    Early above- and below-ground responses of subboreal conifer seedlings to various levels of deciduous canopy removal

    We examined the growth of understory conifers, following partial or complete deciduous canopy removal, in a field study established in two regions in Canada. In central British Columbia, we studied the responses of three species (Pseudotsuga menziesii var. glauca (Beissn.) Franco, Picea glauca (Moench) Voss x Picea engelmannii Parry ex Engelm., and Abies lasiocarpa (Hook.) Nutt.), and in northwestern Quebec, we studied one species (Abies balsamea (L.) Mill.). Stem and root diameter and height growth were measured 5 years before and 3 years after harvesting. Both root and stem diameter growth increased sharply following release but seedlings showed greater root growth, suggesting that in the short term, improvement in soil resource capture and transport, and presumably stability, may be more important than an increase in stem diameter and height growth. Response was strongly size dependent, which appears to reflect greater demand for soil resources as well as higher light levels and greater tree vigour before release for taller individuals. Growth ratios could not explain the faster response generally attributed to true fir species or the unusual swift response of spruces. Good prerelease vigour of spruces, presumably favoured by deciduous canopies, could explain their rapid response to release

    Assessing the impact of AGS-004, a dendritic cell-based immunotherapy, and vorinostat on persistent HIV-1 Infection

    Approaches to deplete persistent HIV infection are needed. We investigated the combined impact of the latency reversing agent vorinostat (VOR) and AGS-004, an autologous dendritic cell immunotherapeutic, on the HIV reservoir. HIV+, stably treated participants in whom resting CD4+ T cell-associated HIV RNA (rca-RNA) increased after VOR exposure ex vivo and in vivo received 4 doses of AGS-004 every 3 weeks, followed by VOR every 72 hours for 30 days, and then the cycle repeated. Change in VOR-responsive host gene expression, HIV-specific T cell responses, low-level HIV viremia, rca-RNA, and the frequency of resting CD4+ T-cell infection (RCI) was measured at baseline and after each cycle. No serious treatment-related adverse events were observed among five participants. As predicted, VOR-responsive host genes responded uniformly to VOR dosing. Following cycles of AGS-004 and VOR, rca-RNA decreased significantly in only two participants, with a significant decrease in SCA observed in one of these participants. However, unlike other cohorts dosed with AGS-004, no uniform increase in HIV-specific immune responses following vaccination was observed. Finally, no reproducible decline of RCI, defined as a decrease of >50%, was observed. AGS-004 and VOR were safe and well-tolerated, but no substantial impact on RCI was measured. In contrast to previous clinical data, AGS-004 did not induce HIV-specific immune responses greater than those measured at baseline. More efficacious antiviral immune interventions, perhaps paired with more effective latency reversal, must be developed to clear persistent HIV infection

    Acculturation and cognitive performance of migrant children in the Netherlands

    Acculturative strategies and cognitive acculturation were studied among 118 Dutch migrant children (age: 7-12 years). Both an exploratory factor analysis and a Linear Logistic Test Model analysis provided evidence for the bipolar unidimensionality of Berry's four acculturation strategies; integration constituted one pole and assimilation, separation, and marginalization the other. Integration was by far the most popular strategy. Cognitive performance was measured by the Snijders-Oomen Nonverbal Intelligence Test, school report measures, and a computer-assisted choice reaction-time measure. A good fit was found for a structural model that postulated cognitive acculturation as the latent variable linking background measures (i.e. acculturation strategy, second language usage, age, and gender) to the cognitive measures. Relationships between background variables and cognitive performance were stronger in the first than in the second generation. Both older children and children who favour an integration strategy showed a higher degree of cognitive acculturation

    A Word Tracking Task as an alternative to horizontal eye movements in the reduction of vividness and emotionality of aversive memories in EMDR

    When treating a patient with PTSD, therapists often use eye movement desensitization and reprocessing (EMDR). In EMDR patients make horizontal eye movements (HEM) while the image of a traumatic memory is recalled. Various studies showed that making HEM during recall of an aversive memory results in a decline in vividness and emotionality of this memory. This study aimed to create an alternative task that would be less physically demanding for the therapist than applying HEM. This task should, according to the working memory (WM) hypothesis, tax the WM as much as HEM. To accomplish this goal, a word-tracking task (WTT) was created in which an oval that moved over a matrix with color-words had to be followed with the eyes. Experiment I showed that the WTT taxes WM, though not as much as HEM. In experiment II, both the WTT and HEM resulted in a decrease in vividness and emotionality of an aversive memory. The results obtained raise questions about a supposed linear relationship between the WM-taxing and EMDR-efficacy of tasks. Further investigation of this relationship is recommended. Also recommended is further study in a clinical population. The WTT seems to be a good alternative for horizontal eye movements in EMDR
