    FurinDB: A Database of 20-Residue Furin Cleavage Site Motifs, Substrates and Their Associated Drugs

    FurinDB (freely available online at http://www.nuolan.net/substrates.html) is a database of furin substrates. This database includes experimentally verified furin cleavage sites, substrates, species, experimental methods, original publications of experiments and associated drugs targeting furin substrates. The current database release contains 126 furin cleavage sites from three species: mammals, bacteria and viruses. A main feature of this database is that all furin cleavage sites are recorded as a 20-residue motif, including one core region (eight amino acids, P6–P2′) and two flanking solvent accessible regions (eight amino acids, P7–P14, and four amino acids, P3′–P6′), that represent our current understanding of the molecular biology of furin cleavage. This database is important for understanding the molecular evolution and relationships between sequence motifs, 3D structures, cellular functions and physical properties required by furin for cleavage, and for elucidating the molecular mechanisms and the progression of furin cleavage associated human diseases, including pathogenic infections, neurological disorders, tumorigenesis, tumor invasion, angiogenesis, and metastasis. FurinDB database will be a solid addition to the publicly available infrastructure for scientists in the field of molecular biology

    Metasurface-Based Free-Space Multi-port Beam Splitter with Arbitrary Power Ratio

    A beam splitter (BS) is one of the most critical building blocks in optical systems. Despite various attempts of flat-type BSs to miniaturize the conventional cube BS reported, it remains a challenge to realize an ultrathin optical BS with multi-port output, non-uniform splitting ratio and steerable outgoing directions. Herein, we have demonstrated a free-space optical multi-port beam splitter (MPBS) based on a polarization-independent all-dielectric metasurface. By applying an optimized phase-pattern paradigm via a gradient-descent-based iterative algorithm to amorphous silicon (a-Si) metasurfaces, we have prepared a variety of MPBS samples with arbitrarily predetermined output port number (2~7), power ratio and spatial distribution of output beams. The experimental results reveal that the fabricated MPBSs could achieve high total splitting efficiency (TSE, above 74.7%) and beam-splitting fidelity (similarity, above 78.4%) within the bandwidth of 100 nm (1500~1600 nm). We envision that such MPBS could provide fabulous flexibility for optical integrated system and diverse applications

    Nonhomogeneous seawater Sr isotopic composition in the coastal oceans : a novel tool for tracing water masses and submarine groundwater discharge

The definitive version was published in Geochemistry Geophysics Geosystems 12 (2011): Q05002, doi:10.1029/2010GC003372.Here we present high-precision (2σ = ±3 ppm) 87Sr/86Sr measurements in coastal waters, together with salinity, to evaluate water mass mixing and the influence of submarine groundwater discharge (SGD) in coastal waters and marginal seas. Nonhomogeneous Sr isotopic variations in water columns were documented in the Southern Okinawa Trough (SOT), South China Sea, and Kao-ping Canyon (KPC), where seawater 87Sr/86Sr varied up to 70 ppm. Seawater Sr isotopic composition changes only slightly in the upper 200 m of the SOT but was detectable and highly correlated with salinity, indicating a mixing between radiogenic North Pacific Tropical Water (high 87Sr/86Sr and high salinity) at 100–150 m and a less radiogenic component with low 87Sr/86Sr and low salinity at ∼200 m. Vertical profiles of seawater 87Sr/86Sr along the KPC show significant variations, suggesting dynamic mixing affected by continental inputs (i.e., river runoff and SGD) in this region. These results highlight the potential use of seawater Sr isotopes as a powerful tracer for determining mixing ratios and the dynamic mixing of oceanic water masses, especially in coastal and marginal seas.This research was supported by a postdoctoral fellowship from Top University (NCKU), Taiwan, to K.‐F.     CASE STUDY ON THE LARGE DIAMETER PIPE JACKING FOR UTILITY TUNNEL

    Pipe jacking have been widely used in utility tunnel constructions as an environment-friendly method in China. This study is focus on the critical technologies used in the pipe jacking for the utility tunnel in Huanggang Mingzhu road. The inner and outer diameter of this utility tunnel are 4m and 4.8m respectively, which is the largest circular pipe jacking project in China at present. This utility tunnel is designed under the urban main road with a heavy traffic, so the control accuracy of pipe jacking construction is required to be high. According to the characteristics of the project and actual construction technical measures, the key construction technologies including pipe jacking equipment, launching of small spacing, slurry circulating, the drag reduction technology, and the control of surface settlement are discussed in this paper. Meanwhile, the jacking force and ground settlement during pipe jacking construction are monitored. The results show that the selected pipe jacking machine has good adaptability to the geological conditions of the project. The actual jacking force is much smaller than the theoretical value, and the two intermediate jacking stations are not activated. In addition, the road surface deformation is -8–5mm during the whole process of pipe jacking construction, which has no impact on surface traffic

    Double Dome and Reemergence of Superconductivity in Pristine 6R-TaS2 under Pressure

    Investigating the implications of interlayer coupling on superconductivity is essential for comprehending the intrinsic mechanisms of high temperature superconductors. Van der Waals heterojunctions have attracted extensive research due to their exotic interlayer coupling. Here, we present a natural heterojunction superconductor of 6R-TaS2 that demonstrates a double-dome of superconductivity, in addition to, the reemergence of superconducting under high pressures. Our first principles calculation shows that the first dome of superconductivity in 6R-TaS2 can be attributed to changes in interlayer coupling and charge transfer. The second superconducting dome and the reemergence of superconductivity can be ascribed to changes in the density of states resulting from Fermi surface reconstruction, in which the DOS of T-layer and S p-orbitals play a crucial role. We have reported the first observation in TMDs that non-metallic atoms playing a dominant role in the reemergence of superconducting and the influence of two Lifshitz transitions on superconducting properties

    InstructSeq: Unifying Vision Tasks with Instruction-conditioned Multi-modal Sequence Generation

    Empowering models to dynamically accomplish tasks specified through natural language instructions represents a promising path toward more capable and general artificial intelligence. In this work, we introduce InstructSeq, an instruction-conditioned multi-modal modeling framework that unifies diverse vision tasks through flexible natural language control and handling of both visual and textual data. InstructSeq employs a multimodal transformer architecture encompassing visual, language, and sequential modeling. We utilize a visual encoder to extract image features and a text encoder to encode instructions. An autoregressive transformer fuses the representations and generates sequential task outputs. By training with LLM-generated natural language instructions, InstructSeq acquires a strong comprehension of free-form instructions for specifying visual tasks. This provides an intuitive interface for directing capabilities using flexible natural instructions. Without any task-specific tuning, InstructSeq achieves compelling performance on semantic segmentation, referring expression segmentation/comprehension, and image captioning. The flexible control and multi-task unification empower the model with more human-like versatility and generalizability for computer vision.     A user-centred collective system design approach for Smart Product-Service Systems:A case study on fitness product design

    Emerging technologies have significantly contributed to the evolution of traditional product-service systems (PSS) into smart PSS. This transformation demands a fresh perspective and a more inventive design approach. In response, this study proposes a new User-Centred Collective System Design (CSD) framework and process for Smart PSS design, aiming to enhance stakeholder engagement during the entire design process, thus promoting highly effective and creative design solutions. A case study, titled ‘Next-G Smart Fitness PSS Design’, was carried out to test and implement this approach, contrasting the results of the CSD method with a designer-centred method. The outcomes showed a marked improvement in product novelty and user desirability of the design outcomes when using the proposed design framework. The proposed CSD framework could offer beneficial insights and user-centric viewpoints for practitioners dealing with complex challenges linked to smart PSS design
