    Population decline is linked to migration route in the Common Cuckoo

    Migratory species are in rapid decline globally. Although most mortality in long-distance migrant birds is thought to occur during migration, evidence of conditions on migration affecting breeding population sizes has been completely lacking. We addressed this by tracking 42 male Common Cuckoos from the rapidly declining UK population during 56 autumn migrations in 2011–14. Uniquely, the birds use two distinct routes to reach the same wintering grounds, allowing assessment of survival during migration independently of origin and destination. Mortality up to completion of the Sahara crossing (the major ecological barrier encountered in both routes) is higher for birds using the shorter route. The proportion of birds using this route strongly correlates with population decline across nine local breeding populations. Knowledge of variability in migratory behaviour and performance linked to robust population change data may therefore be necessary to understand population declines of migratory species and efficiently target conservation resources

    Use of UV Treated Milk Powder to Increase Vaccine Efficacy in the Elderly

    Aging populations experience a decline in adaptive immune system function also known as immunosenesence. Protein nutrition has been shown to stimulate and strengthen the immune system, and such approaches are needed for this growing segment of the population. A controlled, randomized, double blind pilot study was conducted to compare two different protein sources (soy and dairy) as nutritional supplementation to enhance vaccine response. Our objective was to examine the immune stimulating effects of dairy protein subjected to ultraviolet radiation (UV-C) radiation treatment process instead of pasteurization. Participants were 21 healthy individuals over 60 years of age who consumed 6 g of the dairy protein or a comparison, soy isoflavone protein, twice a day for 8 weeks. DTaP vaccine administered at week 4. Non-parametric t-tests revealed a significant increase in Tetanus antibodies in the dairy group compared to the soy group at week 8. These findings suggest additional benefits of UV-C treated unheated dairy protein as a solution to counteract immunosenescence, but warrant further study in elderly and other populations that might benefit from immune system stimulation

    In this paper the quantum cosmological consequences of introducing a term cubic in the Ricci curvature scalar RR into the Einstein--Hilbert action are investigated. It is argued that this term represents a more generic perturbation to the action than the quadratic correction usually considered. A qualitative argument suggests that there exists a region of parameter space in which neither the tunneling nor the no-boundary boundary conditions predict an epoch of inflation that can solve the horizon and flatness problems of the big bang model.     Comparison of Pond Production of Phase-III Sunshine Bass Fed 32-, 36-, and 40%-Crude-Protein Diets with Fixed Energy : Protein Ratios

    We stocked phase-III sunshine bass (white bass Morone chrysops ♀ × striped bass M. saxatilis ♂) at a rate of 6,188 fingerlings/ha into twelve 0.04-ha earthen ponds supplied with continuous aeration. Three dietary treatments were randomly assigned to quadruplicate ponds. Sunshine bass were fed to apparent satiation once daily after average initial weight (mean ± SE = 214 ± 5 g) and total length (245 ± 1.6 mm) were determined. Diets were formulated to conserve the estimated digestible energy:crude protein (CP) ratio (9.3 kcal/g protein) and represented the following CP and energy values fed to fish: 32% CP (3,000 kcal/kg), 36% CP (3,360 kcal/kg), and 40% CP (3,760 kcal/kg). Harvest data suggest that nutrient density is a variable that can be manipulated to optimize production and reduce production costs. Production rates (mean ± SE) were 2,851 ± 600 kg/ha for the 32%-CP diet, 2,895 ± 341 kg/ha for the 36%-CP diet, and 2,953 ± 142 kg/ha for the 40%-CP diet; production rates were not significantly different among dietary treatments. Survival was excellent and did not appear to be related to dietary treatment. Dressed (gilled and gutted) fish averaged 80% of whole-fish weight, and the dressed percentage did not vary as a function of nutrient density. Feed conversion ratios of 3.0 ± 0.4, 2.8 ± 0.2, and 2.6 ± 0.1 were obtained for the fish fed 32-, 36-, and 40%-CP diets, respectively. Protein conversion ratios (mean = 1.0) were not significantly influenced by dietary treatment. Feed cost increased with increasing dietary CP level; costs were US0.447perkilogramforthe320.447 per kilogram for the 32%-CP diet, 0.493 per kilogram for the 36%-CP diet, and 0.541perkilogramforthe400.541 per kilogram for the 40%-CP diet. The resulting production costs attributable to feed were 1.34, 1.38,and1.38, and 1.41 per kilogram of gain for the 32-, 36-, and 40%-CP diets, respectively. A savings of 0.16perkilogramproduced,orapproximately0.16 per kilogram produced, or approximately 450 per hectare, was realized as a result of feeding either of the two lower-CP, lower-energy diets. Accordingly, we suggest that phase-III sunshine bass can be more economically produced by feeding diets as low as 32% CP with a minimum energy:protein ratio of 9.3 kcal/g CP

    Conformational Transitions in the Membrane Scaffold Protein of Phospholipid Bilayer Nanodiscs

    Phospholipid bilayer nanodiscs are model membrane systems that provide an environment where membrane proteins are highly stable and monodisperse without the use of detergents or liposomes. Nanodiscs consist of a discoidal phospholipid bilayer encircled by two copies of an amphipathic alpha helical membrane scaffold protein, which is modeled from apolipoprotein A-1. Hydrogen exchange mass spectrometry was used to probe the structure and dynamics of the scaffold protein in the presence and absence of lipid. On nanodisc self-assembly, the entire scaffold protein gained significant protection from exchange, consistent with a large, protein-wide, structural rearrangement. This protection was short-lived and the scaffold protein was highly deuterated within 2 h. Several regions of the scaffold protein, in both the lipid-free and lipid-associated states, displayed EX1 unfolding kinetics. The rapid deuteration of the scaffold protein and the presence of correlated unfolding events both indicate that nanodiscs are dynamic rather than rigid bodies in solution. This work provides a catalog of the expected scaffold protein peptic peptides in a nanodisc-hydrogen exchange mass spectrometry experiment and their deuterium uptake signatures, data that can be used as a benchmark to verify correct assembly and nanodisc structure. Such reference data will be useful control data for all hydrogen exchange mass spectrometry experiments involving nanodiscs in which transmembrane or lipid-associated proteins are the primary molecule(s) of interest

    Fermilab E791, a very high statistics charm particle experiment, recently completed its data taking at Fermilab's Tagged Photon Laboratory. Over 20 billion events were recorded through a loose transverse energy trigger and written to 8mm tape in the the 1991-92 fixed target run at Fermilab. This unprecedented data sample containing charm is being analysed on many-thousand MIP RISC computing farms set up at sites in the collaboration. A glimpse of the data taking and analysis effort is presented. We also show some preliminary results for common charm decay modes. Our present analysis indicates a very rich yield of over 200K reconstructed charm decays.Comment: 4 pages, 1 figure, LaTe

    Conformational Analysis of Membrane Proteins in Phospholipid Bilayer Nanodiscs by Hydrogen Exchange Mass Spectrometry

    The study of membrane protein structure and enzymology has traditionally been hampered by the inherent insolubility of membrane proteins in aqueous environments and experimental challenges in emulating an in vivo lipid environment. Phospholipid bilayer nanodiscs have recently been shown to be of great use for the study of membrane proteins since they offer a controllable, stable, and monodisperse model membrane with a native-like lipid bilayer. Here we report the integration of nanodiscs with hydrogen exchange (HX) mass spectrometry (MS) experiments, thereby allowing for analysis of the native conformation of membrane proteins. Gamma-glutamyl carboxylase (GGCX), an ~94 kDa transmembrane protein, was inserted into nanodiscs and labeled with deuterium oxide under native conditions. Analytical parameters including sample-handling and chromatographic separation were optimized to measure the incorporation of deuterium into GGCX. Coupling nanodisc technology with HX MS offers an effective approach for investigating the conformation and dynamics of membrane proteins in their native environment and is therefore capable of providing much needed insight into the function of membrane proteins
