Spatial and temporal variations of bioavailable polycyclic aromatic hydrocarbons in the lower Willamette River
Urban rivers with a history of industrial use can exhibit spatial and temporal variations in contaminant concentrations that may significantly affect risk evaluations and assessment of remediation efforts. To more effectively estimate human and ecological health risks, we evaluated the spatial and temporal distribution of polycyclic aromatic hydrocarbons (PAHs) in an urban river using low-density polyethylene passive sampling devices (PSDs). PSDs simulate cell membranes and lipid tissue and accumulate only freely dissolved, and thus, bioavailable contamination. Using PSDs and high-pressure liquid chromatography we measured bioavailable concentrations of 15 priority pollutant PAHs over five years along 18.5 miles of the lower Willamette River including the Portland Harbor Superfund megasite. This area contains several PAH sources including coal tar and creosote sites, which underwent remediation during this study. Additional potential sources of PAHs include combined sewer overflows, urban runoff, atmospheric deposition and petroleum product leaks and spills. Results reveal increased urban bioaccessible PAH loading during wet seasons and high rain events, successful capping of the McCormick and Baxter Superfund site, increased bioavailable PAHs during dredging operations, and the potential for using PSDs for source apportionment. PAH concentrations, loads, and congener ratios changed significantly depending on location, river flow, precipitation, and contaminant removal activities. Our research demonstrates that human and ecological risk varies significantly by season and remediation technique. Additionally, our refined analytical method eliminated 80% of organic solvents and 100% of chlorinated solvents from the sampling method and also reduced labor and cost. Health professionals can apply the findings and refined technology to more effectively assess exposure to pollution

Predicting Polycyclic Aromatic Hydrocarbon Concentrations in Resident Aquatic Organisms Using Passive Samplers and Partial Least-Squares Calibration
The current work sought to develop predictive
models between time-weighted average polycyclic aromatic
hydrocarbon (PAH) concentrations in the freely dissolved
phase and those present in resident aquatic organisms. We
deployed semipermeable membrane passive sampling devices
(SPMDs) and collected resident crayfish (Pacifastacus
leniusculus) at nine locations within and outside of the
Portland Harbor Superfund Mega-site in Portland, OR.
Study results show that crayfish and aqueous phase samples
collected within the Mega-site had PAH profiles enriched in
high molecular weight PAHs and that freely dissolved PAH
profiles tended to be more populated by low molecular weight
PAHs compared to crayfish tissues. Results also show that of
several modeling approaches, a two-factor partial least-squares (PLS) calibration model using detection limit substitution
provided the best predictive power for estimating PAH concentrations in crayfish, where the model explained ≥72% of the
variation in the data set and provided predictions within ∼3× of measured values. Importantly, PLS calibration provided a means
to estimate PAH concentrations in tissues when concentrations were below detection in the freely dissolved phase. The impact of

Maturation characteristics and life-history strategies of the Pacific lamprey, Entosphenus tridentatus
Lampreys (Petromyzontiformes) have persisted over millennia and now suffer a recent decline in abundance. Complex life histories may have factored in their persistence; anthropogenic perturbations in their demise. The complexity of life histories of lampreys is not understood, particularly for the anadromous Pacific lamprey, Entosphenus tridentatus Gairdner, 1836. Our goals were to describe the maturation timing and associated characteristics of adult Pacific lamprey, and to test the null hypothesis that different life histories do not exist. Females exhibited early vitellogenesis – early maturation stages; males exhibited spermatogonia – spermatozoa. Cluster analyses revealed an “immature” group and a “maturing–mature” group for each sex. We found statistically significant differences between these groups in the relationships between (i) body mass and total length in males; (ii) Fulton’s condition factor and liver lipids in males; (iii) the gonadosomatic index (GSI) and liver lipids in females; (iv) GSI and total length in females; (v) mean oocyte diameter and liver lipids; and (vi) mean oocyte diameter and GSI. We found no significant difference between the groups in the relationship of muscle lipids and body mass. Our analyses support rejection of the hypothesis of a single life history. We found evidence for an “ocean-maturing” life history that would likely spawn within several weeks of entering fresh water, in addition to the formerly recognized life history of spending 1 year in fresh water prior to spawning—the “stream-maturing” life history. Late maturity, semelparity, and high fecundity suggest that Pacific lamprey capitalize on infrequent opportunities for reproduction in highly variable environments.Keywords: Primitive, Petromyzontiformes, Life histor

Incidence of male intersex in adult Pacific lamprey (Entosphenus tridentatus), with a brief discussion of intersex vs. hermaphroditism in lampreys (Petromyzontiformes)
We report the incidence of male intersex in adult Pacific lamprey (Entosphenus tridentatus (Richardson, 1836)) during their pre-spawning migration in fresh water. Although “hermaphrodites” have been suggested in other adult lampreys, this is the first detailed description and discussion of this phenomenon. A total of 0.5% of our adult Pacific lamprey from Willamette Falls (2 out of 427 adults) were intersex, with oocytes in the testes. This phenomenon was identifiable only by histological examination. The testes of the intersex males were immature, in the beginning stages of meiosis. One intersex male possessed primary growth or perinucleolar stage oocytes loosely interspersed throughout the testes, and the other possessed at least 6 mid-vitellogenic oocytes (0.6 mm, mean long diameter) separate from the testes. Effect of thrombin peptide 508 (TP508) on bone healing during distraction osteogenesis in rabbit tibia
Thrombin-related peptide 508 (TP508) accelerates bone regeneration during distraction osteogenesis (DO). We have examined the effect of TP508 on bone regeneration during DO by immunolocalization of Runx2 protein, a marker of osteoblast differentiation, and of osteopontin (OPN) and bone sialoprotein (BSP), two late markers of the osteoblast lineage. Distraction was performed in tibiae of rabbits over a period of 6 days. TP508 (30 or 300 μg) or vehicle was injected into the distraction gap at the beginning and end of the distraction period. Two weeks after active distraction, tissue samples were harvested and processed for immunohistochemical analysis. We also tested the in vitro effect of TP508 on Runx2 mRNA expression in osteoblast-like (MC3T3-E1) cells by polymerase chain reaction analysis. Runx2 and OPN protein were observed in preosteoblasts, osteoblasts, osteocytes of newly formed bone, blood vessel cells and many fibroblast-like cells of the soft connective tissue. Immunostaining for BSP was more restricted to osteoblasts and osteocytes. Significantly more Runx2- and OPN-expressing cells were seen in the group treated with 300 μg TP508 than in the control group injected with saline or with 30 μg TP508. However, TP508 failed to increase Runx2 mRNA levels significantly in MC3T3-E1 cells after 2–3 days of exposure. Our data suggest that TP508 enhances bone regeneration during DO by increasing the proportion of cells of the osteoblastic lineage. Clinically, TP508 may shorten the healing time during DO; this might be of benefit when bone regeneration is slow
Pollutants Increase Song Complexity and the Volume of the Brain Area HVC in a Songbird
Environmental pollutants which alter endocrine function are now known to decrease vertebrate reproductive success. There is considerable evidence for endocrine disruption from aquatic ecosystems, but knowledge is lacking with regard to the interface between terrestrial and aquatic ecosystems. Here, we show for the first time that birds foraging on invertebrates contaminated with environmental pollutants, show marked changes in both brain and behaviour. We found that male European starlings (Sturnus vulgaris) exposed to environmentally relevant levels of synthetic and natural estrogen mimics developed longer and more complex songs compared to control males, a sexually selected trait important in attracting females for reproduction. Moreover, females preferred the song of males which had higher pollutant exposure, despite the fact that experimentally dosed males showed reduced immune function. We also show that the key brain area controlling male song complexity (HVC) is significantly enlarged in the contaminated birds. This is the first evidence that environmental pollutants not only affect, but paradoxically enhance a signal of male quality such as song. Our data suggest that female starlings would bias their choice towards exposed males, with possible consequences at the population level. As the starling is a migratory species, our results suggest that transglobal effects of pollutants on terrestrial vertebrate physiology and reproduction could occur in birds
Dimerization of Translationally Controlled Tumor Protein Is Essential For Its Cytokine-Like Activity
BACKGROUND:Translationally Controlled Tumor Protein (TCTP) found in nasal lavage fluids of allergic patients was named IgE-dependent histamine-releasing factor (HRF). Human recombinant HRF (HrHRF) has been recently reported to be much less effective than HRF produced from activated mononuclear cells (HRFmn). METHODS AND FINDINGS:We found that only NH(2)-terminal truncated, but not C-terminal truncated, TCTP shows cytokine releasing activity compared to full-length TCTP. Interestingly, only NH(2)-terminal truncated TCTP, unlike full-length TCTP, forms dimers through intermolecular disulfide bonds. We tested the activity of dimerized full-length TCTP generated by fusing it to rabbit Fc region. The untruncated-full length protein (Fc-HrTCTP) was more active than HrTCTP in BEAS-2B cells, suggesting that dimerization of TCTP, rather than truncation, is essential for the activation of TCTP in allergic responses. We used confocal microscopy to evaluate the affinity of TCTPs to its putative receptor. We detected stronger fluorescence in the plasma membrane of BEAS-2B cells incubated with Del-N11TCTP than those incubated with rat recombinant TCTP (RrTCTP). Allergenic activity of Del-N11TCTP prompted us to see whether the NH(2)-terminal truncated TCTP can induce allergic airway inflammation in vivo. While RrTCTP had no influence on airway inflammation, Del-N11TCTP increased goblet cell hyperplasia in both lung and rhinal cavity. The dimerized protein was found in sera from allergic patients, and bronchoalveolar lavage fluids from airway inflamed mice. CONCLUSIONS:Dimerization of TCTP seems to be essential for its cytokine-like activity. Our study has potential to enhance the understanding of pathogenesis of allergic disease and provide a target for allergic drug development
Defining Global Gene Expression Changes of the Hypothalamic-Pituitary-Gonadal Axis in Female sGnRH-Antisense Transgenic Common Carp (Cyprinus carpio)
BACKGROUND: The hypothalamic-pituitary-gonadal (HPG) axis is critical in the development and regulation of reproduction in fish. The inhibition of neuropeptide gonadotropin-releasing hormone (GnRH) expression may diminish or severely hamper gonadal development due to it being the key regulator of the axis, and then provide a model for the comprehensive study of the expression patterns of genes with respect to the fish reproductive system. METHODOLOGY/PRINCIPAL FINDINGS: In a previous study we injected 342 fertilized eggs from the common carp (Cyprinus carpio) with a gene construct that expressed antisense sGnRH. Four years later, we found a total of 38 transgenic fish with abnormal or missing gonads. From this group we selected the 12 sterile females with abnormal ovaries in which we combined suppression subtractive hybridization (SSH) and cDNA microarray analysis to define changes in gene expression of the HPG axis in the present study. As a result, nine, 28, and 212 genes were separately identified as being differentially expressed in hypothalamus, pituitary, and ovary, of which 87 genes were novel. The number of down- and up-regulated genes was five and four (hypothalamus), 16 and 12 (pituitary), 119 and 93 (ovary), respectively. Functional analyses showed that these genes involved in several biological processes, such as biosynthesis, organogenesis, metabolism pathways, immune systems, transport links, and apoptosis. Within these categories, significant genes for neuropeptides, gonadotropins, metabolic, oogenesis and inflammatory factors were identified. CONCLUSIONS/SIGNIFICANCE: This study indicated the progressive scaling-up effect of hypothalamic sGnRH antisense on the pituitary and ovary receptors of female carp and provided comprehensive data with respect to global changes in gene expression throughout the HPG signaling pathway, contributing towards improving our understanding of the molecular mechanisms and regulative pathways in the reproductive system of teleost fish
