    Aktivitas Reactive Oxygen Species Makrofag Akibat Stimulasi Gel Lidah Buaya Pada Infeksi Salmonella Typhimurium

    Reactive Oxygen Species (ROS) merupakan salah satu lethal chemical yang dapatmembunuh dan mengeliminasi bakteri pada sel fagosit. Lidah Buaya (Aloevera) banyak dipakai sebagai pengobatan tradisional, tetapi belum ada buktiilmiah sampai tingkat seluler apalagi subseluler dalam hal efek imunostimulanpada penyakit infeksi. Tujuan penelitian ini adalah untuk mengetahui aktivitasimunostimulan dari gel lidah buaya yang ditunjukkan oleh aktivitas ROS makrofagsecara in vivo terhadap infeksi bakteri patogen Salmonella typhimurium. Sebanyak24 ekor mencit BABL/c betina umur 8-10 minggu berat 20-30 gram dikelompokkansecara acak menjadi empat kelompok, masing-masing kelompok enam ekor.Kelompok kontrol tidak diberi gel Aloe vera, sementara kelompok P1, P2, dan P3berturut-turut diberi gel Aloe vera 0,5 ml/ekor/hari; 1,0 ml/ekor/hari, dan 1,5ml/ekor/hari. Pemberian gel Aloe vera dilakukan selama sembilan hari. Pada harike-6, mencit diinfeksi bakteri patogen Salmonella typhimurium intraperitoneal105 CFU. Selanjutnya pada hari ke-10 mencit didislokasi dan dibedah, diambilmakrofag dari peritoneum untuk dianalisis produksi ROS-nya. Hasil penelitianmenunjukkan bahwa pemberian gel Aloe vera berpengaruh signi..ikan terhadappeningkatan produksi ROS makrofag mencit BALB/c yang diinfeksi Salmonellatyphimurium.     Pengaruh Suplementasi Madu Kelengkeng Terhadap Kadar Tsa Dan Mda Tikus Putih Yang Diinduksi Timbal (Pb)

    Timbal (Pb) merupakan salah satu logam berat yang berasal dari emisi pembakaran bahan bakar. Peningkatan penggunaan bahan bakar pada mesin industri dan kendaraan bermotor menyebabkan peningkatan kadar Pb di udara. Masuknya Pb ke dalam tubuh akan mengganggu keseimbangan molekul lain sehingga menjadi radikal bebas. Ketidakseimbangan antara radikal bebas dengan antioksidan menyebabkan stres oksidatif yang ditandai dengan menurunnya total satus antioksidan (TSA) dan meningkatnya malondialdehid (MDA). Madu kelengkeng adalah suplemen kesehatan yang mengandung flavonoid, vitamin C, vitamin E dan beta karoten yang berperan sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui pengaruh madu kelengkeng terhadap kadar TSA dan MDA darah tikus putih yang diinduksi Pb. Desain penelitian ini adalah eksperimental dengan 25 ekor tikus putih jantan galur Wistar yang dibagi dalam 5 kelompok, yaitu kelompok A (tikus normal), B (tikus dengan induksi Pb 10 mg/kgBB) dan C, D, E merupakan kelompok perlakuan suplementasi madu secara berturut-turut 0,45, 0,9, 1,8 ml/200 gramBB dan induksi Pb 10 mg/kgBB selama 14 hari. Data TSA dan MDA dianalisis menggunakan one way anova dan uji LSD untuk mengetahui perbedaan setiap kelompok. Hasil penelitian menunjukkan madu dengan dosis 1,8 ml/200 gramBB dapat meningkatkan kadar TSA dan menurunkan kadar MDA secara signifikan. Simpulan dari penelitian ini adalah madu dapat meningkatkan kadar TSA dan menurunkan kadar MDA pada tikus putih yang diinduksi Pb. Lead (Pb) is heavy metal which comes from waste fuel emissions. The imbalance between free radicals and antioxidants cause oxidative stress which characterized by decreased total antioxidant statue (TAS) and increased malondialdehid (MDA). Longan honey is health suplement which contained flavonoids, vitamin C, vitamin E and beta carotene. This study aims to determine the effect of longan honey against TAS and MDA levels of blood Pb-induced. This is an experimental research design with 25 white male rats Wistar, divided into 5 groups: group A (normal), B (rats with induced Pb 10 mg / kg) and the C, D, E is a group of honey supplementation treatment respectively 0.45, 0.9, 1.8 ml / 200 gramBB and induction of Pb 10 mg / kg for 14 days. TAS and MDA data were analyzed using one-way ANOVA and LSD test to determine differences in each group. The results showed that honey with a dose of 1.8 ml / 200 gramBB could increase levels of TAS and MDA levels were significantly lowered.     Progesterone reduces depression-like behavior in a murine model of Alzheimer’s Disease

    Although anxiety and depression are not the core symptoms of Alzheimer’s Disease (AD), there are changes observed in mood in those with AD, as well as in the aging population. Anxiety and depression may be influenced by progesterone P4 and/or its neuroactive metabolites, dihydroprogesterone (DHP) and 5α-pregnan-3α-ol-20-one (3α,5α-THP). To begin to investigate progestogens’ role in AD, a double transgenic mouse model of early-onset familial AD that co-overexpresses mutant forms of amyloid precursor protein (APPswe) and presenilin 1 Δ exon 9 mutation was utilized. As such, the effects of long-term (from 6 to 12 months of age) administration of P4 to ovariectomized (ovx) wildtype and APPswe+PSEN1Δe9 mice for changes in affective behavior was investigated. APPswe+PSEN1Δ9 mutant mice had increased anxiety-like (i.e., increased emergence latencies, decreased time spent on the open quadrants of the elevated zero maze) and increased depressive-like behavior (i.e., increased time spent immobile) than did wildtype mice. Compared to vehicle-administration, P4 administration (which produced physiological circulating P4, DHP, and 3α,5α-THP levels, particularly in the wildtype mice) decreased depressant-like behavior in the forced swim test. These effects occurred independent of changes in general motor behavior/coordination, pain threshold, and plasma corticosterone levels. Thus, the APPswe+PSEN1Δ9 mutation alters affective behavior, and P4 treatment reversed depressive-like behavior
