    Research Progress on Features and Characteristics of Rural Settlements: Literature Distribution, Key Issues, and Development Trends

    During the development of rural settlements, the loss of distinctive rural characteristics, caused by the contradiction between urban expansion and the ideal of pastoralism, has attracted widespread attention from researchers worldwide. To effectively understand the development and trends of the Research of Features and Characteristics of Rural Settlements (abbreviated as RFCRS), this paper uses the knowledge mapping software CiteSpace to conduct co-citation analysis, research collaboration analysis, keyword clustering, and keyword co-occurrence. The study analyzes the basic concepts, the literature distribution characteristics, research clusters, key issues, and development trends of RFCRS. The research found that the current key research topics in RFCRS include “Eco-logical services and environmental protection of rural settlements”, “Sustainable planning and architectural design issues of rural settlements”, and “Human settlement environment and service facility construction of rural settlements”. This paper predicts that future RFCRS research trends will focus on the study of landscape features and characteristics based on ecology, climate, and aesthetics; study of architectural features and characteristics based on characteristic factors and hierarchical structure; and research on rural revitalization based on sustainable development principles. The paper also offers four priority research suggestions for researchers from different disciplines

    Clinical characteristics and prognosis of 196 Chinese patients with colon cancer

    AimsTo analyze the clinical characteristics and prognostic factors of Chinese patients with colon cancer.MethodsA retrospective analysis of the records of patients with colon cancer underwent surgery between 2014 and 2017 was performed. Univariate analysis in combination with Cox proportional hazard regression model was used to analyze the survival data, so as to reveal the prognostic factors of colon cancer. Data record was based on a standard data form. SPSS version 26.0 was used for data analysis (SPSS, Chicago, IL, United States).ResultsThe 3-year survival rate and the 5-year survival rate was 79.3% and 68.2%, respectively. Univariate analysis showed that radical surgery, laparoscopic surgery, ascites, swollen lymph nodes at the root of the mesentery, liver metastases, nerve invasion, vascular invasion, tumor node metastasis (TNM) staging, positive level of carbohydrate antigen (CA) 19-9, CA125, CA72-4 and combined detection were positive factors in the prognosis of colon cancer (P < 0.05). Multivariate analysis showed that radical surgery and TNM staging were independent factors affecting the prognosis of patients with colon cancer (P < 0.05).ConclusionRadical surgery and TNM staging have a significant impact on the prognosis of patients with colon cancer

    Decreasing TfR1 expression reverses anemia and hepcidin suppression in β-thalassemic mice

    Iron availability for erythropoiesis and its dysregulation in β-thalassemia are incompletely understood. We previously demonstrated that exogenous apotransferrin leads to more effective erythropoiesis, decreasing erythroferrone (ERFE) and derepressing hepcidin in β-thalassemic mice. Transferrin-bound iron binding to transferrin receptor 1 (TfR1) is essential for cellular iron delivery during erythropoiesis. We hypothesize that apotransferrin's effect is mediated via decreased TfR1 expression and evaluate TfR1 expression in β-thalassemic mice in vivo and in vitro with and without added apotransferrin. Our findings demonstrate that β-thalassemic erythroid precursors overexpress TfR1, an effect that can be reversed by the administration of exogenous apotransferrin. In vitro experiments demonstrate that apotransferrin inhibits TfR1 expression independent of erythropoietin- and iron-related signaling, decreases TfR1 partitioning to reticulocytes during enucleation, and enhances enucleation of defective β-thalassemic erythroid precursors. These findings strongly suggest that overexpressed TfR1 may play a regulatory role contributing to iron overload and anemia in β-thalassemic mice. To evaluate further, we crossed TfR1+/- mice, themselves exhibiting iron-restricted erythropoiesis with increased hepcidin, with β-thalassemic mice. Resultant double-heterozygote mice demonstrate long-term improvement in ineffective erythropoiesis, hepcidin derepression, and increased erythroid enucleation in relation to β-thalassemic mice. Our data demonstrate for the first time that TfR1+/- haploinsufficiency reverses iron overload specifically in β-thalassemic erythroid precursors. Taken together, decreasing TfR1 expression during β-thalassemic erythropoiesis, either directly via induced haploinsufficiency or via exogenous apotransferrin, decreases ineffective erythropoiesis and provides an endogenous mechanism to upregulate hepcidin, leading to sustained iron-restricted erythropoiesis and preventing systemic iron overload in β-thalassemic mice

    Screening of an individualized treatment strategy for an advanced gallbladder cancer using patient-derived tumor xenograft and organoid models

    Gallbladder cancer is a highly aggressive malignancy with poor sensitivity to postoperative radiotherapy or chemotherapy; therefore, the development of individualized treatment strategies is paramount to improve patient outcomes. Both patient-derived tumor xenograft (PDX) and patient-derived tumor organoid (PDO) models derived from surgical specimens can better preserve the biological characteristics and heterogeneity of individual original tumors, display a unique advantage for individualized therapy and predicting clinical outcomes. In this study, PDX and PDO models of advanced gallbladder cancer were established, and the consistency of biological characteristics between them and primary patient samples was confirmed using pathological analysis and RNA-sequencing. Additionally, we tested the efficacy of chemotherapeutic drugs, targeted drugs, and immune checkpoint inhibitors using these two models. The results demonstrated that gemcitabine combined with cisplatin induced significant therapeutic effects. Furthermore, treatment with immune checkpoint inhibitors elicited promising responses in both the humanized mice and PDO immune models. Based on these results, gemcitabine combined with cisplatin was used for basic treatment, and immune checkpoint inhibitors were applied as a complementary intervention for gallbladder cancer. The patient responded well to treatment and exhibited a clearance of tumor foci. Our findings indicate that the combined use of PDO and PDX models can guide the clinical treatment course for gallbladder cancer patients to achieve individualized and effective treatment

    Optimum design and research on novel vehicle hybrid excitation synchronous generator

    Hybrid excitation is an organic combination of permanent magnet excitation and electric excitation. Hybrid excitation synchronous generator (HESG) both has the advantages of light quality, less losses and high efficiency like permanent magnet generator and the advantages of good magnetic field adjusting performance like electric excitation generator, so it is very suitable for the vehicle application. This paper presented a novel vehicle HESG which has skew stator core, permanent magnet rotor and both armature winding and field winding in the stator. Using ANSYS software, simulating the electric excitation field and the magnetic field, and finally the main parameters of HESG were designed. The simulation and the test results both show that the novel vehicle PMSG has the advantages of small cogging torque, high efficiency, small harmonic component output voltage and low waveform aberration, so as to meet the design requirements fully

    Patient-derived iPSCs link elevated mitochondrial respiratory complex I function to osteosarcoma in Rothmund-Thomson syndrome

    Rothmund-Thomson syndrome (RTS) is an autosomal recessive genetic disorder characterized by poikiloderma, small stature, skeletal anomalies, sparse brows/lashes, cataracts, and predisposition to cancer. Type 2 RTS patients with biallelic RECQL4 pathogenic variants have multiple skeletal anomalies and a significantly increased incidence of osteosarcoma. Here, we generated RTS patient-derived induced pluripotent stem cells (iPSCs) to dissect the pathological signaling leading to RTS patient-associated osteosarcoma. RTS iPSC-derived osteoblasts showed defective osteogenic differentiation and gain of in vitro tumorigenic ability. Transcriptome analysis of RTS osteoblasts validated decreased bone morphogenesis while revealing aberrantly upregulated mitochondrial respiratory complex I gene expression. RTS osteoblast metabolic assays demonstrated elevated mitochondrial respiratory complex I function, increased oxidative phosphorylation (OXPHOS), and increased ATP production. Inhibition of mitochondrial respiratory complex I activity by IACS-010759 selectively suppressed cellular respiration and cell proliferation of RTS osteoblasts. Furthermore, systems analysis of IACS-010759-induced changes in RTS osteoblasts revealed that chemical inhibition of mitochondrial respiratory complex I impaired cell proliferation, induced senescence, and decreased MAPK signaling and cell cycle associated genes, but increased H19 and ribosomal protein genes. In summary, our study suggests that mitochondrial respiratory complex I is a potential therapeutic target for RTS-associated osteosarcoma and provides future insights for clinical treatment strategies

    Observation of ηcωω\eta_c\to\omega\omega in J/ψγωωJ/\psi\to\gamma\omega\omega

    Using a sample of (1310.6±7.0)×106(1310.6\pm7.0)\times10^6 J/ψJ/\psi events recorded with the BESIII detector at the symmetric electron positron collider BEPCII, we report the observation of the decay of the (11S0)(1^1 S_0) charmonium state ηc\eta_c into a pair of ω\omega mesons in the process J/ψγωωJ/\psi\to\gamma\omega\omega. The branching fraction is measured for the first time to be B(ηcωω)=(2.88±0.10±0.46±0.68)×103\mathcal{B}(\eta_c\to\omega\omega)= (2.88\pm0.10\pm0.46\pm0.68)\times10^{-3}, where the first uncertainty is statistical, the second systematic and the third is from the uncertainty of B(J/ψγηc)\mathcal{B}(J/\psi\to\gamma\eta_c). The mass and width of the ηc\eta_c are determined as M=(2985.9±0.7±2.1)M=(2985.9\pm0.7\pm2.1)\,MeV/c2c^2 and Γ=(33.8±1.6±4.1)\Gamma=(33.8\pm1.6\pm4.1)\,MeV.Comment: 13 pages, 6 figure

    Evidence of a resonant structure in the e+eπ+D0De^+e^-\to \pi^+D^0D^{*-} cross section between 4.05 and 4.60 GeV

    The cross section of the process e+eπ+D0De^+e^-\to \pi^+D^0D^{*-} for center-of-mass energies from 4.05 to 4.60~GeV is measured precisely using data samples collected with the BESIII detector operating at the BEPCII storage ring. Two enhancements are clearly visible in the cross section around 4.23 and 4.40~GeV. Using several models to describe the dressed cross section yields stable parameters for the first enhancement, which has a mass of 4228.6 \pm 4.1 \pm 6.3 \un{MeV}/c^2 and a width of 77.0 \pm 6.8 \pm 6.3 \un{MeV}, where the first uncertainties are statistical and the second ones are systematic. Our resonant mass is consistent with previous observations of the Y(4220)Y(4220) state and the theoretical prediction of a DDˉ1(2420)D\bar{D}_1(2420) molecule. This result is the first observation of Y(4220)Y(4220) associated with an open-charm final state. Fits with three resonance functions with additional Y(4260)Y(4260), Y(4320)Y(4320), Y(4360)Y(4360), ψ(4415)\psi(4415), or a new resonance, do not show significant contributions from either of these resonances. The second enhancement is not from a single known resonance. It could contain contributions from ψ(4415)\psi(4415) and other resonances, and a detailed amplitude analysis is required to better understand this enhancement
