    Ethnopharmacology – From Mexican Hallucinogens to a Global Transdisciplinary Science

    Psychoactive natural substances have been reported from practically all regions of the world, but Mexican indigenous cultures have played a crucial role having influenced medical, toxicological, biological, chemical, pharmaceutical, and, of course, anthropological research. Especially in the 1950’s and 1960’s peyotl, teonanacatl and other psychoactives came to the attention of researchers and revelers alike. In this overview we highlight the developments of ethnopharmacology from the initial development of the term until today using one psychoactive species as an example - Salvia divinorum. In 1962 “ethnopharmacologists”, Albert Hofmann and R. Gordon Wasson, documented and collected a flowering specimen of Ska María Pastora allowing the species botanical description as Salvia divinorum Epling & Játiva. Five years later Efron et al. (1967) organised a symposium “Ethnopharmacologic search for psychoactive drugs” which over the next decades would give its name to a discipline which today is much more broadly defined, dealing with local and traditional medicines, their biological activities and chemistry. Globalisation has resulted in a world-wide commodification of many traditional medicines and psychoactives, as exemplified by S. divinorum. This fascinating Lamiaceae has become globally recognized for its best known active constituent salvinorin A, a kappa-opioid antagonist which has a unique effect on human physiology. While today ethnopharmacology is a thriving discipline, the interest in psychoactive substances is no longer central to the discipline. The search for anti-cancer agents (which also started in earnest in the 1960’s) had been of particular relevance and today includes among its many foci: • The scientific study of local and traditional knowledge not only in remote regions, but for example, also in urban immigrant communities • Research linking ethnopharmacology to biodiversity research both in terms of a sustainable use of natural resources (ecosystems) • Pharmacological studies with the aim of understanding the effects of complex mixtures on specific diseases or disease targets • The safety of herbal medicines • Anthropological and historical approaches on the use of medicinal and food plants and the link between food and medical uses of plants and fungi. / 50 years on ethnopharmacology is very different from what D.     Endovascular Treatment of the Descending Thoracic Aorta

    AbstractObjectives: to report our initial experience with endovascular stent graft repair of a variety of thoracic aortic pathology.Design: retrospective single center study.Material and methods: between February 2000 and January 2002, endovascular stent graft repair was performed in 26 patients: traumatic aortic isthmus rupture (n=3), Type B dissection (n=11) and descending thoracic aortic aneurysm (n=12). The deployed stent graft systems were AneuRx-Medtronic (n=1), Talent-Medtronic (n=13) and Excluder-Gore (n=12).Results: successful deployment of the stent grafts in the intended position was achieved in all patients. No hospital mortality neither paraplegia were observed. Late, non procedure related, death occurred in four patients (15%). Access artery complications with rupture of the iliac artery occurred in two patients and were managed by iliac-femoral bypass. The left subclavian artery was overstented in seven patients (27%). Only the first patient received a carotido-subclavian bypass. The mean maximal aortic diameter decreased significantly in patients treated for descending thoracic aneurysm. Only one patient had an endoleak type II after 6 months without enlargement of the aneurysm. Complete thrombosis of the thoracic false lumen occurred in all but one patient treated for Type B dissection 6 months postoperatively. Two patients underwent a consecutive stent graft placement, due to a large re-entry tear distal to the first stent graft.Conclusions: endovascular stent graft repair for Type B dissection, descending thoracic aneurysm and aortic isthmus rupture is a promising less-invasive alternative to surgical repair. Further studies are mandatory to determine its long-term efficacy

    Derivatives for smooth representations of GL(n,R) and GL(n,C)

    The notion of derivatives for smooth representations of GL(n) in the p-adic case was defined by J. Bernstein and A. Zelevinsky. In the archimedean case, an analog of the highest derivative was defined for irreducible unitary representations by S. Sahi and called the "adduced" representation. In this paper we define derivatives of all order for smooth admissible Frechet representations (of moderate growth). The archimedean case is more problematic than the p-adic case; for example arbitrary derivatives need not be admissible. However, the highest derivative continues being admissible, and for irreducible unitarizable representations coincides with the space of smooth vectors of the adduced representation. In [AGS] we prove exactness of the highest derivative functor, and compute highest derivatives of all monomial representations.     Clinical experience with the Bicarbon heart valve prosthesis

    BACGROUND: We have previously reported mid-term results of a study, which ended in January 2000, on the Bicarbon valve. The study concluded that the valve showed excellent clinical results, associated with a low incidence of valve-related complications. In the present study, the same patients were prospectively followed for an additional 5 years. METHODS: Forty-four patients had aortic valve replacement (AVR), 48 had mitral valve replacement (MVR), and 13 had both aortic and mitral valve replacement (DVR). The mean age of the 105 patients was 61.2 ± 11.3 years. The mean follow-up was 6.1 ± 1.9 years with a cumulative follow-up of 616 patient-years. RESULTS: There were 5 early deaths (4.7%: 4 in the AVR group and 1 in the MVR group) and 21 late deaths (3.4%/patient-year: 5 valve related deaths and 16 valve unrelated deaths). Survival at 8 years was 75.2 ± 7.0% in the AVR group, 76.6 ± 6.2% in the MVR group, and 55.4 ± 16.1% in the DVR group. The linearized incidence of thrombo-embolic complications, hemorrhagic complications, and paravalvular leaks in all patients was 0.65 ± 1.48%, 0.81 ± 1.69%, and 0.16 ± 0.54%/patient-year respectively. No other complications were observed. CONCLUSION: The Bicarbon prosthetic heart valve has shown excellent long-term clinical results, associated with a low incidence of valve-related complications

    Chasing Migration Genes: A Brain Expressed Sequence Tag Resource for Summer and Migratory Monarch Butterflies (Danaus plexippus)

    North American monarch butterflies (Danaus plexippus) undergo a spectacular fall migration. In contrast to summer butterflies, migrants are juvenile hormone (JH) deficient, which leads to reproductive diapause and increased longevity. Migrants also utilize time-compensated sun compass orientation to help them navigate to their overwintering grounds. Here, we describe a brain expressed sequence tag (EST) resource to identify genes involved in migratory behaviors. A brain EST library was constructed from summer and migrating butterflies. Of 9,484 unique sequences, 6068 had positive hits with the non-redundant protein database; the EST database likely represents ∼52% of the gene-encoding potential of the monarch genome. The brain transcriptome was cataloged using Gene Ontology and compared to Drosophila. Monarch genes were well represented, including those implicated in behavior. Three genes involved in increased JH activity (allatotropin, juvenile hormone acid methyltransfersase, and takeout) were upregulated in summer butterflies, compared to migrants. The locomotion-relevant turtle gene was marginally upregulated in migrants, while the foraging and single-minded genes were not differentially regulated. Many of the genes important for the monarch circadian clock mechanism (involved in sun compass orientation) were in the EST resource, including the newly identified cryptochrome 2. The EST database also revealed a novel Na+/K+ ATPase allele predicted to be more resistant to the toxic effects of milkweed than that reported previously. Potential genetic markers were identified from 3,486 EST contigs and included 1599 double-hit single nucleotide polymorphisms (SNPs) and 98 microsatellite polymorphisms. These data provide a template of the brain transcriptome for the monarch butterfly. Our “snap-shot” analysis of the differential regulation of candidate genes between summer and migratory butterflies suggests that unbiased, comprehensive transcriptional profiling will inform the molecular basis of migration. The identified SNPs and microsatellite polymorphisms can be used as genetic markers to address questions of population and subspecies structure
