    At the crossing-places: representations of masculinity in selected 21st century children's texts

    This study explores the representations of masculinity in selected contemporary children’s adventure literature. According to John Stephens (2002:x), a problem for boys, both in narrative fictions and in the world, is that hegemonic masculinity ‘appears simultaneously to propose a schema for behaviour and to insist on their subordination as children, to conflate agency with hegemonic masculinity, and to disclose that, for them, such agency is illusory’. This issue, among others, forms the basis of the research as this paradox is particularly evident in texts that fall within the adventure genre, where protagonists present an image of empowered masculinity that has little or no correlation in real, that is, non-literary, childhood. Nevertheless, despite this apparent conflict, the discourses portrayed in these texts continue to influence society (in varying degrees) as they are promoted, perpetuated and disseminated through cultural productions. Moreover, as this research rests on the premise of a belief ‘in the cultural productivity of fictions’ (Knights 1999:vii), it focuses on literary material that forms part of the landscape of childhood in contemporary society. Therefore, this study analyses selected 21st century children’s texts in order to identify and discuss the representations of masculinity in these texts in the context of their publication at a time when hegemonic masculinity has long been a topic of popular and academic debate. The primary texts include the Arthur series by Kevin Crossley-Holland, Anthony Horowitz’s Alex Rider series, the Young Bond series by Charlie Higson and Steve Cole and the Bodyguard books by Chris Bradford.By using discourse theory as a lens to complement the masculinity studies approach, this research investigates the questions posed under the problem statement and presents findings that demonstrate that the gender models presented in the texts are, for the most part, cast in ‘the masculinist and patriarchal conventions that characterised imperialist adventure’ (Capdevila 2003:216). Thus, it is evident that the children’s adventure genre seems to be rather tardy in keeping with the times.     Advanced learner perceptions of psychological well-being and school satisfaction in two educational settings

    The intent of this study was to explore the effects of two settings on self-concept and school satisfaction of academically advanced high school students. The research designs were causal-comparative and correlational. Participants were 224 partial-day academic Governor\u27s School students and 56 academically advanced students in district schools in a southern U.S. state.;Self-concept for the total group appeared adequate to strong, and the two student groups were similar in total self-concept and its dimensions ( p \u3e .01), as measured by the Piers-Harris Children\u27s Self-Concept Scale -- Second Edition (Piers- Harris 2). However, frequent worry, nervousness, and feelings of differentness were revealed by a number of participants. The findings for the School Attitude Assessment Survey -- Revised suggested that the Governor\u27s School students were more satisfied with the Governor\u27s Schools than with their district schools (p \u3c .05), although academic self-perception, on average, was significantly lower in the Governor\u27s Schools. They also appeared more satisfied with the Governor\u27s Schools than the district students were with their schools (p \u3c .05). Several positive correlations were found between self- concept and school attitudes.;For the total sample, while obtaining similar results to those of the males in most self-concept dimensions, the females obtained a significantly lower mean score for the Piers-Harris 2 Freedom From Anxiety domain ( p \u3c.05). In addition, significantly higher goal valuation and motivation/self-regulation, and lower academic self-perception (p \u3c .05), were found for the sample of 159 females in the district setting. Implications for educational practice, counseling interventions, and future research are provided

    AbstractOne of the pathological hallmarks of ALS is the presence of axonal spheroids and perikaryal accumulations/aggregations comprised of the neuronal intermediate filament proteins, neurofilaments and peripherin. These abnormalities represent a point of convergence of both familial and sporadic forms of the disease and understanding their formation may reveal shared pathways in what is otherwise considered a highly heterogeneous disorder. Here we provide a review of the basic biology of neurofilaments and peripherin and the evidence linking them with ALS disease pathogenesis

    A single-molecule method for measuring fluorophore labeling yields for the study of membrane protein oligomerization in membranes

    Membrane proteins are often observed as higher-order oligomers, and in some cases in multiple stoichiometric forms, raising the question of whether dynamic oligomerization can be linked to modulation of function. To better understand this potential regulatory mechanism, there is an ongoing effort to quantify equilibrium reactions of membrane protein oligomerization directly in membranes. Single-molecule photobleaching analysis is particularly useful for this as it provides a binary readout of fluorophores attached to protein subunits at dilute conditions. However, any quantification of stoichiometry also critically requires knowing the probability that a subunit is fluorescently labeled. Since labeling uncertainty is often unavoidable, we developed an approach to estimate labeling yields using the photobleaching probability distribution of an intrinsic dimeric control. By iterative fitting of an experimental dimeric photobleaching probability distribution to an expected dimer model, we estimate the fluorophore labeling yields and find agreement with direct measurements of labeling of the purified protein by UV-VIS absorbance before reconstitution. Using this labeling prediction, similar estimation methods are applied to determine the dissociation constant of reactive CLC-ec1 dimerization constructs without prior knowledge of the fluorophore labeling yield. Finally, we estimate the operational range of subunit labeling yields that allows for discrimination of monomer and dimer populations across the reactive range of mole fraction densities. Thus, our study maps out a practical method for quantifying fluorophore labeling directly from single-molecule photobleaching data, improving the ability to quantify reactive membrane protein stoichiometry in membranes

    Long-pore Electrostatics in Inward-rectifier Potassium Channels

    Inward-rectifier potassium (Kir) channels differ from the canonical K+ channel structure in that they possess a long extended pore (∼85 Å) for ion conduction that reaches deeply into the cytoplasm. This unique structural feature is presumably involved in regulating functional properties specific to Kir channels, such as conductance, rectification block, and ligand-dependent gating. To elucidate the underpinnings of these functional roles, we examine the electrostatics of an ion along this extended pore. Homology models are constructed based on the open-state model of KirBac1.1 for four mammalian Kir channels: Kir1.1/ROMK, Kir2.1/IRK, Kir3.1/GIRK, and Kir6.2/KATP. By solving the Poisson-Boltzmann equation, the electrostatic free energy of a K+ ion is determined along each pore, revealing that mammalian Kir channels provide a favorable environment for cations and suggesting the existence of high-density regions in the cytoplasmic domain and cavity. The contribution from the reaction field (the self-energy arising from the dielectric polarization induced by the ion's charge in the complex geometry of the pore) is unfavorable inside the long pore. However, this is well compensated by the electrostatic interaction with the static field arising from the protein charges and shielded by the dielectric surrounding. Decomposition of the static field provides a list of residues that display remarkable correspondence with existing mutagenesis data identifying amino acids that affect conduction and rectification. Many of these residues demonstrate interactions with the ion over long distances, up to 40 Å, suggesting that mutations potentially affect ion or blocker energetics over the entire pore. These results provide a foundation for understanding ion interactions in Kir channels and extend to the study of ion permeation, block, and gating in long, cation-specific pores
