Expression of Functional Recombinant Mussel Adhesive Protein Mgfp-5 in Escherichia coli
Mussel adhesive proteins have been suggested as a basis for environmentally friendly adhesives for use in aqueous conditions and in medicine. However, attempts to produce functional and economical recombinant mussel adhesive proteins (mainly foot protein type 1) in several systems have failed. Here, the cDNA coding for Mytilus galloprovincialis foot protein type 5 (Mgfp-5) was isolated for the first time. Using this cDNA, we produced a recombinant Mgfp-5 fused with a hexahistidine affinity ligand, which was expressed in a soluble form in Escherichia coli and was highly purified using affinity chromatography. The adhesive properties of purified recombinant Mgfp-5 were compared with the commercial extracted mussel adhesive Cell-Tak by investigating adhesion force using atomic force microscopy, material surface coating, and quartz crystal microbalance. Even though further macroscale assays are needed, these microscale assays showed that recombinant Mgfp-5 has significant adhesive ability and may be useful as a bioadhesive in medical or underwater environments.X119196sciescopu
Agrobacterium-mediated genetic transformation of Miscanthus sinensis
Miscanthus species are tall perennial rhizomatous grasses with C4 photosynthesis originating from East Asia, and they are considered as important bioenergy crops for biomass production. In this study, Agrobacterium-mediated transformation system for M. sinensis was developed using embryogenic calli derived from mature seeds. In order to establish a stable system, optimum conditions to obtain highly regenerable and transformation-competent embryogenic calli were investigated, and embryogenic calli were efficiently induced with callus induction medium containing 3 mg L-1 2,4-dichlorophenoxyacetic acid and 25 mM l-proline, at pH 5.7 with an induction temperature of 28 A degrees C. In addition, the embryogenic callus induction and regeneration potentials were compared between seven M. sinensis germplasms collected from several sites in Korea, which revealed that the germplasm SNU-M-045 had superior embryogenic callus induction and regeneration potentials. With this germplasm, the genetic transformation of M. sinensis was performed using Agrobacterium tumefaciens EHA105 carrying pCAMBIA1300 with a green fluorescence protein gene as a reporter. After putative transgenic plants were obtained, the genomic integration of transgenes was confirmed by genomic PCR, transgene expression was validated by Northern blot analysis, and the number of transgene integration was confirmed by DNA gel blot analysis. Furthermore, the Agrobacterium-mediated transformation of M. sinensis was also performed with pCAMBIA3301 which contains an herbicide resistance gene (BAR), and we obtained transgenic M. sinensis plants whose herbicide resistance was confirmed by spraying with BASTA(A (R)). Therefore, we have established a stable Agrobacterium-mediated transformation system for M. sinensis, and also successfully produced herbicide-resistant Miscanthus plants by introducing BAR gene via the established method.X111210Ysciescopu
Separation of a benzene and nitric oxide mixture by a molecule prism
A study was performed on the separation of nitric oxide and benzene mixture by a molecule prism. The energy conservation and wave properties of molecules were used to obtain the molecule-optical index of refraction for a nonresonant infrared laser pulse. A chromatographic resolution of 0.62 for the spatial separation of a mixture was obtained using the focused Nd:YAG laser pulse as a molecule prism.open273
Origin of New Broad Raman D and G Peaks in Annealed Graphene
Since graphene, a single sheet of graphite, has all of its carbon atoms on the surface, its property is very sensitive to materials contacting the surface. Herein, we report novel Raman peaks observed in annealed graphene and elucidate their chemical origins by Raman spectroscopy and atomic force microscopy (AFM). Graphene annealed in oxygen-free atmosphere revealed very broad additional Raman peaks overlapping the D, G and 2D peaks of graphene itself. Based on the topographic confirmation by AFM, the new Raman peaks were attributed to amorphous carbon formed on the surface of graphene by carbonization of environmental hydrocarbons. While the carbonaceous layers were formed for a wide range of annealing temperature and time, they could be effectively removed by prolonged annealing in vacuum. This study underlines that spectral features of graphene and presumably other 2-dimensional materials are highly vulnerable to interference by foreign materials of molecular thickness.open116167Nsciescopu
VIABILITY, FATTY ACID COMPOSITION, AND STRUCTURE OF THE CORALLINE ALGA CORALLINA PILULIFERA
The decrease in the seaweed flora in some rocky areas, known as algal whitening or barren ground, is associated with some species of coralline algae. To determine the biological characteristics of a representative species of branched coralline alga, the number of medullary tiers was counted and ranged from 12 to 16. The 18S rDNA, psbA, and rbcL genes were used to confirm the identification of Corallina pilulifera. Measuring viability using triphenyl tetrazolium chloride showed highly viability from December to January. Cultural conditions of 16 C, 16 h light:8 h dark cycle, and 40 mu E m(-2) s(-1) light intensity were optimal for maintaining the viability of the coralline alga for up to three days. The fatty acids included 31.4% omega-3 eicosapentaenoic acid. Scanning electron microscopy of the surface structure revealed unique round wells about 7.9 +/- 1.3 mu m in diameter. The coralline alga, preventing fleshy seaweeds, may be used as a potential template for the creation of new environmentally friendly biomimetic antifouling material against the attachment of soft foulants, especially micro- and macroalgae.X111Ysciescopu
RGO/Nylon-6 composite mat with unique structural features and electrical properties obtained from electrospinning and hydrothermal process
In this work, the reduced graphene oxide (RGO) sheets were effectively uploaded through nylon-6 fibers using combined process of electrospinning and hydrothermal treatment. Good dispersion of graphene oxide (GO) with nylon-6 solution could allow to upload GO sheets through nylon-6 fibers and facilitate the formation of spider-wave-like nano-nets during electrospinning. GO sheets present on/into nylon-6 spider-wave-like nano-nets were further reduced to RGO using hydrothermal treatment. The impregnated GO sheets into nylon-6 nanofibers and their reduction during hydrothermal treatment were confirmed by FE-SEM, TEM, FT-IR and Raman spectra. The electrical characteristics of pristine nylon-6, GO/nylon-6 and RGO/nylon-6 nanofibers were investigated and it was found that RGO/nylon-6 composite mat had better electrical conductivity than others. The formation of spider-wave-like nano-nets as well as indirect route of incorporation of RGO sheets on electrospun nylon-6 mat may open a new direction for future graphene/polymer electronics. © 2013 The Korean Fiber Society and Springer Science+Business Media Dordrecht

Einstein's relativity. We reconstruct the exact de Sitter solution in
scalar-tensor gravity, in which the non-minimal coupling scalar is rolling
along the potential. This solution may have some relation to the early
inflation and present acceleration of the universe. We investigated its
primordial quantum perturbation around the adiabatic vacuum. We put forward for
the first time that exact de Sitter generates non-exactly scale invariant
perturbations. In the conformal coupling case, this model predicts that the

Molecular lens applied to benzene and carbon disulfide molecular beams
A molecular lens of the nonresonant dipole force formed by focusing a nanosecond IR laser pulse has been applied to benzene and CS2 molecular beams. Using the velocity map imaging technique for molecular ray tracing, characteristic molecular lens parameters including the focal length (f ), minimum beam width (W), and distance to the minimum beam width position (D) were determined. The laser intensity dependence of the observed lens parameters was in good agreement with theoretical predictions. W was independent of the laser peak intensity (I-0), whereas f and D varied linearly with 1/I-0. The differences in lens parameters between the molecular species were well correlated with the polarizability per mass values of the molecules. A high chromatographic resolution of Rs = 0.84 was achieved between the images of benzene molecular beams undeflected and deflected by the lens. Complexation and coacervation of like-charged polyelectrolytes inspired by mussels
It is well known that polyelectrolyte complexes and coacervates can form on mixing oppositely charged polyelectrolytes in aqueous solutions, due to mainly electrostatic attraction between the oppositely charged polymers. Here, we report the first (to the best of our knowledge) complexation and coacervation of two positively charged polyelectrolytes, which provides a new paradigm for engineering strong, self-healing interactions between polyelectrolytes underwater and a new marine mussel-inspired underwater adhesion mechanism. Unlike the conventional complex coacervate, the like-charged coacervate is aggregated by strong short-range cation-p interactions by overcoming repulsive electrostatic interactions. The resultant phase of the like-charged coacervate comprises a thin and fragile polyelectrolyte framework and round and regular pores, implying a strong electrostatic correlation among the polyelectrolyte frameworks. The like-charged coacervate possesses a very low interfacial tension, which enables this highly positively charged coacervate to be applied to capture, carry, or encapsulate anionic biomolecules and particles with a broad range of applications.113320Ysciescopu
