    Study on Durability Characteristics of Self - Compacting Concrete with Fly Ash

    This paper investigates the study of workability and durability characteristics of Self-Compacting Concrete (SCC) with Viscosity Modifying Admixture (VMA), and containing Class F fly ash. The mix design for SCC was arrived as per the Guidelines of European Federation of National Associations Representing for Concrete (EFNARC). In this investigation, SCC was made by usual ingredients such as cement, fine aggregate, coarse aggregate, water and mineral admixture fly ash at various replacement levels (10%, 20%, 30%, 40% and 50%). The super plasticizer used was Glenium B233 and the viscosity modifying agent used was Glenium Stream 2. The experiments are carried out by adopting a water-powder ratio of 0.45. Workability of the fresh concrete is determined by using tests such as: slump flow, T50, V-funnel, L-Box and U-box tests. The durability of concrete is tested by acid resistance, sulphate attack and saturated water absorption at the age of 28, 56 and 90 days

    Chemistry deals with herbal constituents are often coined as phytochemistry. Herbal constituents have profound improvements in drug discovery for several existing diseases. Many of these constituents are restricted from pharmaceutical discoveries due to two important reasons: pharmacodynamics and pharmacokinetics. There are many new technological strategies and comparisons have been studied to improve the herbal discoveries in the pharmaceutical market. This review paper will highlight historical evidence of nanogels which is the most important strategy applied to several herbal medicines with high patience compliance, delivery rate, and efficiency. Nanogels are nanoparticles combined with cross-linked polymer networks with desirable features to carry hydrophilic or hydrophobic drugs in a more stable condition. Nanogels are highly preferred substances for herbal medicine in terms of stability and rapid response to the external stimuli factors. Nanogel can facilitate the herbal products with higher cellular penetration than existing and hence, it proves to be the new dimension for both oral and transdermal drug delivery for several unmet diseases like cancer, diabetes, and chronic disorders.     ANCILLARY SERVICE REQUIREMENT BASED AUTOMATIC GENERATION CONTROL ASSESSMENT IN A DEREGULATED POWER SYSTEM WITH HES AND IPFC UNITS

    This paper intends the evaluation measures for obtaining the Ancillary ServicesRequirement (ASR) indices stand on Automatic Generation Control (AGC) in a deregulated power system.Ancillary services are vital to support the transmission of electric power from vendor to user with the responsibility of control areas and transmitting utilities within those control areas to maintain steadfast operations of the interconnected power system under deregulated environment. In this swot, Proportional Integral Derivative with derivative Filter (PIDF) is projected for the AGC loop of a two-area thermal power system with reheat bicycle mix condensation turbine. The control constraints of the PIDF controller are optimized using the Big Bang Big Crunch (BBBC) algorithm.ASR keys are computed stranded on the dynamic response of the control input deviations and the mechanical power generation deviations of each area for dissimilar nature of possible transactions. These indices designate the ancillary service requirements and are required to improve the competence of the physical operation of the power system with the augmented transmission capacity in the network. An advanced application of Hydrogen Energy Storage (HES), when coordinated with the Interline Power Flow Controller (IPFC) for the development of AGC loop of a two-area thermal power system is also painstaking. Simulation reveals that the proposed PIDF controller tuned with BBBC algorithm perk up the dynamic output response of the test system. Moreover, it can also be pragmatic that the ASR Indices are computed for a two-area thermal power system with HES and IPFC units indicates that the new advanced control for a better restoration of the power system output responses and ensure enhanced ASR indices in order to afford the superior margin of steadiness

    Peripheral refractive changes associated with myopia progression

    Purpose.: To evaluate the changes in peripheral refraction profiles associated with myopia progression and treatment modalities used in the Cambridge Anti-Myopia Study. Methods.: One hundred and seventy-seven myopes in the age range of 14 to 22 years were enrolled in the study. The mean spherical equivalent refractive error was −3.12 ± 1.87 diopters (D) and the refractive error of each participant was corrected with contact lenses. The participants were randomly assigned to one of four treatment groups, which included: altered spherical aberration and vision training, altered spherical aberration only, vision training only, and control. Peripheral refractive error was measured using an open field autorefractor in the central 60° of the retina in 10° steps. The refractive error was measured using cycloplegic autorefraction. Two-year refractive progression data and initial peripheral refraction measurements were available in 113 participants. Measurements of peripheral refraction and cycloplegic refraction were obtained at three visits over 2 years in 12-month intervals for 92 participants. Results.: All subjects showed a relative peripheral hyperopia, especially in the nasal retina. A limited magnitude of myopia progression of −0.34 ± 0.36 D over 2 years was found in each of the four groups on average. There were no significant differences in the rate of progression between any of the treatment groups (P > 0.05). Initial peripheral J45 astigmatic refractive error at 20° and 30° in the nasal retina was weakly correlated with progression of myopia over 2 years (r = −0.27, P = 0.004 and r = −0.20, P = 0.040, respectively; n = 113). The change in spherical equivalent peripheral refractive error at 30° nasal retina over time was also significantly correlated with progression of myopia especially at 24 months (r = −0.24, P = 0.017, n = 92). Conclusions.: Relative peripheral hyperopia is associated with myopia. Myopia progression may be weakly linked to changes in the peripheral refraction profiles in the nasal retina. However, a causative link between peripheral refractive error and myopia progression could not be established

    The Cambridge Anti-myopia Study: variables associated with myopia progression

    Purpose: To identify variables associated with myopia progression and to identify any interaction between accommodative function, myopia progression, age, and treatment effect in the Cambridge Anti-Myopia Study. Methods: Contact lenses were used to improve static accommodation by altering ocular spherical aberration, and vision training was performed to improve dynamic accommodation. One hundred forty-two subjects, aged 14–21 years, were recruited who had a minimum of −0.75D of myopia. Subjects were assigned to contact lens treatment only, vision training only, contact lens treatment and vision training, or control group. Spherical aberration, lag of accommodation, accommodative convergence/accommodation (AC/A) ratio, accommodative facility, ocular biometry, and refractive error were measured at regular intervals throughout the 2-year trial. Results: Ninety-five subjects completed the 24-month trial period. There was no significant difference in myopia progression between the four treatment groups at 24 months. Age, lag of accommodation, and AC/A ratio were significantly associated with myopia progression. There was a significant treatment effect at 12 months in the contact lens treatment group in younger subjects, based on a median split, aged under 16.9 years (p = 0.005). This treatment effect was not maintained over the second year of the trial. Younger subjects experienced a greater reduction in lag of accommodation with the treatment contact lens at 3 months (p = 0.03), compared to older contact lens treatment and control groups. There was no interaction between AC/A ratio and contact lens treatment effect. Conclusions: Age, lag of accommodation, and AC/A ratio were significantly associated with myopia progression. Although there was no significant treatment effect at 24 months, an interaction between age and contact lens treatment suggests younger subjects may be more amenable, at least in the short term, to alteration of the visual system using optical treatments

    Therapeutic Potential and Challenges of Targeting Receptor Tyrosine Kinase ROR1 with Monoclonal Antibodies in B-Cell Malignancies

    Based on its selective cell surface expression in chronic lymphocytic leukemia (CLL) and mantle cell lymphoma (MCL), receptor tyrosine kinase ROR1 has recently emerged as a promising target for therapeutic monoclonal antibodies (mAbs). To further assess the suitability of ROR1 for targeted therapy of CLL and MCL, a panel of mAbs was generated and its therapeutic utility was investigated.A chimeric rabbit/human Fab library was generated from immunized rabbits and selected by phage display. Chimeric rabbit/human Fab and IgG1 were investigated for their capability to bind to human and mouse ROR1, to mediate antibody-dependent cellular cytotoxicity (ADCC), complement-dependent cytotoxicity (CDC), and internalization, and to agonize or antagonize apoptosis using primary CLL cells from untreated patients as well as MCL cell lines. A panel of mAbs demonstrated high affinity and specificity for a diverse set of epitopes that involve all three extracellular domains of ROR1, are accessible on the cell surface, and mediate internalization. The mAb with the highest affinity and slowest rate of internalization was found to be the only mAb that mediated significant, albeit weak, ADCC. None of the mAbs mediated CDC. Alone, they did not enhance or inhibit apoptosis.Owing to its relatively low cell surface density, ROR1 may be a preferred target for armed rather than naked mAbs. Provided is a panel of fully sequenced and thoroughly characterized anti-ROR1 mAbs suitable for conversion to antibody-drug conjugates, immunotoxins, chimeric antigen receptors, and other armed mAb entities for preclinical and clinical studies

    Next-generation sequencing of pancreatic cyst wall specimens obtained using micro-forceps for improving diagnostic accuracy

    Background and study aims: Pancreatic cysts are common incidental findings, with an estimated prevalence of 13-15% in imaging done for other reasons. Diagnosis often relies on collection of cyst fluid, but tissue sampling using micro-forceps may allow for a more reliable diagnosis and higher yield of DNA for next-generation sequencing (NGS). The primary aim was to assess the performance of NGS in identifying mucinous cyst. The secondary aims were to assess DNA yield between the cyst fluid and cyst wall tissue, complication rate and performance of conventional investigations. Patients and methods: 24 patients referred for endoscopic ultrasound were recruited. Biopsies were taken using micro-forceps and the AmpliSeq Cancer Hotspot panel was used for NGS, a PCR assay targeting several hotspots within 50 genes, including GNAS, KRAS and VHL. Results: The concentration of DNA extracted from 24 cyst wall samples was significantly higher than in the 9/24 available matched cyst fluid samples. The sensitivity, specificity and diagnostic accuracy of NGS for diagnosing mucinous cyst was 93%, 50% and 84%, standard of care -66.6%, 50% and 63.1% and for standard of care with NGS was 100%, 50% and 89.4% respectively. Cyst wall biopsy was able to diagnose 19/24 cysts (4 high risk, 7 intraductal papillary mucinous neoplasm, 4 cysts of mucinous origin and 4 benign). Conclusions: NGS data correlates well with histology and may aid in diagnosis and risk stratification of pancreatic cysts. Cyst wall biopsy performs well in diagnosing cysts but was inadequate in 5/24 patients

    Cytomegalovirus induces abnormal chondrogenesis and osteogenesis during embryonic mandibular development

    <p>Abstract</p> <p>Background</p> <p>Human clinical studies and mouse models clearly demonstrate that cytomegalovirus (CMV) disrupts normal organ and tissue development. Although CMV is one of the most common causes of major birth defects in humans, little is presently known about the mechanism(s) underlying CMV-induced congenital malformations. Our prior studies have demonstrated that CMV infection of first branchial arch derivatives (salivary glands and teeth) induced severely abnormal phenotypes and that CMV has a particular tropism for neural crest-derived mesenchyme (NCM). Since early embryos are barely susceptible to CMV infection, and the extant evidence suggests that the differentiation program needs to be well underway for embryonic tissues to be susceptible to viral infection and viral-induced pathology, the aim of this study was to determine if first branchial arch NCM cells are susceptible to mCMV infection prior to differentiation of NCM derivatives.</p> <p>Results</p> <p>E11 mouse mandibular processes (MANs) were infected with mouse CMV (mCMV) for up to 16 days <it>in vitr</it>o. mCMV infection of undifferentiated embryonic mouse MANs induced micrognathia consequent to decreased Meckel's cartilage chondrogenesis and mandibular osteogenesis. Specifically, mCMV infection resulted in aberrant stromal cellularity, a smaller, misshapen Meckel's cartilage, and mandibular bone and condylar dysmorphogenesis. Analysis of viral distribution indicates that mCMV primarily infects NCM cells and derivatives. Initial localization studies indicate that mCMV infection changed the cell-specific expression of FN, NF-κB2, RelA, RelB, and Shh and Smad7 proteins.</p> <p>Conclusion</p> <p>Our results indicate that mCMV dysregulation of key signaling pathways in primarily NCM cells and their derivatives severely disrupts mandibular morphogenesis and skeletogenesis. The pathogenesis appears to be centered around the canonical and noncanonical NF-κB pathways, and there is unusual juxtaposition of abnormal stromal cells and surrounding matrix. Moreover, since it is critically important that signaling molecules are expressed in appropriate cell populations during development, the aberrant localization of components of relevant signaling pathways may reveal the pathogenic mechanism underlying mandibular malformations.</p
