    Development of Rifampicin-Indocyanine Green-Loaded Perfluorocarbon Nanodroplets for Photo-Chemo-Probiotic Antimicrobial Therapy

    Acne vulgaris, generally resulted from overgrowth of Propionibacterium acnes (P. acnes), is one of the most difficult-to-treat facial dermatoses and more than 90% of adolescents experienced the disease worldwide. Because the innate non-lymphoid immune system cannot effectively eliminate excessive P. acnes from the skin surface, so far the therapy of acne vulgaris is still mainly dependent on antibiotic treatment. However, long-term or overdose of antibiotics may initiate microbial drug resistance and/or generate unexpected side effects that seriously hamper the use of antibiotics in the clinic. To overcome the aforementioned challenges, the novel rifampicin (RIF)-indocyanine green (ICG)-loaded perfluorocarbon (PFC) nanodroplets (RIPNDs) that may offer combined photo-, chemo-, and probiotic efficacies to P. acnes eradication were developed in this study. The RIPND was first characterized as a sphere-like nanoparticle with surface charge of āˆ’20.9 Ā± 2.40 mV and size of 240.7 Ā± 6.73 nm, in which the encapsulation efficiencies of RIF and ICG were 54.0 Ā± 10.5% and 95.0 Ā± 4.84%, respectively. In comparison to the freely dissolved ICG, the RIPNDs conferred an enhanced thermal stability to the entrapped ICG, and were able to provide a comparable hyperthermia effect and markedly increased production of singlet oxygen under near infrared (NIR; 808 nm, 6 W/cm2) exposure. Furthermore, the RIPNDs were able to induce fermentation of S. epidermidis but not P. acnes, indicating that the RIPNDs may serve as a selective fermentation initiator for the target probiotics. Based on the microbial population index analyses, P. acnes with 1 Ɨ 106 cells/mL can be completely eradicated by 12-h co-culture with S. epidermidis fermentation products followed by treatment of RIPNDs (ā‰„20-Ī¼M ICG/3.8-Ī¼M RIF) + NIR for 5 min, whereby the resulted microbial mortality was even higher than that caused by using 16-fold enhanced amount of loaded RIF alone. Overall these efforts show that the RIPNDs were able to provide improved ICG stability, selective fermentability to S. epidermidis, and enhanced antimicrobial efficacy compared to equal dosage of free RIF and/or ICG, indicating that the developed nanodroplets are highly potential for use in the clinical anti-P. acne treatment with reduced chemotoxicity

    Atypical Presentation of Spinal Epidural Abscessā€”Prolonged and Intractable Abdominal Pain

    SummaryDespite advances in medicine, early diagnosis of spinal epidural abscess remains a challenge to clinicians. The most common symptoms of spinal epidural abscess include back pain, fever, and neuralgic deficits. However, spinal epidural abscess can also present with vague and nonspecific symptoms. In this case, a 68-year-old male had abdominal pain in the right upper quadrant lasting 3 weeks and was diagnosed with a gastric ulcer. After treatment, his symptoms did not resolve. Fever and back pain became evident as his disease progressed, followed by right lower limb weakness and the inability to walk. He was taken to the emergency department of our hospital, and the weakness of his lower extremities worsened during hospitalization. His right leg became completely paralyzed despite the use of intravenous antibiotics. A spinal computed tomography scan was performed emergently (magnetic resonance imaging was unavailable) and revealed an epidural abscess involving T5ā€“6 with adjacent osteomyelitis. The patient underwent posterior decompressive laminectomy with pus drainage in the T4ā€“7 region. His neuralgic examinations improved soon after the operation, but ambulation remained limited. Early diagnosis is crucial to the prognosis of spinal epidural abscess, because delayed diagnosis usually results in complete paralysis even death. Thus, clinicians should be aware of atypical presentations of spinal epidural abscess

    Microstructure and Corrosion Behavior of Ni-Alloy/CrN Nanolayered Coatings

    The Ni-alloy/CrN nanolayered coatings, Ni-Al/CrN and Ni-P/CrN, were deposited on (100) silicon wafer and AISI 420 stainless steel substrates by dual-gun sputtering technique. The influences of the layer microstructure on corrosion behavior of the nanolayered thin films were investigated. The bilayer thickness was controlled approximately 10 nm with a total coating thickness of 1m. The single-layer Ni-alloy and CrN coatings deposited at 350āˆ˜C were also evaluated for comparison. Through phase identification, phases of Ni-P and Ni-Al compounds were observed in the single Ni-alloy layers. On the other hand, the nanolayered Ni-P/CrN and Ni-Al/CrN coatings showed an amorphous/nanocrystalline microstructure. The precipitation of Ni-Al and Ni-P intermetallic compounds was suppressed by the nanolayered configuration of Ni-alloy/CrN coatings. Through Tafel analysis, the corr and corr values ranged from ā€“0.64 to ā€“0.33ā€‰V and 1.42Ɨ10āˆ’5 to 1.14Ɨ10āˆ’6ā€‰A/cm2, respectively, were deduced for various coating assemblies. The corrosion mechanisms and related behaviors of the coatings were compared. The coatings with a nanolayered Ni-alloy/CrN configuration exhibited a superior corrosion resistance to single-layer alloy or nitride coatings

    Many countries are facing significant challenges posed by aging populations, and many of these elderly people are increasingly socially isolated, raising the need to develop senior-friendly services and settings for community life. ICT-enabled technologies can be used to help the elderly maintain adequate degrees of social participation. This paper reports the design and production of an innovative recreation installation featuring elderly friendly technologies to enhance the social lives of elderly Chinese people. The contents are composed of modernized visualization elements of traditional Chinese painting designed to engage elderly Chinese people and promote interaction and participation.</p

    Phosphorylation at Ser473 regulates heterochromatin protein 1 binding and corepressor function of TIF1beta/KAP1

    <p>Abstract</p> <p>Background</p> <p>As an epigenetic regulator, the transcriptional intermediary factor 1Ī² (TIF1Ī²)/KAP1/TRIM28) has been linked to gene expression and chromatin remodeling at specific loci by association with members of the heterochromatin protein 1 (HP1) family and various other chromatin factors. The interaction between TIF1Ī² and HP1 is crucial for heterochromatin formation and maintenance. The HP1-box, PXVXL, of TIF1Ī² is responsible for its interaction with HP1. However, the underlying mechanism of how the interaction is regulated remains poorly understood.</p> <p>Results</p> <p>This work demonstrates that TIF1Ī² is phosphorylated on Ser473, the alteration of which is dynamically associated with cell cycle progression and functionally linked to transcriptional regulation. Phosphorylation of TIF1Ī²/Ser473 coincides with the induction of cell cycle gene <it>cyclin A2 </it>at the S-phase. Interestingly, chromatin immunoprecipitation demonstrated that the promoter of <it>cyclin A2 </it>gene is occupied by TIF1Ī² and that such occupancy is inversely correlated with Ser473 phosphorylation. Additionally, when HP1Ī² was co-expressed with TIF1Ī²/S473A, but not TIF1Ī²/S473E, the colocalization of TIF1Ī²/S473A and HP1Ī² to the promoters of <it>Cdc2 </it>and <it>Cdc25A </it>was enhanced. Non-phosphorylated TIF1Ī²/Ser473 allowed greater TIF1Ī² association with the regulatory regions and the consequent repression of these genes. Consistent with possible inhibition of TIF1Ī²'s corepressor function, the phosphorylation of the Ser473 residue, which is located near the HP1-interacting PXVXL motif, compromised the formation of TIF1Ī²-HP1 complex. Finally, we found that the phosphorylation of TIF1Ī²/Ser473 is mediated by the PKCĪ“ pathway and is closely linked to cell proliferation.</p> <p>Conclusion</p> <p>The modulation of HP1Ī²-TIF1Ī² interaction through the phosphorylation/de-phosphorylation of TIF1Ī²/Ser473 may constitute a molecular switch that regulates the expression of particular genes. Higher levels of phosphorylated TIF1Ī²/Ser473 may be associated with the expression of key regulatory genes for cell cycle progression and the proliferation of cells.</p
