake national nature reserve in China

    Introduction: The deviation between high willingness and poor actual behaviors in community participation has become an obstacle to achieving effective management and resource protection of protected areas. Therefore, it is necessary to investigate this deviation and its influencing factors.Methods: Based on a field survey of community residents in the Shengjin Lake National Nature Reserve (SJLNNR) in Anhui Province, China, this study uses a Logistic-ISM model to analyze the key factors influencing the deviation and the hierarchical structure supporting those key factors.Results: There is a significant deviation between participation willingness and actual behavior in SJLNNR. This deviation is characterized by eight key factors. Among these, awareness of the necessity of establishing SJLNNR is a direct factor at the surface level. Participation of important or recognizable people; average annual family income; understanding of environmental protection laws and regulations; realization of environmental improvement effect; satisfaction with SJLNNR authority; and future earnings expectations are all intermediate indirect factors. Furthermore, resident member is a fundamental factor. Resident members refer to those who live at home for six months or more during a calendar year.Discussion: These results indicate that there are three possible practical reasons for the deviation: 1) The lack of young and middle-aged adults is the root cause. 2) Inadequate ecological compensation is the direct trigger. 3) Individual internal psychological factors have significant effects. Furthermore, this study provides policy implications for converting willingness into actual participation in the community and promoting sustainable development

    Therapeutic Potential of Nitrogen Mustard Based Hybrid Molecules

    As medicine advances, cancer is still among one of the major health problems, posing significant threats to human health. New anticancer agents features with novel scaffolds and/or unique mechanisms of action are highly desirable for the treatment of cancers, especially those highly aggressive and drug-resistant ones. Nitrogen mustard has been widely used as an anticancer drug since the discovery of its antitumor effect in the 1942. However, the lack of selectivity to cancer cells restricts the wide usage of a mass of nitrogen mustard agents to achieve further clinical significance. Discovery of antitumor hybrids using nitrogen mustards as key functional groups has exhibited enormous potential in the drug development. Introduction of nitrogen mustards resulted in improvement in the activity, selectivity, targetability, safety, pharmacokinetics and pharmacodynamics properties of corresponding lead compounds or agents.     Online dosimetric evaluation of larynx SBRT: A pilot study to assess the necessity of adaptive replanning

    PURPOSE: We have initiated a multi-institutional phase I trial of 5-fraction stereotactic body radiotherapy (SBRT) for Stage III-IVa laryngeal cancer. We conducted this pilot dosimetric study to confirm potential utility of online adaptive replanning to preserve treatment quality. METHODS: We evaluated ten cases: five patients enrolled onto the current trial and five patients enrolled onto a separate phase I SBRT trial for early-stage glottic larynx cancer. Baseline SBRT treatment plans were generated per protocol. Daily cone-beam CT (CBCT) or diagnostic CT images were acquired prior to each treatment fraction. Simulation CT images and target volumes were deformably registered to daily volumetric images, the original SBRT plan was copied to the deformed images and contours, delivered dose distributions were re-calculated on the deformed CT images. All of these were performed on a commercial treatment planning system. In-house software was developed to propagate the delivered dose distribution back to reference CT images using the deformation information exported from the treatment planning system. Dosimetric differences were evaluated via dose-volume histograms. RESULTS: We could evaluate dose within 10 minutes in all cases. Prescribed coverage to gross tumor volume (GTV) and clinical target volume (CTV) was uniformly preserved; however, intended prescription dose coverage of planning treatment volume (PTV) was lost in 53% of daily treatments (mean: 93.9%, range: 83.9-97.9%). Maximum bystander point dose limits to arytenoids, parotids, and spinal cord remained respected in all cases, although variances in carotid artery doses were observed in a minority of cases. CONCLUSIONS: Although GTV and CTV SBRT dose coverage is preserved with in-room three-dimensional image guidance, PTV coverage can vary significantly from intended plans and dose to critical structures may exceed tolerances. Online adaptive treatment re-planning is potentially necessary and clinically applicable to fully preserve treatment quality. Confirmatory trial accrual and analysis remains ongoing

    Stable, Environmental Specific and Novel QTL Identification as Well as Genetic Dissection of Fatty Acid Metabolism in Brassica napus

    Fatty acid (FA) composition is the typical quantitative trait in oil seed crops, of which study is not only closely related to oil content, but is also more critical for the quality improvement of seed oil. The double haploid (DH) population named KN with a high density SNP linkage map was applied for quantitative trait loci (QTL) analysis of FA composition in this study. A total of 406 identified QTL were detected for eight FA components with an average confidence interval (CI) of 2.92 cM, the explained phenotypic variation (PV) value ranged from 1.49 to 45.05%. Totally, 204 consensus and 91 unique QTL were further obtained via meta-analysis method for the purpose of detecting multiple environment expressed and pleiotropic QTL, respectively. Of which, 74 stable expressed and 22 environmental specific QTL were also revealed, respectively. In order to make clear the genetic mechanism of FA metabolism at individual QTL level, conditional QTL analysis was also conducted and more than two thousand conditional QTL which could not be detected under the unconditional mapping were detected, which indicated the complex interrelationship of the QTL controlling FA content in rapeseed. Through comparative genomic analysis and homologous gene annotation, 61 candidates related to acyl lipid metabolism were identified underlying the CI of FA QTL. To further visualize the genetic mechanism of FA metabolism, an intuitive and meticulous network about acyl lipid metabolism was constructed and some closely related candidates were positioned. This study provided a more accurate localization for stable and pleiotropic QTL, and a deeper dissection of the molecular regulatory mechanism of FA metabolism in rapeseed
