    Biochemical characterization and DNA repair pathway interactions of Mag1-mediated base excision repair in Schizosaccharomyces pombe

    The Schizosaccharomyces pombe mag1 gene encodes a DNA repair enzyme with sequence similarity to the AlkA family of DNA glycosylases, which are essential for the removal of cytotoxic alkylation products, the premutagenic deamination product hypoxanthine and certain cyclic ethenoadducts such as ethenoadenine. In this paper, we have purified the Mag1 protein and characterized its substrate specificity. It appears that the substrate range of Mag1 is limited to the major alkylation products, such as 3-mA, 3-mG and 7-mG, whereas no significant activity was found towards deamination products, ethenoadducts or oxidation products. The efficiency of 3-mA and 3-mG removal was 5–10 times slower for Mag1 than for Escherichia coli AlkA whereas the rate of 7-mG removal was similar to the two enzymes. The relatively low efficiency for the removal of cytotoxic 3-methylpurines is consistent with the moderate sensitivity of the mag1 mutant to methylating agents. Furthermore, we studied the initial steps of Mag1-dependent base excision repair (BER) and genetic interactions with other repair pathways by mutant analysis. The double mutants mag1 nth1, mag1 apn2 and mag1 rad2 displayed increased resistance to methyl methanesulfonate (MMS) compared with the single mutants nth1, apn2 and rad2, respectively, indicating that Mag1 initiates both short-patch (Nth1-dependent) and long-patch (Rad2-dependent) BER of MMS-induced damage. Spontaneous intrachromosomal recombination frequencies increased 3-fold in the mag1 mutant suggesting that Mag1 and recombinational repair (RR) are both involved in repair of alkylated bases. Finally, we show that the deletion of mag1 in the background of rad16, nth1 and rad2 single mutants reduced the total recombination frequencies of all three double mutants, indicating that abasic sites formed as a result of Mag1 removal of spontaneous base lesions are substrates for nucleotide excision repair, long- and short-patch BER and RR

    The importance of Einstein's geometrization philosophy, as an alternative to the least action principle, in constructing general relativity (GR), is illuminated. The role of differential identities in this philosophy is clarified. The use of Bianchi identity to write the field equations of GR is shown. Another similar identity in the absolute parallelism geometry is given. A more general differential identity in the parameterized absolute parallelism geometry is derived.     Evaluating e-Government services from a citizens' prespective: A reference process model

    Evaluating and optimizing e-government services is imperative for governments especially due to the capacity of e-services to transform public administrations and assist the interactions of governments with citizens, businesses and other government agencies. Existing widely applied evaluation approaches neglect to incorporate citizens’ satisfaction measures. Several citizen satisfaction models and indicators have been suggested in academia; however a reference process model that can assist practitioners to apply these performance measures is missing. In this paper we draw upon the evaluation approach proposed by the EU funded project CEES and propose a reference process model that captures re-usable practices for e-government evaluation from a citizens’ perspective.     Hemodynamic prediction in patent ductus arteriosus morphologies

    Patent ductus arteriosus (PDA) is a condition in which the ductus arteriosus remain opened after birth, causing the blood to shunt through from the aorta to the pulmonary artery. However, due to the complicated nature of the arterial geometry, the flow characteristic inside the PDA is not fully understood, since simplified model are commonly used in researches. This study aims to identify the hemodynamic characteristic in three different patient-specific morphologies. Computational modeling via computational fluid dynamic (CFD) is implemented to predict the blood flow behavior in different PDA morphologies. The result shows that low wall shear stress is observed at the region where the flow recirculation occurs. High wall shear stress is observed in the pulmonary artery due to increase of flow velocity at the insertion point of PDA. The PDA morphologies exhibit left-to-right shunt, which diverts approximately 10% of blood flow from the aorta to the pulmonary artery. The highest shunted blood flow is found in TR LPA morphology. High value of OSI indicates the changes of wall shear stress vector. It is observed that DS LPA has the highest area covered by OSI which presents better hemodynamic characteristic as compared to other morphology

    Predictors of COVID-19 Vaccination Rate in USA: A Machine Learning Approach

    In this study, we examine state-level features and policies that are most important in achieving a threshold level vaccination rate to curve the effects of the COVID-19 pandemic. We employ CHAID, a decision tree algorithm, on three different model specifications to answer this question based on a dataset that includes all the states in the United States. Workplace travel emerges as the most important predictor; however, the governors’ political affiliation (PA) replaces it in a more conservative feature set that includes economic features and the growth rate of COVID-19 cases. We also employ several alternative algorithms as a robustness check. Results from these checks confirm our original findings regarding workplace travels and political affiliation. The accuracy under different model specifications ranges from 80%–88%, whereas the sensitivity is between 92.5%–100%. Our findings provide actionable policy insights to increase vaccination rates and combat the COVID-19 pandemic

    Perceiving threat in others: The role of body morphology.

    People make judgments of others based on appearance, and these inferences can affect social interactions. Although the importance of facial appearance in these judgments is well established, the impact of the body morphology remains unclear. Specifically, it is unknown whether experimentally varied body morphology has an impact on perception of threat in others. In two preregistered experiments (N = 250), participants made judgments of perceived threat of body stimuli of varying morphology, both in the absence (Experiment 1) and presence (Experiment 2) of facial information. Bodies were perceived as more threatening as they increased in mass with added musculature and portliness, and less threatening as they increased in emaciation. The impact of musculature endured even in the presence of faces, although faces contributed more to the overall threat judgment. The relative contributions of the faces and bodies seemed to be driven by discordance, such that threatening faces exerted the most influence when paired with non-threatening bodies, and vice versa. This suggests that the faces and bodies were not perceived as entirely independent and separate components. Overall, these findings suggest that body morphology plays an important role in perceived threat and may bias real-world judgments

    Studies of ion transport and electrochemical properties of plasticized composite polymer electrolytes

    The composite polymer electrolytes (CPEs) composed of polyacrylonitrile (PAN) as host polymer, lithium tetraflouroborate (LiBF4) as dopant salt, dissoĂ…lved in the mixture of ethylene carbonate (EC) and dimethyl phthalate (DMP) as plasticizing solvent, with the addition of silica (SiO2) as inorganic filler were prepared by the solution casting technique. The CPE films were prepared by varying the concentrations of SiO2 from 1 to 5 wt. %. The CPE film containing 3 wt. % of SiO2 exhibits the highest ionic conductivity of 1.36 Ă— 10-2 S cm-1 at room temperature while for temperature dependence studies, the plot obtained obeyed Arrhenius rule and the calculated activation energy was 0.11 eV. The ionic conductivity of the CPEs was found to depend on the concentration of ion pairs of dopant salt as showed by FTIR spectra. The calculated value of lithium ions transport number, tLi+ for the highest conducting CPE film was 0.15. This result indicates that anionic species are the main contributor to the total conductivity of the CPE. The CPE film has an electrochemical stability higher than the non-filler film
