    Antibacterial characterization of Bacillus velezensis LG37 and mining of genes related to biosynthesis of antibacterial substances

    Bacillus velezensis LG37 secretes various antibacterial substances and inhibits the growth of other bacteria. Here, we analyzed the antibacterial characteristics and the screening and verification of genes related to the synthesis of the antibacterial substance of LG37 by antibacterial activities experiment, Local BLAST+, and RT-PCR. LG37 was isolated from aquaculture water and preserved in our laboratory. The phylogenetic tree was used to analyze the genetic relationship between LG37 and the bacteriostatic test indicator strain. LG37 had a more substantial inhibitory effect on closely related strains, while the inhibitory effect on the more distantly related strains was weak. Combined with the results of genome sequencing, the ribosomal peptide (RP) bacteriocin gene and non-ribosomal peptide synthetase (NRPSs) related gene clusters were screened and analyzed. A total of six gene-coding RP bacteriocins and two genes coding surfactins and fengycin A NRPSs gene cluster were screened. Local BLAST+ analysis revealed a total of 11 NRPSs gene clusters. The active expression of the NRPSs and RP encoding genes was further validated by RT-PCR. The findings revealed various genes and gene clusters encoding RP bacteriocins and NRPSs in B. velezensis LG37. The bacterium is potentially valuable in diverse applications in aquaculture

    Portable Light Pen 3D Vision Coordinate Measuring System- Probe Tip Center Calibration

    For different tasks, probe tip should be changed in the 3D vision coordinate measuring system and the accurate determination of probe tip center position is critical. A novel and simple approach for calibrating the probe tip center position of the light pen is presented in this paper. Hundreds of images of the light pen with different postures are collected while the probe tip is kept in firm contact with a reference conical hole. The probe tip position is determined by computing the rotation matrix and translation vector from the obtained images by using the least square fitting method. The experimental results demonstrate the effectiveness of the proposed approach. Its repeatability reaches 0.033 mm, 0.030 mm, and 0.043 mm in x, y, and z axes, respectively, and its convergence speed is satisfactory

    Neonatal rhesus monkey is a potential animal model for studying pathogenesis of EV71 infection

    AbstractData from limited autopsies of human patients demonstrate that pathological changes in EV71-infected fatal cases are principally characterized by clear inflammatory lesions in different parts of the CNS; nearly identical changes were found in murine, cynomolgus and rhesus monkey studies which provide evidence of using animal models to investigate the mechanisms of EV71 pathogenesis. Our work uses neonatal rhesus monkeys to investigate a possible model of EV71 pathogenesis and concludes that this model could be applied to provide objective indicators which include clinical manifestations, virus dynamic distribution and pathological changes for observation and evaluation in interpreting the complete process of EV71 infection. This induced systemic infection and other collected indicators in neonatal monkeys could be repeated; the transmission appears to involve infecting new monkeys by contact with feces of infected animals. All data presented suggest that the neonatal rhesus monkey model could shed light on EV71 infection process and pathogenesis

    Genomic Insights Into the Admixture History of Mongolic- and Tungusic-Speaking Populations From Southwestern East Asia

    As a major part of the modern Trans-Eurasian or Altaic language family, most of the Mongolic and Tungusic languages were mainly spoken in northern China, Mongolia, and southern Siberia, but some were also found in southern China. Previous genetic surveys only focused on the dissection of genetic structure of northern Altaic-speaking populations; however, the ancestral origin and genomic diversification of Mongolic and Tungusic–speaking populations from southwestern East Asia remain poorly understood because of the paucity of high-density sampling and genome-wide data. Here, we generated genome-wide data at nearly 700,000 single-nucleotide polymorphisms (SNPs) in 26 Mongolians and 55 Manchus collected from Guizhou province in southwestern China. We applied principal component analysis (PCA), ADMIXTURE, f statistics, qpWave/qpAdm analysis, qpGraph, TreeMix, Fst, and ALDER to infer the fine-scale population genetic structure and admixture history. We found significant genetic differentiation between northern and southern Mongolic and Tungusic speakers, as one specific genetic cline of Manchu and Mongolian was identified in Guizhou province. Further results from ADMIXTURE and f statistics showed that the studied Guizhou Mongolians and Manchus had a strong genetic affinity with southern East Asians, especially for inland southern East Asians. The qpAdm-based estimates of ancestry admixture proportion demonstrated that Guizhou Mongolians and Manchus people could be modeled as the admixtures of one northern ancestry related to northern Tungusic/Mongolic speakers or Yellow River farmers and one southern ancestry associated with Austronesian, Tai-Kadai, and Austroasiatic speakers. The qpGraph-based phylogeny and neighbor-joining tree further confirmed that Guizhou Manchus and Mongolians derived approximately half of the ancestry from their northern ancestors and the other half from southern Indigenous East Asians. The estimated admixture time ranged from 600 to 1,000 years ago, which further confirmed the admixture events were mediated via the Mongolians Empire expansion during the formation of the Yuan dynasty

    In deregulated power market, hydro producer has in principle no other objective than to produce electricity and sell with maximum profit with lower market risk. Attention must focus on profit uncertainty caused by uncertainty in spot prices and reservoir inflow. The purpose of this review is to assess the state-of-the-art in hydropower operations considering profit risk under uncertainty and consider future directions for additional research and application. Physical and financial tools to hedge risk in bilateral market and risk-assessment methods are all discussed in detail. Furthermore, production resources can also be used to manage risk to a certain extent.     Numerical Simulation Research on Radial Force of Centrifugal Pump with Guide Vanes

    To reveal the internal unsteady flow state of the guide vane centrifugal pump, in this paper, the standard SST k‐ω turbulent flow model is used for unsteady numerical simulation of the centrifugal pump. The characteristics of the flow field inside the centrifugal pump are analyzed, the resultant force and vector distribution of the radial force of the guide vane and impeller of the centrifugal pump under different flow rates are obtained, which were verified by experiments. The results show that the main reason of radial force of the impeller is the pressure asymmetry in each flow passage. The radial force will show periodic fluctuations due to the rotor-stator interference between the impeller and the guide vanes under different flow rates. The radial force on the impeller decreases gradually with the increase of the flow, the distribution is hexagonal or hexagonal shape, and the number of impeller blades is the same. The results can provide reference for the design of impeller and guide vane of centrifugal pump

    Abstract Cardiocerebrovascular diseases (CVDs) are the leading cause of death worldwide, consuming huge healthcare budget. For CVD patients, the prompt assessment and appropriate administration is the crux to save life and improve prognosis. Thrombolytic therapy, as a non-invasive approach to achieve recanalization, is the basic component of CVD treatment. Still, there are risks that limits its application. The objective of this review is to give an introduction on the utilization of thrombolytic therapy in cardiocerebrovascular blockage diseases, including coronary heart disease and ischemic stroke, and to review the development in risk assessment of thrombolytic therapy, comparing the performance of traditional scales and novel artificial intelligence-based risk assessment models
