Loss-of-function mutations in SNX14 cause autosomal recessive spinocerebellar ataxia 20, which is a form of early-onset cerebellar ataxia that lacks molecular mechanisms and mouse models. We generated Snx14-deficient mouse models and observed severe motor deficits and cell-autonomous Purkinje cell degeneration. SNX14 deficiency disrupted microtubule organization and mitochondrial transport in axons by destabilizing the microtubule-severing enzyme spastin, which is implicated in dominant hereditary spastic paraplegia with cerebellar ataxia, and compromised axonal integrity and mitochondrial function. Axonal transport disruption and mitochondrial dysfunction further led to degeneration of high-energy-demanding Purkinje cells, which resulted in the pathogenesis of cerebellar ataxia. The antiepileptic drug valproate ameliorated motor deficits and cerebellar degeneration in Snx14-deficient mice via the restoration of mitochondrial transport and function in Purkinje cells. Our study revealed an unprecedented role for SNX14-dependent axonal transport in cerebellar ataxia, demonstrated the convergence of SNX14 and spastin in mitochondrial dysfunction, and suggests valproate as a potential therapeutic agent. We thank Tim Huang for helpful discussion, Wei Mo for sharing mouse lines, Li Zhong for sharing reagents, Aidong Han, Luming Yao, Caiming Wu, Mingxia Zhu, Qingfeng Liu, Lin Zhu, Shuo Zhang, Haiping Zheng, and Changchuan Xie for technical assistance, and Cui Li for providing bioinformatics software. CCA-YOLO: An Improved Glove Defect Detection Algorithm Based on YOLOv5

    No full text
Aiming to address the issue of low efficiency and a high false-detection rate in artificial defect detection in nitrile medical gloves, CCA-YOLO was proposed on the basis of YOLOv5 to realize the detection of tear and scratch defects. CCA-YOLO added a small-target detection layer to the YOLOv5 network backbone and further proposed an innovative channel coordinate attention mechanism. According to the different characteristics of tears and scratches, focal and efficient IoU loss and α-IoU loss functions were introduced to further improve the positioning accuracy. The data enhancement method was used to generate a dataset of nitrile gloves, which was divided into datasets for horizontal angular tear detection, vertical angular tear detection, and scratch detection. The problem of class imbalance with few defect samples was solved. Our experiments show that CCA-YOLO can effectively identify tear and scratch defects in nitrile medical gloves in the self-made datasets. Compared with YOLOv5, the mean average precision (mAP) of the three models for horizontal angular tear detection, vertical angular tear detection, and scratch detection can reach 99.3%, 99.8%, and 99.6%, showing increments of 4.2%, 5.3%, and 12.4%, respectively, thereby meeting the performance requirements of glove defect detection

Characteristics and Risk Assessment of Soil Polluted by Lead around Various Metal Mines in China

The contamination of soil by lead (Pb) is a serious and widespread problem in China, especially in mining areas. This paper summarized the available data regarding Pb-contaminated soils around various metal mines in China. Based on these data, the Pb concentration in the soil and its temporal and spatial changes were analyzed. Potential ecological hazards and adult lead models were also used to estimate ecological and health risks. The results indicated that the concentration of Pb was closely related with the type of mine. Compared with other types of mine, soil around lead-zinc (Pb-Zn) and tin (Sn) mines with high Pb contents in the metallic ores and high pollutant emission coefficient were more strongly polluted by Pb. The characteristic spatial and temporal variations of Pb pollution status in China were clarified, and the results showed that the concentration was high in the southern, southwestern, and central regions of China where many mining areas were located, and the mean value passed a turning point in 2012. Ecological risk assessments indicated that some areas around mines were at considerable to high risk, and the risk was relatively severe in Pb-Zn mining areas. According to the adult lead model, Pb-Zn mines had a greater impact on blood Pb concentration than the other types of mine

Cobot Motion Planning Algorithm for Ensuring Human Safety Based on Behavioral Dynamics

Recently, the safety of workers has gained increasing attention due to the applications of collaborative robots (cobot). However, there is no quantitative research on the impact of cobot behavior on humans’ psychological reactions, and these results are not applied to the cobot motion planning algorithms. Based on the concept of the gravity field, this paper proposes a model of the psychological safety field (PSF), designs a comprehensive experiment on different speeds and minimum distances when approaching the head, chest, and abdomen, and obtains the ordinary surface equation of psychological stress about speed and minimum distance by using data fitting. By combining social rules and PSF models, we improve the robot motion planning algorithm based on behavioral dynamics. The validation experiment results show that our proposed improved robot motion planning algorithm can effectively reduce psychological stress. Eighty-seven point one percent (87.1%) of the experimental participants think that robot motion planned by improved robot motion planning algorithms is more “friendly”, can effectively reduce psychological stress, and is more suitable for human–robot interaction scenarios

Abstract Background Eczema is the most common form of dermatitis and also the starting point of atopic march. Although many eczema‐associated allergic and immunologic disorders have been studied, there remains a gap in the systematic quantitative knowledge regarding the relationships between all childhood disorders and eczema. This study aimed to systematically explore eczema‐associated childhood diseases using a real‐world, long‐term clinical dataset generated from millions of children in China. Methods Data were collected at 8,907,735 outpatient healthcare visits from 2,592,147 children between January 1, 2013, and August 15, 2019, at the largest comprehensive pediatric medical center in Zhejiang Province of China. The period prevalence differences in various pediatric diseases between children with and without eczema were used to test the independence of various pediatric disorders and eczema using Fisher's exact test. Bonferroni correction was used to adjust the p value in multiple testing. Odds ratio >2 with 95% confidence interval not including 1 and adjusted p < 0.05 was used to identify eczema‐associated diseases. Results Overall, 234 pediatric disorders were identified from more than 6000 different pediatric disorders. An interactive eczema‐associated disease map that has related quantitative epidemiological features called ADmap was published at http://pedmap.nbscn.org/admap. Thirty‐six of these disease associations have not been reported in previous studies. Conclusion This systematic exploratory study confirmed the associations of many well‐known diseases with eczema in Chinese children and also identified some novel and interesting associations. These results are valuable for the development of a comprehensive approach to the management of eczema in childhood

At sub-3 nm nodes, the scaling of lateral devices represented by a fin field-effect transistor (FinFET) and gate-all-around field effect transistors (GAAFET) faces increasing technical challenges. At the same time, the development of vertical devices in the three-dimensional direction has excellent potential for scaling. However, existing vertical devices face two technical challenges: “self-alignment of gate and channel” and “precise gate length control”. A recrystallization-based vertical C-shaped-channel nanosheet field effect transistor (RC-VCNFET) was proposed, and related process modules were developed. The vertical nanosheet with an “exposed top” structure was successfully fabricated. Moreover, through physical characterization methods such as scanning electron microscopy (SEM), atomic force microscopy (AFM), conductive atomic force microscopy (C-AFM) and transmission electron microscopy (TEM), the influencing factors of the crystal structure of the vertical nanosheet were analyzed. This lays the foundation for fabricating high-performance and low-cost RC-VCNFETs devices in the future
