    Homeobox Transcription Factors Are Required for Conidiation and Appressorium Development in the Rice Blast Fungus Magnaporthe oryzae

    The appropriate development of conidia and appressoria is critical in the disease cycle of many fungal pathogens, including Magnaporthe oryzae. A total of eight genes (MoHOX1 to MoHOX8) encoding putative homeobox transcription factors (TFs) were identified from the M. oryzae genome. Knockout mutants for each MoHOX gene were obtained via homology-dependent gene replacement. Two mutants, ΔMohox3 and ΔMohox5, exhibited no difference to wild-type in growth, conidiation, conidium size, conidial germination, appressorium formation, and pathogenicity. However, the ΔMohox1 showed a dramatic reduction in hyphal growth and increase in melanin pigmentation, compared to those in wild-type. ΔMohox4 and ΔMohox6 showed significantly reduced conidium size and hyphal growth, respectively. ΔMohox8 formed normal appressoria, but failed in pathogenicity, probably due to defects in the development of penetration peg and invasive growth. It is most notable that asexual reproduction was completely abolished in ΔMohox2, in which no conidia formed. ΔMohox2 was still pathogenic through hypha-driven appressoria in a manner similar to that of the wild-type. However, ΔMohox7 was unable to form appressoria either on conidial germ tubes, or at hyphal tips, being non-pathogenic. These factors indicate that M. oryzae is able to cause foliar disease via hyphal appressorium-mediated penetration, and MoHOX7 is mutually required to drive appressorium formation from hyphae and germ tubes. Transcriptional analyses suggest that the functioning of M. oryzae homeobox TFs is mediated through the regulation of gene expression and is affected by cAMP and Ca2+ signaling and/or MAPK pathways. The divergent roles of this gene set may help reveal how the genome and regulatory pathways evolved within the rice blast pathogen and close relatives

    Re-defining response and treatment effects for neuro-oncology immunotherapy clinical trials

    In much of medical oncology, including neuro-oncology, there is great interest to evaluate the therapeutic potential of immune-based therapies including vaccines, adoptive T cell strategies and modulators of immune checkpoint regulators such as cytotoxic T lymphocyte antigen 4 and programmed death 1. Immune-based treatments exert an indirect anti-tumor effect by generating potent, tumor-targeting immune responses. Robust anti-tumor immune responses have been shown to achieve encouraging radiographic responses across the spectrum of applied immunotherapeutics which are felt to be indicative of a bona fide anti-tumor effect. Conversely, worsening of imaging findings, particularly early in the course of immunotherapy administration, can be challenging to interpret with growing evidence demonstrating that at least a subset of such patients ultimately will derive meaningful clinical benefit. The immune related response criteria were generated to provide guidance regarding the interpretation of such complex imaging findings, for general medical oncologists prescribing immunotherapeutics. An analogous effort that addresses challenges associated with imaging assessment and incorporates nuances associated with neuro-oncology patients is underway and is referred to as the immunotherapy response assessment in neuro-oncology criteria

    Housing: An Under-Explored Influence on Children’s Well-Being and Becoming

    Research on housing has tended to focus on adult outcomes, establishing relationships between housing and a number of aspects of health and well-being. Research exploring the influence of housing on children has been more limited, and has tended to focus on adult concerns around risk behaviours, behavioural problems and educational attainment. While these outcomes are important, they neglect the impact of housing on children’s lives beyond these concerns. There are a number of reasons to believe that housing would play an important role in children’s well-being more broadly. Family stress and strain models highlight how housing difficulties experienced by adults may have knock on effects for children, while Bronfenbrenner’s ecological approach to human development emphasises the importance of children’s experiences of their environments, of which the home is among the most important. This paper summaries the existing evidence around housing and child outcomes, predominantly educational and behavioural outcomes, and argues for the extension of this work to consider the impact of housing on children’s lives more broadly, especially their subjective well-being

    Integrated control of Allium white rot with Trichoderma viride, tebuconazole and composted onion waste

    Two isolates of Trichoderma viride (L4 and S17A) were assessed for biological control of Allium white rot (AWR) with different onion accessions and cultivars, alone and in combination with a tebuconazole-based seed treatment or composted onion waste. In glasshouse tests, 23 new bulb-onion accessions from previous work to detect resistance to Sclerotium cepivorum showed no differences in susceptibility to AWR but, when combined with S17A, disease was reduced by up to two-thirds over all accessions. Trichoderma viride L4 and S17A also reduced the proportion of infected plants for five commercial bulb-onion cultivars and one advanced breeding line by at least one-third. Further glasshouse tests using a salad-onion cultivar showed that L4, S17A, tebuconazole or composted onion waste controlled AWR and at least halved the proportion of diseased plants. Combination treatments of T. viride with either tebuconazole or compost enhanced control and, in some treatments, disease was almost eliminated. In field trials, control of AWR by S17A was significant for 17 out of 18 individual or mixed bulb-onion accessions, with disease reduced overall by more than half. In another field experiment, S17A failed significantly to reduce AWR for two out of three commercial bulb-onion cultivars, while tebuconazole reduced the final proportion of AWR-infected plants over all cultivars from 0.47 to 0.09. Combining S17A and tebuconazole resulted in a similar level of AWR to using tebuconazole alone. The use of T. viride in an integrated strategy with other treatments to enhance control of S. cepivorum is discussed
