    Farm-level returns and costs of yellow catfish (Pelteobagrus fulvidraco) aquaculture in Guangdong and Zhejiang provinces, China

    AbstractFreshwater aquaculture in China is expanding and intensifying as this country experiences rapid economic growth, and understanding farm-level profitability is necessary if farmers are to make reasonable decisions about their production plans. We conducted a survey of yellow catfish farmers in 2014 in Guangdong and Zhejiang provinces in order to estimate farm-level profitability of pond aquaculture. We selected representative prefectures from the 2 provinces as study areas and used convenience sampling. Eighty-seven farmers were interviewed between April and May 2014 and the questionnaire collected detailed information on: (1) farmers’ demographics (age, gender, education, training, and experience); (2) production inputs (land, labor, fingerlings, feed, chemicals, machinery, and other miscellaneous costs); and (3) outputs (weight and revenue of harvested fish). Responses of 61 farmers included in the data analysis were post-stratified into 3 categories of farm size (<1.47ha, 1.47–3.67ha, and >3.67ha). We calculated production cost components, returns, and returns-costs ratios by farm size in each province. The overall returns-costs ratio was 1.31 in Guangdong and 1.17 in Zhejiang. Farmers in Guangdong invested more in land and machinery and had higher percentages of labor costs and chemical expenditures, but achieved better returns-costs ratios than farmers in Zhejiang. Higher land rent might be associated with greater yields of yellow catfish in Guangdong, which were almost twice those of Zhejiang

    Neural Super-Resolution for Real-time Rendering with Radiance Demodulation

    Rendering high-resolution images in real-time applications (e.g., video games, virtual reality) is time-consuming, thus super-resolution technology becomes more and more crucial in real-time rendering. However, it is still challenging to preserve sharp texture details, keep the temporal stability and avoid the ghosting artifacts in the real-time rendering super-resolution. To this end, we introduce radiance demodulation into real-time rendering super-resolution, separating the rendered image or radiance into a lighting component and a material component, due to the fact that the light component tends to be smoother than the rendered image and the high-resolution material component with detailed textures can be easily obtained. Therefore, we perform the super-resolution only on the lighting component and re-modulate with the high-resolution material component to obtain the final super-resolution image. In this way, the texture details can be preserved much better. Then, we propose a reliable warping module by explicitly pointing out the unreliable occluded regions with a motion mask to remove the ghosting artifacts. We further enhance the temporal stability by designing a frame-recurrent neural network to aggregate the previous and current frames, which better captures the spatial-temporal correlation between reconstructed frames. As a result, our method is able to produce temporally stable results in real-time rendering with high-quality details, even in the highly challenging 4 ×\times 4 super-resolution scenarios

    A case study of time-series regression modeling: Risk factors for pond-level mortality of farmed grass carp (Ctenopharyngodon idella) on a southern Chinese farm

    Limited research has been done using multivariable statistical methods to assess factors associated with fish mortality in warm-water finfish aquaculture in China. We carried out a case study to test the hypothesized association between pond-level daily mortality of farmed grass carp and predisposing environmental and husbandry factors. Based on logbook data from a single farm in Guangdong province (China) in 2013, two-stage time-series regression (TSR) analyses were conducted to estimate the lagged effect of these predisposing factors on grass carp mortality. Factors assessed included temperature fluctuations, movement of fish into and out of ponds, and 3 types of treatments (antibiotics-antiparasitics, traditional Chinese medicine-probiotics, and chemicals to improve water quality). First, coefficients were estimated using a generalized linear negative-binomial model for each pond, and these coefficient estimates were combined using meta-analytic techniques. Sensitivity analyses were done to compare effects of changes in the 3 modeling components: distributional forms, number of spline knots, and types of autocorrelation terms. Model results in the case study indicated 2 risk factors might be associated with increased mortality of grass carp: (1) movements-in of new fish during the previous 14 days; and (2) increasing water temperature during the previous 7 days. Sensitivity analyses indicated good consistency of the estimates with different modeling components. Our findings highlight the utility of assessing daily farm records using TSR to develop hypotheses about potential risk factors for grass carp mortality in China

    Characterization and Genomic Analysis of SFPH2, a Novel T7virus Infecting Shigella

    Shigellosis, caused by Shigella, is a major global health concern, with nearly 164.7 million cases and over a million deaths occurring annually worldwide. Shigella flexneri is one of the most common subgroups of Shigella with a high incidence of multidrug-resistance. The phage therapy approach is an effective method for controlling multidrug-resistant bacteria. However, only a few Shigella phages have been described to date. In this study, a novel lytic bacteriophage SFPH2 was isolated from a sewage sample obtained from a hospital in Beijing, China, using a multidrug-resistant S. flexneri 2a strain (SF2) isolated from the fecal sample of a dysentery patient. SFPH2 is a member of the Podoviridae virus family with an icosahedral capsid and a short, non-contractile tail. It was found to be stable over a wide range of temperatures (4–50°C) and pH values (pH 3–11). Moreover, SFPH2 could infect two other S. flexneri serotypes (serotypes 2 variant and Y). High-throughput sequencing revealed that SFPH2 has a linear double-stranded DNA genome of 40,387 bp with 50 open reading frames. No tRNA genes were identified in the genome. Comparative analysis of the genome revealed that the SFPH2 belongs to the subfamily Autographivirinae and genus T7virus. The genome shows high similarity with other enterobacterial T7virus bacteriophages such as Citrobacter phage SH4 (95% identity and 89% coverage) and Cronobacter phage Dev2 (94% identity and 92% coverage). A comparison of the fiber proteins showed that minor differences in the amino acid residues might specify different protein binding regions and determine host species. In conclusion, this is the first report of a T7virus that can infect Shigella; SFPH2 has a functional stability under a wide range of temperatures and pH values, showing the potential to be widely applied to control Shigella–associated clinical infections and reduce the transmission rates of S. flexneri serotype 2a and its variants in the environment

    Silicon and oxygen synergistic effects for the discovery of new high-performance nonfullerene acceptors

    In organic electronics, an aromatic fused ring is a basic unit that provides -electrons to construct semiconductors and governs the device performance. The main challenge in developing new pi -skeletons for tuning the material properties is the limitation of the available chemical approach. Herein, we successfully synthesize two pentacyclic siloxy-bridged pi -conjugated isomers to investigate the synergistic effects of Si and O atoms on the geometric and electronic influence of pi -units in organic electronics. Notably, the synthesis routes for both isomers possess several advantages over the previous approaches for delivering conventional aromatic fused-rings, such as environmentally benign tin-free synthesis and few synthetic steps. To explore their potential application as photovoltaic materials, two isomeric acceptor-donor-acceptor type acceptors based on these two isomers were developed, showing a decent device efficiency of 10%, which indicates the great potential of this SiO-bridged ladder-type unit for the development of new high-performance semiconductor materials.Developing a new pi -skeletal aromatic fused-ring for tuning material properties in organic electronics is still a challenge due to limited chemical approach. Here, the authors enrich the chemistry by synthesizing SiO-bridged ladder-type pi -skeletons with enhanced planarity and deeper energy levels than CO-bridged counterpart

    Aqueous Extract of Mori Folium Exerts Bone Protective Effect Through Regulation of Calcium and Redox Homeostasis via PTH/VDR/CaBP and AGEs/RAGE/Nox4/NF-κB Signaling in Diabetic Rats

    Purpose: The present study is aimed to explore whether the aqueous extract of Mori Folium (MF) exhibits bone protective effect by regulating calcium and redox homeostasis in diabetic rats, and to identify the signaling pathways involved in this process.Methods: Diabetic rats were established using high-sugar and high-fat diet and streptozotocin (STZ) (30 mg/kg for 3 consecutive days). The serum levels of osteocalcin (OC), insulin-like growth factor-1 (IGF-1), tartrate-resistant acid phosphatase (TRAP), phosphorus (P), calcium (Ca), 1,25-dihydroxyvitamin D3 [1,25(OH)2D3], parathormone (PTH), advanced glycation end products (AGEs), superoxide dismutase (SOD), and malondialdehyde (MDA), total antioxidant capacity (TAC), 8-hydroxy-2′-deoxyguanosine (8-OH-dG), and interleukin 6 (IL-6) were determined by ELISA or biochemical assays. Histopathological alterations in the femurs were evaluated by the stainings of hematoxylin-eosin (H&amp;E) and alizarin red S. In addition, femoral strength was detected by a three-point bending assay, bone microstructure was detected with micro-computer tomography. Bone material properties were examined by Fourier-transform infrared spectroscopy. Furthermore, the expressions of IGF-1, runt-related transcription factor 2 (Runx2), osteoprotegerin (OPG), receptor activator of nuclear factor kappa-B ligand (RANKL), cathepsin K, AGEs, receptor of advanced glycation end products (RAGE), NADPH oxidase 4 (Nox4), and nuclear factor kappa-B (NF-κB) in the femurs and tibias, and the alterations in the levels of calcium-binding protein-28k (CaBP-28k), transient receptor potential V6 (TRPV6), and vitamin D receptor (VDR) in the kidneys and duodenums were determined by western blot and immunohistochemical analysis.Results: Treatment of diabetic rats with MF aqueous extract induces an increase in the levels of OC and IGF-1 as well as a decrease in TRAP level in serum. MF treatment also upregulates the expression of OPG, downregulates the expressions of AGEs, RAGE, Nox4, NF-κB, and RANKL, which leads to improve bone microstructure and strength exhibited by an increase in cortical area ratio, cortical thickness, and trabecular area ratio as well as ultimate load, elastic modulus, and bending stress in the femurs and tibias of diabetic rats. In addition, MF aqueous extract preserves bone material properties by decreasing the ratio of fatty acid/collagen and increasing the ratio of mineral/matrix in the femurs of diabetic rats. Moreover, MF treatment increases the levels of P, Ca, and 1,25(OH)2D3, and decreases the level of PTH in the serum, as well as upregulates the expressions of TRPV6 and VDR in the duodenums and CaBP-28k in the kidneys of diabetic rats. Additionally, MF has ability of rebuilding redox homeostasis and eliminating inflammatory stress by increasing the levels of SOD and TAC as well as decreasing the levels of IL-6, AGEs, MDA, and 8-OH-dG.Conclusions: MF treatment may improve bone quality through maintenance of calcium homeostasis via regulating the PTH/VDR/CaBP signaling, and elimination of oxidative stress via regulating the AGEs/RAGE/Nox4/NF-κB signaling. These results may suggest the potential of MF in preventing the development of diabetic osteoporosis

    Leveraging 16S rRNA Microbiome Sequencing Data to Identify Bacterial Signatures for Irritable Bowel Syndrome

    Irritable bowel syndrome (IBS) is a chronic gastrointestinal disorder characterized by abdominal pain or discomfort. Previous studies have illustrated that the gut microbiota might play a critical role in IBS, but the conclusions of these studies, based on various methods, were almost impossible to compare, and reproducible microorganism signatures were still in question. To cope with this problem, previously published 16S rRNA gene sequencing data from 439 fecal samples, including 253 IBS samples and 186 control samples, were collected and processed with a uniform bioinformatic pipeline. Although we found no significant differences in community structures between IBS and healthy controls at the amplicon sequence variants (ASV) level, machine learning (ML) approaches enabled us to discriminate IBS from healthy controls at genus level. Linear discriminant analysis effect size (LEfSe) analysis was subsequently used to seek out 97 biomarkers across all studies. Then, we quantified the standardized mean difference (SMDs) for all significant genera identified by LEfSe and ML approaches. Pooled results showed that the SMDs of nine genera had statistical significance, in which the abundance of Lachnoclostridium, Dorea, Erysipelatoclostridium, Prevotella 9, and Clostridium sensu stricto 1 in IBS were higher, while the dominant abundance genera of healthy controls were Ruminococcaceae UCG-005, Holdemanella, Coprococcus 2, and Eubacterium coprostanoligenes group. In summary, based on six published studies, this study identified nine new microbiome biomarkers of IBS, which might be a basis for understanding the key gut microbes associated with IBS, and could be used as potential targets for microbiome-based diagnostics and therapeutics

    HIV-1-Infected and Immune-Activated Macrophages Induce Astrocytic Differentiation of Human Cortical Neural Progenitor Cells via the STAT3 Pathway

    Diminished adult neurogenesis is considered a potential mechanism in the pathogenesis of HIV-1-associated dementia (HAD). In HAD, HIV-1-infected and immune-activated brain mononuclear phagocytes (MP; perivascular macrophages and microglia) drive central nervous system (CNS) inflammation and may alter normal neurogenesis. We previously demonstrated HIV-1-infected and lipopolysaccharide (LPS) activated monocyte-derived macrophages (MDM) inhibit human neural progenitor cell (NPC) neurogenesis, while enhancing astrogliogenesis through the secretion of the inflammatory cytokines such as tumor necrosis factor-alpha (TNF-α), in vitro and in vivo. Here we further test the hypothesis that HIV-1-infected/activated MDM promote NPC astrogliogenesis via activation of the transcription factor signal transducer and activator of transcription 3 (STAT3), a critical factor for astrogliogenesis. Our results show that LPS-activated MDM-conditioned medium (LPS-MCM) and HIV-infected/LPS-activated MDM-conditioned medium (LPS+HIV-MCM) induced Janus kinase 1 (Jak1) and STAT3 activation. Induction of the Jak-STAT3 activation correlated with increased glia fibrillary acidic protein (GFAP) expression, demonstrating an induction of astrogliogenesis. Moreover, STAT3-targeting siRNA (siSTAT3) decreased MCM-induced STAT3 activation and NPC astrogliogenesis. Furthermore, inflammatory cytokines (including IL-6, IL-1β and TNF-α) produced by LPS-activated and/or HIV-1-infected MDM may contribute to MCM-induced STAT3 activation and astrocytic differentiation. These observations were confirmed in severe combined immunodeficient (SCID) mice with HIV-1 encephalitis (HIVE). In HIVE mice, siRNA control (without target sequence, sicon) pre-transfected NPCs injected with HIV-1-infected MDM showed more astrocytic differentiation and less neuronal differentiation of NPCs as compared to NPC injection alone. siSTAT3 abrogated HIV-1-infected MDM-induced astrogliogenesis of injected NPCs. Collectively, these observations demonstrate that HIV-1-infected/activated MDM induces NPC astrogliogenesis through the STAT3 pathway. This study generates important data elucidating the role of brain inflammation in neurogenesis and may provide insight into new therapeutic strategies for HAD
