    Effects of acute ammonia exposure and post-exposure recovery on nonspecific immunity in Clam Cyclina sinensis

    This study aimed to assess the toxicity of ammonia on clam Cyclina sinensis and the post-exposure recovery. With increased exposure to TAN, the alkaline phosphatase (AKP) activities after exposure showed a trend of growing initially and subsequently decreasing, whereas the AKP activities after post-exposure recovery showed an increasing trend. The AKP activities after post-exposure recovery were significantly higher than those in control. The acid phosphatase (ACP) activities in T1 and T2 after post-exposure recovery were higher than those in the control, whereas the ACP activities in T3, T4, and T5 after post-exposure recovery were significantly higher than those in the control. The lysozyme (LZM) activities in T1 and T2 after exposure were significantly higher than those in control, whereas the LZM activities in T3, T4, and T5 after exposure were significantly lower than those in the control. Overall, ACP and LZM in the clams exposed to a low level of TAN (≤ 40 mg/L) can recover to the normal levels completely. However, a 48h recovery period scarcely seems adequate to compensate for AKP, ACP, and LZM activities in the clams exposed to a high level of TAN (> 40 mg/L)

    Recombination analysis reveals a double recombination event in hepatitis E virus

    Recombination of Hepatitis E Virus (HEV) has rarely been reported. In the present study, phylogenetic and recombination analyses were performed on 134 complete HEV genomes. Three potentially significant recombination events, including both intra-genotype and one inter-genotype, were identified by recombination detection analysis. Recombination events I and II occurred intra-genotype and inter-genotype, respectively, among three isolates, including the lineage represented by CHN-XJ-SW13 (GU119961, swine isolate), E067-SIJ05C (AB369690, human isolate), and JJT-Kan (AB091394, human isolate), and lead to the recombinant swine isolate swCH31 (DQ450072). Recombination event III occurred between the lineage represented by the NA1 (M73218) and K52-87 (L25595), which resulted in the recombinant Xingjiang-1 (D11092). Our analyses proved that that recombination could occur between human and swine HEV strains, double recombination events existed in HEV, and recombination event could happen within ORF2 region of HEV. These results will provide valuable hints for future research on HEV diversity

    Recombination analysis based on the complete genome of bocavirus

    Bocavirus include bovine parvovirus, minute virus of canine, porcine bocavirus, gorilla bocavirus, and Human bocaviruses 1-4 (HBoVs). Although recent reports showed that recombination happened in bocavirus, no systematical study investigated the recombination of bocavirus. The present study performed the phylogenetic and recombination analysis of bocavirus over the complete genomes available in GenBank. Results confirmed that recombination existed among bocavirus, including the likely inter-genotype recombination between HBoV1 and HBoV4, and intra-genotype recombination among HBoV2 variants.     Antipsychotics-induced improvement of cool executive function in individuals living with schizophrenia

    Cool executive dysfunction is a crucial feature in people living with schizophrenia which is related to cognition impairment and the severity of the clinical symptoms. Based on electroencephalogram (EEG), our current study explored the change of brain network under the cool executive tasks in individuals living with schizophrenia before and after atypical antipsychotic treatment (before_TR vs. after_TR). 21 patients with schizophrenia and 24 healthy controls completed the cool executive tasks, involving the Tower of Hanoi Task (THT) and Trail-Marking Test A-B (TMT A-B). The results of this study uncovered that the reaction time of the after_TR group was much shorter than that of the before_TR group in the TMT-A and TMT-B. And the after_TR group showed fewer error numbers in the TMT-B than those of the before_TR group. Concerning the functional network, stronger DMN-like linkages were found in the before_TR group compared to the control group. Finally, we adopted a multiple linear regression model based on the change network properties to predict the patient’s PANSS change ratio. Together, the findings deepened our understanding of cool executive function in individuals living with schizophrenia and might provide physiological information to reliably predict the clinical efficacy of schizophrenia after atypical antipsychotic treatment

    microRNA-29a-3p, Up-Regulated in Human Gastric Cells and Tissues with H.Pylori Infection, Promotes the Migration of GES-1 Cells via A20-Mediated EMT Pathway

    Background/Aims: Helicobacter pylori (H. pylori) infection is closely related to human gastric mucosa-associated diseases. Several recent studies on miRNAs have expanded our insights on H.pylori pathogenesis. This study aimed to investigate the biological roles and underlying molecular mechanisms of miR-29a-3p in human gastric cells and tissues with H.pylori infection. Methods: miR-29a-3p expression was quantified by quantitative RT-PCR (qRT-PCR). A miR-29a-3p target gene was validated by bioinformatics analysis, western blotting and dual luciferase reporter gene assays. Western blotting and immunohistochemistry (IHC) assay were performed to detect the protein expression. Transwell assay was used to determine the cell migration ability. Results: MiR-29a-3p was up-regulated in H.pylori-positive gastric mucosa tissues and H.pylori-infected gastric cells. The up-regulation of miR-29a-3p was dose-dependent in BGC-823 and GES-1 cells infected with H.pylori. Using gain- and loss-of-function experiments in vitro, we demonstrated that miR-29a-3p promoted the migration of gastric epithelial cells. We further characterized A20 as a direct target of miR-29a-3p. The expression of A20 was decreased in H.pylori-positive gastric mucosa tissues compared with H.pylori-negative gastric mucosa tissues. A20 downregulation was time- and dose-dependent in GES-1 and BGC-823 cells infected with H.pylori. In GES-1 and BGC-823 cells infected with H.pylori, the miR-29a-3p mimic significantly blocked A20 expression, which suggests that H.pylori decreased A20 expression through up-regulating miR-29a-3p in GES-1 and BGC-823 cells infected with H.pylori. The knockdown of A20 by siRNA enhanced the migration of human gastric epithelial cells and promoted the expression of Snail, Vimentin, and N-cadherin and inhibited the expression of E-cadherin. Conclusion: The miR-29a-3p may act as a tumor promotive miRNA by regulating cells migration through directly targeting of A20 gene in human gastric epithelial cells infected with H.pylori

    Effect of salinity on growth performance and resistance of the clam Cyclina sinensis against Vibrio parahaemolyticus infection

    We examined the growth performance, immune parameters and the susceptibility to Vibrio parahaemolyticus in the clam Cyclina sinensis, which had been reared at different salinity levels of 10‰, 20‰ and 30‰ for 60 days. At the end of the feeding experiment, the biggest shell length and body weight was found in 20‰, followed by 30‰ and 10‰. No significant differences in superoxide dismutase (SOD) activity were observed among the clams held in 10‰, 20‰ and 30‰. Na+/K+ -ATPase (NKA) activity of the clams held in 20‰ and 30‰ were significantly lower than that in group 10‰. The lowest activities of lysozyme (LZM) and glutamic-pyruvic transaminase (GPT) were found in group 10‰. The Integrated Biomarker Response index (IBR) values of the clams had an inverse relationship with salinity: 11.28, 3.40 and 2.85 in 10‰, 20‰ and 30‰, respectively. At the end of the feeding experiment, the clams were infected with V. parahaemolyticus. As time after infection goes on, the survival rate of clams reared in 20‰ was not significantly different with the other two treatments from 24 to 48h after infection. However, from 72 to 120h after infection, it was significantly lower than those reared in 10‰, while it was significantly higher than those reared in 30‰. It is concluded that the clam C. sinensis reared in 10‰ seawater may reduce growth performance and immune ability, whereas increase resistance against V. parahaemolyticus infection
