    Role of Polyamines in the Response to Salt Stress of Tomato

    Plants irrigated with saline solutions undergo osmotic and oxidative stresses, which affect their growth, photosynthetic activity and yield. Therefore, the use of saline water for irrigation, in addition to the increasing soil salinity, is one of the major threats to crop productivity worldwide. Plant tolerance to stressful conditions can be improved using different strategies, i.e., seed priming and acclimation, which elicit morphological and biochemical responses to overcome stress. In this work, we evaluated the combined effect of priming and acclimation on salt stress response of a tomato cultivar (Solanum lycopersicum L.), very sensitive to salinity. Chemical priming of seeds was performed by treating seeds with polyamines (PAs): 2.5 mM putrescine (PUT), 2.5 mM spermine (SPM) and 2.5 mM spermidine (SPD). Germinated seeds of primed and non-primed (controls) were sown in non-saline soil. The acclimation consisted of irrigating the seedlings for 2 weeks with tap water, followed by irrigation with saline and non-saline water for 4 weeks. At the end of the growth period, morphological, physiological and biochemical parameters were determined. The positive effects of combined treatments were evident, when primed plants were compared to non-primed, grown under the same conditions. Priming with PAs improved tolerance to salt stress, reduced the negative effects of salinity on growth, improved membrane integrity, and increased photosynthetic pigments, proline and enzymatic and non-enzymatic antioxidant responses in all salt-exposed plants. These results may open new perspectives and strategies to increase tolerance to salt stress in sensitive species, such as tomato

    Plants grown in saline soils undergo osmotic and oxidative stresses, affecting growth and photosynthesis and, consequently, the yield. Therefore, the increase in soil salinity is a major threat to crop productivity worldwide. Plant’s tolerance can be ameliorated by applying simple methods that induce them to adopt morphological and physiological adjustments to counteract stress. In this work, we evaluated the effects of seed priming on salt stress response in three cultivars of rapeseed (Brassica napus L.) that had different tolerance levels. Seed chemical priming was performed with 2.5 mM spermine (SPM), 5 mM spermidine (SPD), 40 mM NaCl and 2.5 mM Ca (NO3)2. Primed and not primed seeds were sown on saline and not saline (controls) media, and morphological and physiological parameters were determined. Since SPD treatment was effective in reducing salinity negative effects on growth, membrane integrity and photosynthetic pigments, we selected this priming to further investigate plant salt stress response. The positive effects of this seed treatment on growth and physiological responses were evident when primed plants were compared to not primed ones, grown under the same saline conditions. SPD priming ameliorated the tolerance towards saline stress, in a genotype-independent manner, by increasing photosynthetic pigments, proline amounts and antioxidant responses in all cultivars exposed to salt. These results may open new perspectives for crop productivity in the struggle against soil salinization

    Celebrity effect in public health and bioethics: An \u201cItalian case\u201d in endometrio-sis prevention communication

    Objective: To explore endometriosis-related digital seeking behaviour in Italy, after the showgirl Rossella Brescia announced to be affected by this gynecological disease. Methods: We performed a retrospective search of web-activities related to endometriosis, carried out in Italy, in the last 5 years, using Google Trends with \u201cendometriosis\u201d as keyword and \u201csearch topic\u201d as search strategy. We used the section Google News in order to check the impact of media coverage on web behaviour. Results: Web-activities related to Rossella Brescia and endometriosis correlated in a statistically borderline significant way, even though peaks in endometriosis searches coincided with bursts in digital interest towards Rossella Brescia. Among the endometriosis-associated rising queries, \u201cRossella Brescia endometriosis\u201d and \u201cRossella Brescia\u201d were two of the most searched. Among Rossella Brescia-associated rising queries, \u201cRossella Brescia disease\u201d yielded the first place (break out), whilst \u201cRossella Brescia endometriosis\u201d the fourth place (breakout), \u201cendometriosis\u201d the fifth place (breakout) and \u201cRossella Brescia ill\u201d the ninth place (breakout). Conclusions: Practitioners should become aware of the importance of new media in communicating the disease with their patients and workers in the field of public health should strengthen their presence online, exploiting celebrity effect in order to disseminate rigorous but accessible information and raise public awareness against the disease, methods of diagnosis and prevention

    Reduction in Fatigue Symptoms Following the Administration of Nutritional Supplements in Patients with Multiple Sclerosis

    Despite recent advances in immune-modulatory drugs, pharmacological therapies have been proven ineffective in severe presentations of multiple sclerosis (MS), including secondary progressive MS. At present, therapeutic interventions' performance is primarily focused on ameliorating symptoms to improve the patient's quality of life (QOL). Among complementary treatments, nutrition has been considered a decisive factor to control symptoms and enhance the wellness of MS patients. Although no special diets are associated with MS, the impact of diet and dietary supplements on the course of progressive forms of the disease has been studied during the last few years. Fatigue is among the most common and disabling symptoms reported by MS patients. Fatigue has been defined in the Multiple Sclerosis Council for Clinical Practice Guidelines (MSCCPG, 1998) as a "subjective lack of physical and/or mental energy that the individual perceives as an interference with habitual and desired activities". This study aimed to compare the psychometric functioning of the "Fatigue Severity Scale" (FSS) and the "Modified Fatigue Impact Scale" (MFIS) in our sample of people with MS. Specifically, during chronic treatment, the change in these two parameters with two vitamin-rich dietary supplements (Citozym® and Ergozym®) was evaluated. The impact of these nutritional supplements revealed differences in antioxidant and anti-inflammatory parameters among the volunteers in the treatment group, with a subsequent improvement in fatigue. In conclusion, the results obtained have confirmed the effectiveness of complementary nutritional therapies, evaluated essentially based on hematological biomarkers, through which it is possible to act on disability to improve the QOL of MS patients

    Has the ice bucket challenge really increased people's awareness of amyotrophic lateral sclerosis? Insights and ethical implications from Google Trends and wikipedia: A 2 years-follow up

    Amyotrophic lateral sclerosis (ALS), commonly known as Lou Gehrig's disease, is the most common form of motor neuron diseases (MNDs), causing the death of neurons controlling voluntary muscles. In order to increase public awareness and to promote fundraising, a charity activity known as the "Ice Bucket Challenge" (IBC), initially started only as a joke, was launched in July and August 2014, by Pete Frates, ex Boston College baseball player and himself suffering from ALS. Immediately, it became "viral" among social media and social network users, worldwide. We used Google Trends (GT) and accessed to Wikipedia page to document interest towards ALS 2 years after the IBC. In coincidence with the IBC initiative, a peak in web queries could be noticed, as well as a burst in daily accesses to Wikipedia page. However, this increase in web activities (+450% for GT) was characterized by a brief memory and a short half-life: before and after IBC initiative, GT-based RSVs were 18.2\uc2\ub11.7% and 17.8\uc2\ub11.9%, respectively. Despite alleged claims of the effectiveness of social networks-based campaigns, apart from money donation and a temporarily burst of web queries and accesses to specialized web-sites, before and after the IBC the level of web-related activities has remained practically constant. The direct involvement of scientists and stakeholders, besides that of celebrities and famous people, would be of crucial importance. Only in this way, initiatives such as the IBC could turn from mere entertaining events even though money-attracting into real educational moments. Otherwise, they would be other missed opportunities

    Layered Double Hydroxides as a Drug Delivery Vehicle for S-Allyl-Mercapto-Cysteine (SAMC)

    The intercalations of anionic molecules and drugs in layered double hydroxides (LDHs) have been intensively investigated in recent years. Due to their properties, such as versatility in chemical composition, good biocompatibility, high density and protection of loaded drugs, LDHs seem very promising nanosized systems for drug delivery. In this work, we report the intercalation of S-allyl-mercapto-cysteine (SAMC), which is a component of garlic that is well-known for its anti-tumor properties, inside ZnAl-LDH (hereafter LDH) nanostructured crystals. In order to investigate the efficacy of the intercalation and drug delivery of SAMC, the intercalated compounds were characterized using X-ray powder diffraction (XRD), Fourier-transform infrared spectroscopy (FT-IR) and scanning electron microscopy (SEM). The increase in the interlayer distance of LDH from 8.9 Å, typical of the nitrate phase, to 13.9 Å indicated the intercalation of SAMC, which was also confirmed using FT-IR spectra. Indeed, compared to that of the pristine LDH precursor, the spectrum of LDH-SAMC was richly structured in the fingerprint region below 1300 cm−1, whose peaks corresponded to those of the functional groups in the SAMC molecular anion. The LDH-SAMC empirical formula, obtained from UV-Vis spectrophotometry and thermogravimetric analysis, was [Zn0.67Al0.33(OH)2]SAMC0.15(NO3)0.18·0.6H2O. The morphology of the sample was investigated using SEM: LDH-SAMC exhibited a more irregular size and shape of the flake-like crystals in comparison with the pristine LDH, with a reduction in the average crystallite size from 3 µm to about 2 µm. In vitro drug release studies were performed in a phosphate buffer solution at pH 7.2 and 37 °C and were analyzed using UV-Vis spectrophotometry. The SAMC release from LDH-SAMC was initially characterized by a burst effect in the first four hours, during which, 32% of the SAMC is released. Subsequently, the release percentage increased at a slower rate until 42% after 48 h; then it stabilized at 43% and remained constant for the remaining period of the investigation. The LDH-SAMC complex that was developed in this study showed the improved efficacy of the action of SAMC in reducing the invasive capacity of a human hepatoma cell line

    Introduction The ERAS protocol (Enhanced Recovery After Surgery) is a multimodal pathway aimed to reduce surgical stress and to allow a rapid postoperative recovery. Application of the ERAS protocol to colorectal cancer surgery has been limited to a minority of hospitals in Italy. To promote the systematic adoption of ERAS in the entire regional hospital network in Piemonte an Audit and Feedback approach (A&F) has been adopted together with a cluster randomised trial to estimate the true impact of the protocol on a large, unselected population. Methods A multicentre stepped wedge cluster randomised trial is designed for comparison between standard perioperative management and the management according to the ERAS protocol. The primary outcome is the length of hospital stay (LOS). Secondary outcomes are: incidence of postoperative complications, time to patients' recovery, control of pain and patients' satisfaction. With an A&F approach the adherence to the ERAS items is monitored through a dedicated area in the study web site. The study includes 28 surgical centres, stratified by activity volume and randomly divided into four groups. Each group is randomly assigned to a different activation period of the ERAS protocol. There are four activation periods, one every 3 months. However, the planned calendar and the total duration of the study have been extended by 6 months due to the COVID-19 pandemic. The expected sample size of about 2200 patients has a high statistical power (98%) to detect a reduction of LOS of 1 day and to estimate clinically meaningful changes in the other endpoints. Ethics and dissemination The study protocol has been approved by the Ethical Committee of the coordinating centre and by all participating centres. Study results will be timely circulated within the hospital network and published in peer-reviewed journals. Trial registration number NCT04037787
