    Hyperpolarized xenon-129 production and applications

    Hyperpolarized 3He and 129Xe were initially developed and used in the nuclear physics community. Lately they are primarily used in Medical Resonance Imaging (MRI). Although first MRI polarized gas images were acquired using 129Xe, the research community has focused mostly on 3He, due to the well-known polarizing methods and higher polarization numbers achieved. The main purpose of this thesis is to present a novel design of a large-scale SEOP polarizer for producing large quantities of highly polarized 129Xe. High Rb-Xe spin-exchange rates through long-lived van de Waals molecules at low total pressure, implemented in a novel counterflow polarizer design, resulted in xenon polarization as high as 50% for 1.2 liters/hour, with a maximum of 64% for 0.3 l/h. We characterized and improved the polarization process by finding the optimum operating parameters of the polarizer. Two new methods to efficiently use high-power diode lasers are described: a new optical arrangement for a better beam shaping of fiber coupled lasers and the first external-cavity spectrum narrowing of a stack of laser diode arrays. A new accumulation technique for the hyperpolarized xenon was developed and full recovery of polarization after a freeze-thaw cycle was demonstrated for the first time. Two approaches for xenon delivery, frozen and gas states, were developed. Hyperpolarized xenon transportation to Brigham and Women\u27s Hospital was successfully accomplished for collaborative research. First MRI images using hyperpolarized xenon acquired at BWH are presented. Final chapter is focused on describing a low field human MRI scanner using hyperpolarized 3He. We built a human scale imager with open access for orientational studies of the lung functionality. Horizontal and vertical human lung images were acquired as a first stage of this project

    How Teachers Can Use Large Language Models and Bloom's Taxonomy to Create Educational Quizzes

    Question generation (QG) is a natural language processing task with an abundance of potential benefits and use cases in the educational domain. In order for this potential to be realized, QG systems must be designed and validated with pedagogical needs in mind. However, little research has assessed or designed QG approaches with the input from real teachers or students. This paper applies a large language model-based QG approach where questions are generated with learning goals derived from Bloom's taxonomy. The automatically generated questions are used in multiple experiments designed to assess how teachers use them in practice. The results demonstrate that teachers prefer to write quizzes with automatically generated questions, and that such quizzes have no loss in quality compared to handwritten versions. Further, several metrics indicate that automatically generated questions can even improve the quality of the quizzes created, showing the promise for large scale use of QG in the classroom setting.Comment: 8 pages, 8 figures.     How Useful are Educational Questions Generated by Large Language Models?

    Controllable text generation (CTG) by large language models has a huge potential to transform education for teachers and students alike. Specifically, high quality and diverse question generation can dramatically reduce the load on teachers and improve the quality of their educational content. Recent work in this domain has made progress with generation, but fails to show that real teachers judge the generated questions as sufficiently useful for the classroom setting; or if instead the questions have errors and/or pedagogically unhelpful content. We conduct a human evaluation with teachers to assess the quality and usefulness of outputs from combining CTG and question taxonomies (Bloom's and a difficulty taxonomy). The results demonstrate that the questions generated are high quality and sufficiently useful, showing their promise for widespread use in the classroom setting.Comment: Accepted to AIED Late Breaking Results 2023 - to be published in their proceeding

    Unmodifiable variables related to thyroid cancer incidence

    The incidence of thyroid cancer is significantly different between male and female patients. Thyroid cancer is also the only form of cancer where age can be considered a staging variable. Identifying biological prognostic factors such as age or sex is important as it helps select an optimal personalized therapy. The present analysis is an observational, prospective study that enrolled all patients with thyroid disease who were operated upon at a single center. The study aimed to determine the most frequent age at presentation, the predominance of one sex over the other, the incidence of malignant thyroid disease, and the relative risk for each sex to develop thyroid carcinoma. The incidence of thyroid carcinoma was higher for women than for men, with a higher relative risk in the female subgroup. Incidence was also highest in the 50-60-year-old group. Given that studies show better survival for women and for younger patients, even when presenting with advanced disease, compared with older, male patients, such prognostic indicators should be a factor in the treatment decision

    Does sex of the patient play a role in survival for MSI colorectal cancer?

    Microsatellite instability (MSI) is a feature of colorectal tumors that develops as a result of inactivation of the DNA mismatch repair system. It is found in about 15% of all colorectal cancers and is an important prognostic molecular marker when assessing patients with colorectal cancer. It can influence prognosis and treatment decisions in both the advanced and early stages. Although in early stages this marker suggests a favorable prognosis and presents an important argument against adjuvant treatment in stage II disease, in metastatic stages it no longer associated with such an optimistic outcome. The present trial is a prospective, single-center study which included 122 colorectal cancer patients who were tested for MSI using immunohistochemistry. The trial included patients with stage II to IV colorectal cancer, treated in the Prof. Dr. Agrippa Ionescu Emergency Hospital, Bucharest, Romania. Follow-up data were collected during a 24-month period. The study attempted to determine whether differences exist in overall survival for MSI (microsatellite instability) vs. MSS (microsatellite stable) colorectal cancer and to ascertain whether sex of the patient influences prognosis in MSI patients, irrespective of stage or treatment. Results demonstrated no significant differences in survival for MSI vs MSS colorectal patients, and patients’ gender proved not to influence the outcome in MSI patients

    On the generalized Freedman-Townsend model

    Consistent interactions that can be added to a free, Abelian gauge theory comprising a finite collection of BF models and a finite set of two-form gauge fields (with the Lagrangian action written in first-order form as a sum of Abelian Freedman-Townsend models) are constructed from the deformation of the solution to the master equation based on specific cohomological techniques. Under the hypotheses of smoothness in the coupling constant, locality, Lorentz covariance, and Poincare invariance of the interactions, supplemented with the requirement on the preservation of the number of derivatives on each field with respect to the free theory, we obtain that the deformation procedure modifies the Lagrangian action, the gauge transformations as well as the accompanying algebra. The interacting Lagrangian action contains a generalized version of non-Abelian Freedman-Townsend model. The consistency of interactions to all orders in the coupling constant unfolds certain equations, which are shown to have solutions.Comment: LaTeX, 62 page

    Identification of inhibitors of Plasmodium falciparum phosphoethanolamine methyltransferase using an enzyme-coupled transmethylation assay

    <p>Abstract</p> <p>Background</p> <p>The phosphoethanolamine methyltransferase, PfPMT, of the human malaria parasite <it>Plasmodium falciparum</it>, a member of a newly identified family of phosphoethanolamine methyltransferases (PMT) found solely in some protozoa, nematodes, frogs, and plants, is involved in the synthesis of the major membrane phospholipid, phosphatidylcholine. PMT enzymes catalyze a three-step S-adenosylmethionine-dependent methylation of the nitrogen atom of phosphoethanolamine to form phosphocholine. In <it>P. falciparum</it>, this activity is a limiting step in the pathway of synthesis of phosphatidylcholine from serine and plays an important role in the development, replication and survival of the parasite within human red blood cells.</p> <p>Results</p> <p>We have employed an enzyme-coupled methylation assay to screen for potential inhibitors of PfPMT. In addition to hexadecyltrimethylammonium, previously known to inhibit PfPMT, two compounds dodecyltrimethylammonium and amodiaquine were also found to inhibit PfPMT activity <it>in vitro</it>. Interestingly, PfPMT activity was not inhibited by the amodiaquine analog, chloroquine, or other aminoquinolines, amino alcohols, or histamine methyltransferase inhibitors. Using yeast as a surrogate system we found that unlike wild-type cells, yeast mutants that rely on PfPMT for survival were sensitive to amodiaquine, and their phosphatidylcholine biosynthesis was inhibited by this compound. Furthermore NMR titration studies to characterize the interaction between amoidaquine and PfPMT demonstrated a specific and concentration dependent binding of the compound to the enzyme.</p> <p>Conclusion</p> <p>The identification of amodiaquine as an inhibitor of PfPMT <it>in vitro </it>and in yeast, and the biophysical evidence for the specific interaction of the compound with the enzyme will set the stage for the development of analogs of this drug that specifically inhibit this enzyme and possibly other PMTs.</p

    Co-Evaluation of Pattern Matching Algorithms on IoT Devices with Embedded GPUs

    Pattern matching is an important building block for many security applications, including Network Intrusion Detection Systems (NIDS). As NIDS grow in functionality and complexity, the time overhead and energy consumption of pattern matching become a significant consideration that limits the deployability of such systems, especially on resource-constrained devices.\ua0On the other hand, the emergence of new computing platforms, such as embedded devices with integrated, general-purpose Graphics Processing Units (GPUs), brings new, interesting challenges and opportunities for algorithm design in this setting: how to make use of new architectural features and how to evaluate their effect on algorithm performance. Up to now, work that focuses on pattern matching for such platforms has been limited to specific algorithms in isolation.In this work, we present a systematic and comprehensive benchmark that allows us to co-evaluate both existing and new pattern matching algorithms on heterogeneous devices equipped with embedded GPUs, suitable for medium- to high-level IoT deployments. We evaluate the algorithms on such a heterogeneous device, in close connection with the architectural features of the platform and provide insights on how these features affect the algorithms\u27 behavior. We find that, in our target embedded platform, GPU-based pattern matching algorithms have competitive performance compared to the CPU and consume half as much energy as the CPU-based variants.\ua0Based on these insights, we also propose HYBRID, a new pattern matching approach that efficiently combines techniques from existing approaches and outperforms them by 1.4x, across a range of realistic and synthetic data sets. Our benchmark details the effect of various optimizations, thus providing a path forward to make existing security mechanisms such as NIDS deployable on IoT devices
