    Crystal structure of the heterotrimeric integrin-binding region of laminin-111

    Laminins are cell - adhesive glycoproteins that are essential for basement membrane assembly and function. Integrins are important laminin receptors, but their binding site on the heterotrimeric laminin s is poorly defined structurally. We report the crystal structure at 2.13 Å resolution of a minimal integrin - binding fragment of mouse laminin - 111, consisting of ~50 resid ues of α 1 β 1 γ 1 coiled coil and the first three lam inin G - like (LG) domains of the α 1 chain . The LG domains adopt a triangular arrangem ent, with the C - terminus of the coiled coil situated between LG1 and LG2. The critical integrin - binding glutamic acid residue in the γ 1 chain tail is surface - exposed and predicted to bind to the metal ion - dependent adhesion site in the integrin β 1 subunit. Additional contacts to the integrin are likely to be made by the LG1 and LG2 surfaces adjacent to the γ 1 chain tail, which are notably conserved and free of obstructing glycans

    In this research study Linear Programming (LP) Model is applied to determine the maximum profit from the three major crops (wheat, maize and alfalfa) in Central Yasin District Ghizer, where agriculture is the main source of income, however, due to population growth the agricultural land is used to construct buildings, which has minimized the land for agricultural use. In such a challenging condition a method to suggest the cultivation of a maximum profitable crop is the need of time. Therefore, LP model is an appropriate method to decide the profitable crop and to maximize the profit. The study was conducted employing quantitative method of research, with the purpose to compare the maximum profit per annum from the major crops, and to determine the beneficial crop among the three major crops in Central Yasin. All the farmers of the study area who grow the crops were the target population of the study and samples from the target population were selected using random sampling technique. Data was collected by conducting survey in the study area, where self-made questionnaires were distributed among the samples individually and responses were noted on the spot. The raw data is organized and analyzed using Microsoft Excel for LP model formulation. The result showed that the average land used for the cultivation of wheat, maize and alfalfa in the study area is 0.924 acres, 0.664 acres and 0.75 acres respectively. Furthermore, results of the LP model showed that alfalfa is the optimal solution rather than wheat and maize, and it (alfalfa) gives an average annual net profit of Rs.195634.488

    Variations in bilingual processing of positive and negative information

    Past research suggests that the emotional content of words has greater impact when presented in a bilingual's first language (L1) compared to their second language (L2). This is predicted to be a consequence of automatic processing of emotional words in L1 compared to slower, semantic processing in L2. In the current study 58 Chinese-English bilinguals from Hong Kong rated the valence and arousal of positive, neutral, and negative words presented in Chinese (L1) and English (L2). In contrast to predictions, perceived emotionality of the words was higher in L2, with positive words rated more positively and negative words rated more negatively when presented in English compared to Chinese. The findings suggest that words presented in L2 did not have lower emotional impact than L1, the results indicate that emotional processing of words may be influenced by language proficiency and language complexity

    MOLECULAR CHARACTERIZATION BY USING RANDOM AMPLIFIED POLYMORPHIC DNA (RAPD) ANALYSIS OF SALMONELLA ENTERITIDIS ISOLATES RECOVERED FROM AVIAN AND HUMAN SOURCES

    Random amplified polymorphic DNA (RAPD) analysis was applied for molecular characterization of five Salmonella enteritidis strains from different avian sources and human cases of infection. A total of 16 primers were used and only five primers showed good discriminatory power for all five isolates. Dendrogram showed a common lineage among all five isolates. There was a close genetic relationship among isolates of eggs and human sources, while there was less pronounced homology among isolates of broiler meat and human sources. On the basis of results we have found that an endemic strain of S. enteritidis is prevalent between the poultry derived food and humans which gives us an insight to genetic diversity of S. enteritidis from these sources

    Hyperinsulinism-hyperammonaemia syndrome: novel mutations in the GLUD1 gene and genotype-phenotype correlations

    Background: Activating mutations in the GLUD1 gene (which encodes for the intra-mitochondrial enzyme glutamate dehydrogenase, GDH) cause the hyperinsulinism–hyperammonaemia (HI/HA) syndrome. Patients present with HA and leucine-sensitive hypoglycaemia. GDH is regulated by another intra-mitochondrial enzyme sirtuin 4 (SIRT4). Sirt4 knockout mice demonstrate activation of GDH with increased amino acid-stimulated insulin secretion. Objectives: To study the genotype–phenotype correlations in patients with GLUD1 mutations. To report the phenotype and functional analysis of a novel mutation (P436L) in the GLUD1 gene associated with the absence of HA. Patients and methods: Twenty patients with HI from 16 families had mutational analysis of the GLUD1 gene in view of HA (n=19) or leucine sensitivity (n=1). Patients negative for a GLUD1 mutation had sequence analysis of the SIRT4 gene. Functional analysis of the novel P436L GLUD1 mutation was performed. Results: Heterozygous missense mutations were detected in 15 patients with HI/HA, 2 of which are novel (N410D and D451V). In addition, a patient with a normal serum ammonia concentration (21 µmol/l) was heterozygous for a novel missense mutation P436L. Functional analysis of this mutation confirms that it is associated with a loss of GTP inhibition. Seizure disorder was common (43%) in our cohort of patients with a GLUD1 mutation. No mutations in the SIRT4 gene were identified. Conclusion: Patients with HI due to mutations in the GLUD1 gene may have normal serum ammonia concentrations. Hence, GLUD1 mutational analysis may be indicated in patients with leucine sensitivity; even in the absence of HA. A high frequency of epilepsy (43%) was observed in our patients with GLUD1 mutations

    Multi-wavelength observing of a forming solar-like star

    V2129 Oph is a 1.35 solar mass classical T Tauri star, known to possess a strong and complex magnetic field. By extrapolating from an observationally derived magnetic surface map, obtained through Zeeman-Doppler imaging, models of V2129 Oph's corona have been constructed, and used to make predictions regarding the global X-ray emission measure, the amount of modulation of X-ray emission, and the density of accretion shocks. In late June 2009 we will under take an ambitious multi-wavelength, multi-observing site, and near contemporaneous campaign, combining spectroscopic optical, nIR, UV, X-ray, spectropolarimetric and photometric monitoring.     Prevalence of vitamin D deficiency in Pakistan and implications for the future

    Background and aims: Vitamin D deficiency (25-hydroxyvitamin D - VitDD) affects over one billion people worldwide. VitDD results in progression of osteoporosis as well as other conditions. Previous studies have shown high rates of VitDD in Pakistan despite appreciable levels of sunshine. However, none have assessed VitDD across all age groups, genders, incomes and locations to guide future strategies. Methods: Questionnaire and blood sampling among 4830 randomly selected citizens. Results: High levels of VitDD among all age groups, genders, income levels and locations. 53.5% had VitDD, 31.2% had insufficient Vitamin D and only 15.3% normal Vitamin D. Conclusion: High rates of VitDD in Pakistan despite high levels of sunshine and previous Food Acts asking for food fortification with Vitamin D. Public health strategies are needed to address high VitDD rates, including food fortification, i.e. nurture, alongside increasing exposure to sunlight, i.e. nature. This will involve all key stakeholder groups
