miRNA Changes in retinal ganglion cells after optic nerve crush and glaucomatous damage
The purpose of this study was to characterize the miRNA profile of purified retinal ganglion cells (RGC) from healthy and diseased rat retina. Diseased retina includes those after a traumatic optic nerve crush (ONC), and after ocular hypertension/glaucoma. Rats were separated into four groups: healthy/intact, 7 days after laser-induced ocular hypertension, 2 days after traumatic ONC, and 7 days after ONC. RGC were purified from rat retina using microbeads conjugated to CD90.1/Thy1. RNA were sequenced using Next Generation Sequencing. Over 100 miRNA were identified that were significantly different in diseased retina compared to healthy retina. Considerable differences were seen in the miRNA expression of RGC 7 days after ONC, whereas after 2 days, few changes were seen. The miRNA profiles of RGC 7 days after ONC and 7 days after ocular hypertension were similar, but discrete miRNA differences were still seen. Candidate mRNA showing different levels of expression after retinal injury were manipulated in RGC cultures using mimics/AntagomiRs. Of the five candidate miRNA identified and subsequently tested for therapeutic efficacy, miR-194 inhibitor and miR-664-2 inhibitor elicited significant RGC neuroprotection, whereas miR-181a mimic and miR-181d-5p mimic elicited significant RGC neuritogenesis
Ten Quick Tips for Harnessing the Power of ChatGPT/GPT-4 in Computational Biology
The rise of advanced chatbots, such as ChatGPT, has sparked curiosity in the
scientific community. ChatGPT is a general-purpose chatbot powered by large
language models (LLMs) GPT-3.5 and GPT-4, with the potential to impact numerous
fields, including computational biology. In this article, we offer ten tips
based on our experience with ChatGPT to assist computational biologists in
optimizing their workflows. We have collected relevant prompts and reviewed the
nascent literature in the field, compiling tips we project to remain pertinent
for future ChatGPT and LLM iterations, ranging from code refactoring to
scientific writing to prompt engineering. We hope our work will help
bioinformaticians to complement their workflows while staying aware of the
various implications of using this technology. Additionally, to track new and
creative applications for bioinformatics tools such as ChatGPT, we have
established a GitHub repository at

adherence to ChatGPT and other LLMs will increase the efficiency of
computational biologists, ultimately advancing the pace of scientific discovery

Influence of Different Thicknesses on Mechanical and Corrosion Properties of a-C:H Films
The hydrogenated amorphous carbon films (a-C:H) were deposited on p-type Si (100) substrates at different thicknesses by radio frequency plasma enhanced chemical vapor deposition technique (rf-PECVD). Raman spectra display asymmetric diamond-like peaks, representative of the a-C:H films. The decrease of intensity ID/IG ratios revealed the sp3 content arise at different thicknesses of the a-C:H films. In terms of mechanical properties, the high hardness and elastic modulus values show the elastic and plastic deformation behaviors related to sp3 content in amorphous carbon films. Electro chemical properties showed that the a-C:H films exhibited excellent corrosion resistance in air-saturated 3.5 wt% NaCl solution for pH 2 at room temperature. Thickness increasing affected the small sp2 clusters in matrix, restricting the velocity transfer and exchange of electrons. The deposited a-C:H films exhibited excellent mechanical properties and corrosion resistance
