Biomimetic strategies for the deputization of proteoglycan functions
Proteoglycans (PGs), which have glycosaminoglycan chains attached to their protein cores, are essential for maintaining the morphology and function of healthy body tissues. Extracellular PGs perform various functions, classified into the following four categories: i) the modulation of tissue mechanical properties; ii) the regulation and protection of the extracellular matrix; iii) protein sequestration; and iv) the regulation of cell signaling. The depletion of PGs may significantly impair tissue function, encompassing compromised mechanical characteristics and unregulated inflammatory responses. Since PGs play critical roles in the function of healthy tissues and their synthesis is complex, the development of PG mimetic molecules that recapitulate PG functions for tissue engineering and therapeutic applications has attracted the interest of researchers for more than 20 years. These approaches have ranged from semisynthetic graft copolymers to recombinant PG domains produced by cells that have undergone genetic modifications. This review discusses some essential extracellular PG functions and approaches to mimicking these functions
Ameliorative effect of biosynthesized titanium dioxide nanoparticles using garlic extract on the body weight and developmental toxicity of liver in albino rats compared with chemically synthesized nanoparticles
The application of metallic nanoparticles poses risks to human and animal health. Titanium dioxide nanoparticles (TiO2NPs) are the most commonly synthesized metallic oxides in the world. Exposure to TiO2NPs can cause toxicity in the target organisms. This study aimed to evaluate the effects of green and chemical TiO2NPs on maternal and embryo-fetal livers. Green TiO2NPs using garlic extract (GTiO2NPs) and chemical TiO2NPs (CHTiO2NPs) were synthesized and characterized by x-ray powder diffraction and high-resolution transmission electron microscopy. The cytotoxicity of both chemical and green TiO2NPs was determined against HepG2 cell lines. Fifty pregnant female Albino rats were equally and randomly divided into five groups. Group 1 was kept as a control. Groups 2 and 3 were orally treated with 100 and 300 mg/kg body weight of CHTiO2NPs, respectively. Groups 4 and 5 were orally treated with 100 and 300 mg/kg of GTiO2NPs, respectively, from day 6 to 19 of gestation. All dams were euthanized on gestation day 20. All live fetuses were weighed and euthanized. Blood and tissue samples were collected for biochemical, histopathological, and Bax-immunohistochemical expression analyses. Our results indicated that garlic could be used as a reducing agent for the synthesis of TiO2NPs, and the produced NPs have no toxic effect against HepG2 cells compared with CHTiO2NPs. The maternal and fetal bodyweights were greatly reduced among the chemically TiO2NPs induced animals. The mean serum level of AST and ALT activities and the total protein level significantly increased when TiO2NPs were administered at high doses. Histologically, the CHTiO2NPs-treated groups revealed vacuolated and necrotized hepatocytes with congested and dilated blood vessels in the fetal and maternal livers. The immunohistochemistry revealed distinct positive staining of Bax expressed in the hepatocytes. Nevertheless, the biosynthesis of TiO2NPs using garlic extract had a minimal effect on the normal architecture of the liver. It could be concluded that the bioactivity of TiO2NPs can be modified by green synthesis using garlic extract. Compared to the CHTiO2NPs, the exposure to GTiO2NPs showed reduced liver damage in maternal and embryo-fetal rats
Health risk assessment of organochlorine pesticide residues in edible tissue of seafood
Fish is one of the most valuable foods with high-quality animal protein. However, aquaculture, or ingesting contaminated food, allows organochlorine pesticides (OCPs) to enter the fish's body, and therefore, it negatively impacted public health. One-hundred and twenty random samples of Clupea harengus (C. harengus), Mugil cephalus (M. cephalus), Sardinella aurita (S. aurita), Oreochromis niloticus (O. niloticus), Neptunus pelagicus (N. pelagicus) and Sepia savigngi (S. savigngi) (n = 20 each) were collected from local markets in Mansoura city, Egypt. Samples were checked to see whether any residues of OCPs with the application of risk assessment due to their consumption by Mansoura citizens. The findings indicated that summation hexachlorocyclohexane (∑HCH) in examined seafood samples ranged from 0.27 ± 0.13 in N. pelagicus to 61.61 ± 52.03 μg.kg−1 in S. aurita. Also, the γ-HCH isomer was considered the more prominent among isomers. Hexachlorobenzene (HCB) was found in five different species, with mean values of 2.03 ± 1.85, 1.5.7 ± 1.17, 0.94 ± 0.87, 0.35 ± 0.06, and 0.18 ± 0.06 μg.kg−1 in C. harengus, S. aurita, M. cephlaus, O. niloticus, and S. savigngi. Moreover, summation of Heptachlors (∑HPTs) was 10.19 ± 7.63, 1.27 ± 0.26, 2.58 ± 0.11, 0.95 ± 0.12, 0.21 ± 0.11 and 0.32 ± 0.03 μg.kg−1 of wet weight in examined C. harengus, M. cephlaus, S. aurita, O. niloticus, N. pelagicus, and S. savigngi. Aldrin and dieldrin residues were 3.75 ± 1.31 and 4.86 ± 1.33 μg.kg−1 in C. harengu, meanwhile they were 1.61 ± 0.77 and 0.78 ± 0.04 μg.kg−1in M. cephalus. Dichlorodiphenyldichloroethylene (pp-DDE) was dominant in all examined species within different concentrations 5.08 ± 4.12, 0.98 ± 0.10, 3.07 ± 0.91, 0.93 ± 0.27, 0.08 ± 0.01 and 0.35 ± 0.02 μg.kg−1 in C. harengus, M. cephlaus, S. aurita, O. niloticus, N. pelagicus and S. savigngi, respectively. We concluded that all examined seafood samples were lower than the recommended maximum residue limit. Also, the estimated daily intake was less than the permitted daily intake. Ozonated saline intradermal injection: promising therapy for accelerated cutaneous wound healing in diabetic rats
IntroductionThe use of ozonized water is gaining importance in medicine due to its effects on hyperglycemia and wound healing mechanisms.MethodsThis experiment was conducted to assess the impacts of intradermal administration of ozonated water on acute skin wound healing in a diabetic rat model. Sixty-four adult male Wistar rats were randomly divided into two groups: an ozonated water group (O3W) and a control group (CG). Experimental diabetes was chemically induced in the rats by the intraperitoneal administration of 60 mg/kg streptozotocin. One week later, full-thickness skin surgical wounds (1 cm2) were created between the two shoulders of the rats under general anesthesia. The wounds were then daily irrigated with normal saline (CG) or intradermally injected with 1 mL of ozonated water at 10 mg/L O3W. Wound healing was evaluated through macroscopic analysis, measuring wound size, diameter, and percentage of contraction rate before wounding and at 3, 7, 9, 12, 14, 18, 21, 24, and 28 days post-wounding. On days 7, 14, 21, and 28 after induction of the wounds, the body weights and blood glucose levels of rats (8 per group) were measured before the rats were euthanized. Moreover, the morphological structure of the tissue, vascular endothelial and transforming growth factor (VEGF and TGF) affinity and gene expression were examined.ResultsThe O3W group had significantly lower blood glucose levels and wound size and gained body weight. Additionally, epithelial vascularization, stromal edema, TGF, and VEGF gene expression significantly improved in the O3W group.DiscussionTherefore, ozonated water has the potential to enhance and promote cutaneous wound healing in diabetic rats
Risk Factors Associated with Mild Cognitive Impairment аmong Apparently Healthy People and the Role of MicroRNAs
BACKGROUND: Mild cognitive impairment (MCI) is a stage between the expected cognitive decline of normal ageing and the serious decline of dementia.
AIM: To identify risk factors and role of miRNAs associated with mild cognitive impairment (MCI) among employees.
SUBJECTS AND METHOD: A cross-sectional study was carried out on 186 employees aged between 40 and 65 years. Cognitive function was evaluated using ACEIII, MoCA, and Quick cognitive tests. Medical history and lifestyle were assessed. Family 132 & 134 miRNA expressions were assessed by real-time PCR.
RESULTS: MCI was detected among 14 / 186 (7.5%). miRNA 132 expression was the only significant miRNAs to detect MCI with low sensitivity and specificity (70%). The logistic analysis revealed that higher miRNA132 expressions, low monthly intake of; vegetables, unroasted nuts, low education and higher ALT levels were predicting factors for MCI with AOR 1.1 (1.01-3.3), 1.2 (1.04-1.43), 0.8 (0.8-0.98), 2.7 (1.9-7.4) and 1.6 (1.1-2.3) respectively.
CONCLUSION: MiRNAs expression showed low sensitivity and specificity in detecting MCI; only miRNA 132 might be used. Several modifiable factors seem to reduce the risk of MCI

The Titer of Immunity and Signs of Welfare Due to Changes in Group Size in Two Strains of Lohmann Classic Layers
This study examined the effect of the different group sizes on the titers of some immunological indicators and signs of health in two strains of lohmann layers. A total 558 layers (279 lohmann brown and 279 lohmann selected leghorn), aged 50 weeks were homogenously classified into four groups, where 360 birds (180 lohmann brown and 180 lohmann selected leghorn) in 6 cages (60 layers/cage “5 m2”) and 198 birds (99 lohmann brown and 99 lohmann selected leghorn) (33 layers/cage “2.8 m2”) with the same floor space relatively. The antibody titer of avian encephalomyelitis, avian meta-pneumonia, infectious bronchitis and mycoplasma synovia were higher in small group (33 birds) than large group size (60 birds). However, the differences didn’t reach the significance. In the other hand, the lohmann selected leghorn was more susceptible to avian encephalomyelitis, infectious bronchitis and mycoplasma synovia due to the increase in its antibody titers, while the antibody titers of avian meta-pneumonia and mycoplasma gallisepticum were higher in lohmann brown. In large group size, the scores of plumage condition were referred to the best, especially in lohmann brown. Furthermore, the changes in feet condition in lohamman brown were better than lohmann selected leghorn, especially in large group. In order to achieve the full genetic growth potential, layers must be reared under optimal group size. Therefore, any deviation of the managemental condition of layers may impair their performance, cause immunosuppression, and change their physiological responses leading to increase their susceptibility to diseases
Biofilm-Producing Ability of <i>Staphylococcus aureus</i> Obtained from Surfaces and Milk of Mastitic Cows
This study was conducted to evaluate the incidence of mastitis in 153 dairy cows and to evaluate the kinetics of adhesion of isolates obtained from surfaces and milk in comparison with the reference strain (RS), CCM 4223. The surfaces of the floor, teat cup, and cow restraints were aseptically swabbed in three replicates (n = 27). Of the total number of infected cows (n = 43), 11 samples were found to be positive for Staphylococcus aureus, 12 samples tested positive for non-aureus staphylococci, 6 samples tested positive for Streptococcus spp., and 11 samples tested positive for other bacteria (Escherichia coli, Pseudomonas spp.) or a mixed infection. The most represented pathogen in milk (11/43) and on surfaces (14/27) was S. aureus. The kinetics of adhesion of the reference strain and isolates of S. aureus on stainless steel surfaces were determined after 3, 6, 9, 12, 24, and 48 h, and 3, 6, 9, 12, and 15 days of incubation. All strains reached counts higher than 5 Log10 CFU/cm2 needed for biofilm formation, except RS (4.40 Log10 CFU/cm2). The isolates of S. aureus revealed a higher capability to form biofilm in comparison with RS during the first 3 h (p S. aureus on monitored surfaces—floor, teat cup, and cow restraints—and the frequency with which mastitis is caused by S. aureus (p S. aureus, it can result in the formation of biofilm, which is a significant virulence factor
