Identifikacija optimizovanom taqman probom qpcr metodom gena rezistencije na scrapie kod ovaca na teritoriji Republike Srbije
Scrapie is an infectious neurodegenerative disease affecting the central nervous system of sheep and goats that belongs to transmissible spongiform encephalopathies. The disease is caused by the accumulation of proteinase-resistant isoform of the prion protein. The sheep predisposition to scrapie is associated with polymorphisms of the PrP gene. Genetic susceptibility to scrapie is mainly related to codons 136, 154, and 171. ARR sheep are strongly scrapie resistant and VRQ genotype is the most susceptible. Many countries have scrapie eradication programs based on using rams with resistant genotype. The eradication program has not yet been implemented in the Republic of Serbia. To examine the genetic makeup of sheep in Serbia related to scrapie, we optimized TaqMan probes of real-time polymerase chain reaction (qPCR) technique for three codons. Blood samples from 100 sheep were analyzed by qPCR and the majority of the examined sheep were AA homozygous for the 136 codon. For codon 154 the most frequent genotype was RR and for codon 171 the most frequent genotype was QQ.Ovčija svrabež je infektivno neurodegenerativno oboljenje iz grupe transmisivnih encefalopatija koje uzrokuje promene u moždanom tkivu ovaca i koza. Oboljenje je uzrokovano akumulacijom proteinaza-rezistentne izoforme prion proteina. Osetljivost ovaca na ovo oboljenje uslovljena je polimorfi zmom PrP gena. Genetska otpornost na ovčiju svrabež prevashodno je uslovljena kodonima 136, 154 i 171. Najotpornije su ovce sa ARR genotipom, dok se ovce sa VRQ genotipom smatraju najosetljivijim. Mnoge zemlje su sprovele programe eradikacije ovčije svrabeži bazirane na korišćenju priplodnih ovnova koji imaju rezistentan genotip. Ovakav program još nije sproveden u Republici Srbiji. U cilju ispitivanja genetskog statusa ovaca u Srbiji u pogledu otpornosti na ovčiju svrabež, optimizovali smo TaqMan probu, kvantitativne lančane reakcije polimeraze (qPCR) tehnike za sva tri kodona. Diet supplementation helps honey bee colonies in combat infections by enhancing their hygienic behaviour
The hygienic behavior in honey bees is a complex polygenic trait that serves as a natural defense mechanism against bacterial and fungal brood diseases and Varroa destructor mites infesting brood cells. The aim of this study was to evaluate the effect of a dietary amino acids and vitamins supplement “BEEWELL AminoPlus” on hygienic behavior of Apis mellifera colonies combating microsporidial and viral infections. The experiment was performed during a one-year period on 40 colonies alloted to five groups: one supplemented and infected with Nosema ceranae and four viruses (Deformed wing virus - DWV, Acute bee paralysis virus - ABPV, Chronic bee paralysis virus - CBPV and Sacbrood virus – SBV), three not supplemented, but infected with N. ceranae and/ or viruses, and one negative control group. Beside the l isted pathogens, honey bee trypanosomatids were also monitored in all groups.The supplement “BEEWELL AminoPlus” induced a significant and consistent increase of the hygienic behavior in spite of the negative effects of N. ceranae and viral infections. N. ceranae and viruses significantly and consistently decreased hygienic behavior, but also threatened the survival of bee colonies. Oxidative status of honey bees infected with Nosema ceranae microsporidium and supplemented with Agaricus bisporus mushroom extract
Nosema ceranae, a microsporidium species, is among the most common causes of bee diseases. The positive effect of Agaricus bisporus mushroom extract on the survival and immunity of Nosema-infected bees has been reported recently. The effect could be achieved by stimulating the expression of immune-related genes, but also by suppressing nosemosis. The aim of this work was to determine the effect of A. bisporus extract on the oxidative status of bees infected with N. ceranae. In a cage experiment on newly hatched bees, the effect of aqueous extract of champignon (A. bisporus, strain A15) was investigated. Six groups were formed: three groups were infected and received A. bisporus extract through food at different times (days 1, 3, and 6 after hatching), one group received the extract but was not infected (treatment control), one was only infected with Nosema (positive control) and one was neither infected nor received the extract (negative control). The effects were examined on samples taken on days 7 and 15 of the study. The activities of the antioxidant enzymes catalase (CAT), superoxide dismutase (SOD) and glutathione S-transferase (GST) and the concentrations of malondialdehyde (MDA) were determined. In comparison to the positive control, the enzyme activities and MDA concentrations were significantly lower in the groups fed with the mushroom extract supplement. In the negative control, the level of oxidative stress was lower than in the positive control. In comparison with the other groups, the values mostly did not differ significantly. Eight-Year Study of Haemogregarina stepanowi Infection in Poached European Pond Turtles (Emys orbicularis) Held in Belgrade Zoo Quarantine
The eight-year study (2015–2023) was performed on a large sample of poached European
pond turtles infected with Haemogregarina stepanowi and held in a pond that belongs to a quarantine
section of Belgrade Zoo. The protected species of European pond turtles have been found in poor
health, with general weakness, anorexia, and low motility. Comprehensive cytological, hematological,
molecular, and postmortem evaluations have been performed. Initially, Diff Quick staining of the
blood smears revealed rounded or elongated erythrocytes, often bearing premeront or U-shaped
gamont of the hemogregarines inside. The reduced erythrocyte numbers, hemoglobin, and hematocrit
values found in the examined population of infected turtles indicated anemia. Macroscopically, shell
necrosis and massive skin hemorrhages were the most prominent findings observed in diseased
turtles. Microscopically, the lungs, liver, kidneys, and spleen revealed hyperemia, hemorrhages,
and the presence of parasitic stages in tissue samples in 31 of 40 necropsied turtles. Cytological
and microscopic examination of the samples proved to be sufficient for establishing the infection,
but molecular analyses of the 18S sequence were used for phylogenetic studies. Over the years, the
number of diseased and dead turtles has decreased, which could be hypothetically attributed to the
elimination of leeches as the definitive host
Multiple epithelial tumors of skin and lingual mucosa in a mouse (Mus musculus) with characteristics of a papillomavirus infection – case report
Познато је да папиломавируси инфицирају дивље врсте мишева и пацова, као и
друге дивље глодаре, а 2011. године је откривена инфекција и код кућног миша
(Mus musculus).
У овом раду је описан приказ случаја вишеструких епителних тумора коже и
слузнице језика код миша. Миш, женка, стара 8 месеци, након појаве великог
броја чворова на кожи целог тела еутаназирана је и донесена на обдукцију. Два
месеца раније хируршки је уклоњена у том тренутку једина туморска маса на
кожи са хистопатолошком дијагнозом трихоепителиома. Узорци ткива са
обдукције фиксирани су у пуферизованом формалину и након обраде у
аутоматском ткивном процесору обојени хематоксилином и еозином.
Макроскопски су се уочавале мултифокалне, округле до овалне
пролиферативне промене на кожи промера од 4 mm до 3 cm, чврсто-еластичне
конзистенције, сиво-беле боје на пресеку, местимично са улегнутим
некротичним центром. Парцијалним скидањем коже уочено је да описане
промене не продиру у дубља ткива. Две промене сличних макроскопских
карактеристика промера око 4 mm уочене су на дорзалној страни језика.
Хистопатолошким прегледом уочене су јасно ограничене, ендофитичне,
местимично конфлуентне новотворевине сачињене од умножених епителних
ћелија. Оне на слузници језика одговарале су налазу папилома, док су на кожи
поред папилома уочене и туморске масе сачињене од умножених епителних
ћелија пореклом од длачног фоликула местимично са јасним знацима трихогене
диференцијације (трихоепителиоми). Туморске ћелије показале су знаке
папиломавирусне инфекције – увећану плаво-сиву цитоплазму, вакуолизацију
цитоплазме, једарне инклузије, вакуолизацију једра, присуство коилоцита.
Анамнестички подаци, клиничка слика и патоморфолошки налаз указују на
инфективну, највероватније вирусну етиологију туморских пролиферата која
одговара папиломавирусу мишева.It is known that papillomaviruses infect wild species of rats and mice, as well as other
wild rodents, and in 2011 infection in house mice (Mus musculus) was discovered.
In this paper a case of multiple epithelial tumors of skin and lingual mucosa was
reported. Female mouse, 8 months old, due to a large number of skin nodules was
euthanized and sent to necropsy. Two months earlier single nodular skin mass that
was at that time present was surgically removed with a histopathological diagnosis of
trichoepithelioma. Necropsy tissue samples were fixed in buffered formalin,
processed in an automatic tissue processor and stained with hematoxylin and eosin.
Grossly multifocal, round to oval, firm, proliferative cutaneous lesions ranging from
4 mm to 3 cm in diameter, grey-white on cross section, occasionally with depressed
necrotic center were noted. Partial skinning revealed that these changes did not
infiltrate deep tissues. Two similar changes were noted on the dorsal mucosa of the
tongue.
Histopathlogical examination showed sharply demarcated, endophytic, occasionally
confluent neoplasms made of proliferated epithelial cells. Those on the lingual
mucosa were consistent with papilloma, while on the skin besides papillomas there
were masses that consisted of proliferated epithelial cells of hair follicles with signs
of trichogenic differentiation (trichoepitheliomas). Tumor cells showed signs of
papillomavirus infection – enlarged blue-grey cytoplasm, cytoplasm vacuolization,
intranuclear inclusions, nuclear vacuolization, and koilocytes.
Anamnesis, clinical data and pathomorphological reports strongly suggest infective,
most probably viral etiology of described neoplasms that were consisted with

Fungal infections – new/old threat? The importance of veterinary pathology in the diagnostic
Царство гљива (Funghi) обухвата 6 милиона врста у оквиру којих се срећу облици
којима је развијен талус – гљиве/печурке и микроскопске облике – гљивице и
кваснице од којих 625 врста може да изазове инфекцију кичмењака и поред
тренутно актуелних вирусних и бактеријских обољења представљају све значајнија
обољења глобалног карактера која често имају зоонотски карактер.
Микозе (гљивичне инфекције) представљају обољења изазвана гљивицама.
Гљивице могу да инфицирају различитa ткива изазивајући површинске или дубоке
патолошке промене као и системско обољење. Микозе се деле на сапронозе
(сапронозне гљивичне инфекције) код којих су изазивачи опортунистичке гљивице
које се налазе у спољашњој средини у воденим екосистемима и/или земљишту
(хидрофилне и геофилне гљивице) и микотичне зоонозе (зоонозне гљивичне
инфекције) за које су одговорне зоофилне и антропофилне патогене гљивице и
преносе се са инфицираног домаћина.
У најчешће и најзначајније сапронозе су аспергилоза, мукоромикоза, кандидијаза и
инфекције меланизованим гљивицама, а нешто ређе се јављају кокцидиоидомикоза,
хистоплазмоза, паракокцидиоидомикоза и бластомикоза.
Најзначајније микотичне зоонозе, нарочито са аспекта дерматопатологије су
дерматофиције - инфекције изазване са Microsporum canis, Trichophyton mentagrophytes,
Trichophyton verrucosum, Epidermophyton, Sporothrix sp. и Mallasezia sp.
Група микоза као што су хитридиомикоза водоземаца, узрочник „синдрома белог
носа слепих мишева“ и узрочник „обољења жуте гљивице“ код рептила су од
значаја у домену очувања биодиверзитета јер могу да оболе одређене угрожене
животињске врсте.
У ветеринарској медицини гљивице представљају и значајне контаминенте хране за
животиње које путем својих метаболита (микотоксини) нарушавају здравље
животиња. Поред детекције макроскопских промена карактеристичних за микозе,
за процес дијагностике микоза значајно дијагностичко средство представља
хистопатологија. Специјализоване метода бојења које се поред рутинског бојења
хематоксилином и еозином (HE) користе у дијагностици микоза су Грокот (GMS)
које боји гљивице црном бојом и PAS бојење које боји гљивице ружичасто-црвено
и уоквирује гљивице тако да је омогућена визуелизација њихове морфологије.
Поред хистопатологије, за тачну диференцијацију неких врста гљивица може да се
користи имунохистохемијско испитивање. Гљивице представљају узрочнике који
се све чешће наводе као узрочници обољења која често имају зоонотски карактер.
Ветеринарска патологија има значајну улогу у дијагностици ових обољења на више
нивоа - у домену дерматопатологије, неких генерализованих стања - системских
микоза, као и у оквиру очувања угрожених животињских врста.Kingdom Funghi includes as many as 6 million of species of which shapes with
differentiated talus and microscopic forms can be found. 625 species of these have been
reported to cause infection in vertebrates, and beside actual viral and bacterial diseases
mycotic diseases represent the important global diseases which often have zoonotic
character. Mycosis represent diseases caused with fungi. Fungi can infect different tissues
causing superficial and profound pathologic changes, as well as systemic disease. Mycosis
are divided on sapronoses/sapronotic fungal infections caused by opportunistic
environmental fungi that inhabit aquatic ecosystems and/or soil (hydrophilic and geophilic
fungi) rather than a living host, and mycotic zoonoses/zoonotic mycotic infections –
caused by transmissible zoophilic and anthropohilic pathogen fungi which are transmitted
by contagious host.
Most frequent and most importabt sapronoses are aspergillosis, mucoromycosis,
candidiasis, cryptococcosis and infections due to melanised fungi, and less frequent
sapronoses are coccidioidomycosis, histoplasmosis, paracoccidioidomycosis, and
blastomycosis.
Most important fungal zoonoses, especially important in field of dermatopathology are
dermatophytic infections - infections caused by Microsporum canis, Trichophyton
mentagrophytes, Trichophyton verrucosum, Epidermophyton, Sporothrix sp. and Mallasezia sp.
Group of mycosis such as chytridiomycosis of amphibians, then causative agent of “bat
white-nose syndrome” and causative agent of “yellow fungus disease” in reptiles, which
are important in the preservation of biodiversity, because they can jeopardize some
endangered animal species.
In veterinary medicine fungi also represent important food contaminents which have great
impact on animal health causeb with their metabolites (mycotoxin). Besides recognition
of macroscopic changes characteristic for mycoses, histopathology represents an
important diagnostic tool in the diagnostics of these infections. Special stains are used in
diagnosis of fungal infections besides routine haematoxylin and eosin staining (HE).
These staining are Grocott-Gomori's methenamine silver stain (GMS) which stains fungi
black and Periodic acid–Schiff (PAS) which stains fungi pink-red, delineate the fungi so
that their morphology can be visualised. Also, in accurate differentiating of some fungal
species immunohistochemistry can be used. Fungi represent noxes which are more
common stated as cause of diseases which frequent have zoonotic character. Veterinary
pathology plays important role in fungal infection diagnostics weather in domain of
dermatopathology (diagnosis of superficial and subcutaneous mycoses), in recognition of
some general disorders - systemic mycoses, as well as in conservation of endangered

Sheep resistance to scrapie and sampling procedure for diagnosis of pathological prion protein in sheep
Ovčija svrabež (Scrapie, engl.) je neurodegenerativno oboljenje ovaca iz grupe transmisivnih spongiformnih encefalopatija (TSE).
Oboljenja iz grupe TSE se nazivaju prionske bolesti, jer je patološki
prion protein etiološki faktor za razvoj ovih oboljenja. Ovčija svrabež
se javlja u dve forme, kao klasična i atipična. Osetljivost ovaca na ovčiju svrabež zavisi od polimorfizma gena prionskog proteina i prirode
uzročnika prionske bolesti. Kodoni 136, 154 i 171 gena prion proteina
određuju stepen otpornosti, tj. sklonosti za Scrapie. Pošto je svaki alel
obeležen troslovnim kodom, utvrđeno je da su jedinke sa ARR alelom
visoko rezistentne na klasičnu ovčiju svrabež, ali ne pruža nikakvu posebnu zaštitu protiv atipične forme Scrapie-a. Primenom qPCR metode može se sprovesti genotipizacija jedinki u cilju uspešne selekcije
rezistentnih jedinki na ovčiju svrabež. Za metodu je neophodno uzeti
odgovarajući materijal i pripremiti ga za određivanje alela. Određivanje alela qPCR tehnikom sa Taquman probom je moguće iz krvi sa ili
bez dodatka antikoagulansa. Za dijagnostiku prisustva patološkog prion proteina kod ovaca u Laboratoriji za patologiju primenjuje se ELISA
metoda. Neophodno tkivo za dijagnostiku patološkog prion proteina
je obeks produžene moždine. Produžena moždina ovaca se uzorkuje
kroz foramen magnum specijalizovanim kašikama za uzorkovanja tkiva malih preživaraScrapie is a neurodegenerative disease belonging to the group of transmissible
spongiform encephalopathies (TSE). Diseases from the TSE group are also known as prion
diseases because they are caused by the pathological prion protein. Scrapie is a disease
that has two forms, classical and atypical. The susceptibility of sheep to Scrapie depends
on the polymorphism of the prion protein gene and the nature of the agent causing prion
disease. Codons 136, 154 and 171 of the prion protein gene determine a certain degree
of resistance, ie the tendency for Scrapie. Since each allele is marked with a three-letter
code, individuals with the ARR allele were found to be highly resistant to classical Scrapie,
but did not provide any special protection against the atypical form. Using the qPCR
method, genotyping of individuals can be performed for the successful selection of Scrapie
resistant animals. It is necessary to sample the appropriate material for qPCR method and
to prepare it for the determination of alleles. Determination of alleles by qPCR technique
with Taquman test is possible from blood with or without anticoagulants. In the Laboratory
of Pathology, the ELISA method is used to diagnose the presence of pathological prion
protein in sheep. The Material necessary for the diagnosis of pathological prion protein
is the region of obex of medulla oblongata. The medulla oblongata of sheep is sampled
through the foramen magnum with specialized spoons for sampling the tissues of small
ruminants
Identification of the presence of a scrapie resistance gene in sheep on the territory of the Republic of Serbia
Monitoring of scrapie in sheep and goats has been carried out for many years in Serbia.
Despite many years of monitoring, only one case of this disease has been reported in sheep,
but given that, there are numerous cases of the disease in some neighboring countries, it is
very important to determine the presence of resistant genes in the sheep population in Serbia.
Genetic susceptibility to scrapie is influenced mainly by the prion protein polymorphisms

Fibrinonecrotic endometritis and septicemia in farmed chinchilas due to Salmonella typhimurium infection
Чинчиле (Chinchilla lanigera) су мали сисари који припадају реду Rodentia и породици Chinchilidae. У природи живе на простору Анда у Јужној Америци. Данас се гаје у зоо вртовима, као кућни љубимци и узгајају на фармама због крзна. Салмонелоза је зоонотска болест која се јавља у целом свету. Изазивају је бактерије из рода Salmonella, које се сматрају стриктно патогеним код сисара и птица. Инфекција салмонелама код чинчила најчешће се јавља у септикемичној форми. У овом раду описане су патоморфолошке промене код чинчила угинулих од салмонелозе.
На Катедру за патологију достављени су лешеви две угинуле чинчиле код којих је пре угинућа клинички запажен крвави исцедак из стиднице. Спољашњим прегледом угинулих животиња уочено је да су биле у доброј кондицији, длака перианалне регије била је упрљана смеђе-црвеним садржајем, а из стиднице се изливао тамноцрвени садржај. Унутрашњим прегледом запажена је увећана, проширена материца напетог зида, плаво-црвене боје, наглашених крвних судова, испуњена трошним, безструктурним, сиво-ружичастим садржајем. Јетра је била благо тупих рубова, светлосмеђе боје. Плућа су била мозаичног изгледа, са ситним, тачкастим, тамноцрвеним подручјима. Узорци ткива са обдукције фиксирани су у пуферизованом формалину и након обраде у аутоматском ткивном процесору обојени хематоксилином и еозином.
Хистопатолошким прегледом је запажено псеудомембранозно запаљење материце са великом количином фибринонекротичног садржаја у којем су уочене колоније бактерија, док је мишићни слој био прожет интензивним крвављењима. Уочена су крвављења и у мозгу, плућима, срцу, бубрезима, јетри и цревима. Местимично се у крвним судовима мањег промера уочавао фибрински тромб. У плућима је запажен периваскуларни едем, док су у јетри уочена расута некротична подручја и мултифокални хепатитис, а код једне животиње су били присутни и знаци регенерације хепатоцита (увећане ћелије са базофилијом цитоплазме, двоједарне ћелије, крупна једра). Микробиолошким испитивањем узорака органа утврђено је присуство бактерије Salmonella enterica subspecies enterica серотип Typhimurium (Salmonella Typhimurium).Chinchillas (Chinchilla lanigera) are small mammals of the order Rodentia and the family Chinchilidae with natural habitats in the Andes in South America. They are kept in zoos, as pets and farmed for fur. Salmonellosis is a zoonotic disease known worldwide. It is caused by bacteria from the genus Salmonella, which are considered strictly pathogenic in mammals and birds. In chinchillas, infection with Salmonella sp. is usually in the form of septicemia. This paper describes pathomorphological changes in chinchillas died from salmonellosis.
Carcasses of two chinchillas with bloody vulvar discharge noticed clinically were delivered to the Department of pathology. External examination revealed that animals were in good body condition, with the tan-red stained perianal area and dark red secret from the vulva. An internal examination showed an enlarged, bluish-red uterus with accentuated vasculature filled with friable, amorphous greyish-pink content. The liver was slightly enlarged and pale tan. Lungs had a mosaic appearance with punctuated dark red areas. Necropsy tissue samples were fixed in buffered formalin, processed in an automatic tissue processor and stained with hematoxylin and eosin.
Microscopic examination showed pseudomembranous inflammation of the uterus with a large amount of fibrinonecrotic content in which many bacterial colonies were present. At the same time in muscle layers, there was intense hemorrhage. Hemorrhages were also seen in the brain, lungs, heart, kidney, liver and intestine. Occasionally in small blood vessels, there was fibrin thrombus. In the lungs, perivascular edema was noticed. The liver had disseminated necrotic areas and multifocal hepatitis, and in one animal, there were signs of hepatocyte regeneration (enlarged cells with cytoplasmic basophilia, binucleated cells, and large nuclei). Microbiological examination of organ samples revealed the presence of Salmonella enterica subspecies enterica serotype Typhimurium (Salmonella Typhimurium).Zbornik kratkih sadržaj
Mehanizmi lekovitog dejstva propolisa u apiterapiji životinja
Propolis je smeša koju proizvode pčele sakupljajući lepljive materije sa pupoljaka
biljaka i mešajuci ih sa voskom. Pčele propolisom učvršćuju saće i zatvaraju pukotine
unutar gnezda. Propolis su za lečenje i prevenciju bolesti koristile još praistorijske
civilizacije, a poznato je da su ga Stari Egipćani, Grci i Rimljani koristili za lečenje rana,
kožnih lezija i čireva. Razvoj nauke u XX i XXI veku omogućio je dokazivanje pozitivnih
efekata i mehanizama delovanja propolisa. Biološke aktivnosti propolisa u vezi su sa
hemijskim sastavom, koji varira periodično, zavisno od vegetacije. Dokazani
medicinski efekti propolisa su: antimikrobni (antibakterijski, antifungalni i
antivirusni), antioksidativni i imunoregulatorni. Dokazan je efekat propolisa kako na
Gram pozitivne bakterije (rodova Staphylococcus, Streptococcus, Kocuria), tako i na
gram negativne bakterije (rodova Haemophilus, Porphyromonas, Prevotella).
Antigljivični efekat propolisa najviše je istraživan i dokazan za rodove Candida,
Saccharomyces i Trichophyton, dok je najznačajniji antivirusni potencijal potvrđen za
Herpes Simplex Virus tip 1, influenca viruse kao i SARS-CoV-2. Kada je reč o
imunoregulatornom efektu, dokazan je inhibitorni efekat na formiranje granuloma i
eksudata, povećanje vaskularne permeabilnosti i dobar analgetski efekat kod

leukotrijena u peritonealnim makrofagima miševa. Flavonoidi i fenolne kiselie, u
zavisnosti od porekla propolisa, čine glavne imuno-značajne komponenate propolisa.
Neki istraživači su izdvojili fenetil estar kofeinske kiseline kao veoma značajnu
imunoregulatornu komponentu propolisa. Još neki od puteva delovanja propolisa su smanjenje proizvodnje proinflamatornih citokina kao što su TNF-α, IL-6 i povećanje
IL-10, TGF-b i IL-1β. Imunoregulatorni efekti propolisa dovode ili do stimulacije, ili do
suzbijanja imunog odgovora, čineći ga potencijalno primenljivim u alternativnoj i
pomoćnoj terapiju u lečenju raznih bolesti. U ovom radu biće predstavljeni primeri
