    Isolation and functional characterization of proinflammatory acidic phospholipase A2 from Bothrops leucurus snake venom

    AbstractIn the present study, an acidic PLA2, designated Bl-PLA2, was isolated from Bothrops leucurus snake venom through two chromatographic steps: ion-exchange on CM-Sepharose and hydrophobic chromatography on Phenyl-Sepharose. Bl-PLA2 was homogeneous on SDS-PAGE and when submitted to 2D electrophoresis the molecular mass was 15,000Da and pI was 5.4. Its N-terminal sequence revealed a high homology with other Asp49 acidic PLA2s from snake venoms. Its specific activity was 159.9U/mg and the indirect hemolytic activity was also higher than that of the crude venom. Bl-PLA2 induced low myotoxic and edema activities as compared to those of the crude venom. Moreover, the enzyme was able to induce increments in IL-12p40, TNF-α, IL-1β and IL-6 levels and no variation of IL-8 and IL-10 in human PBMC stimulated in vitro, suggesting that Bl-PLA2 induces proinflammatory cytokine production by human mononuclear cells. Bothrops leucurus venom is still not extensively explored and knowledge of its components will contribute for a better understanding of its action mechanism

    O papel de um instrumento de apoio à comunicação matemática numa turma do 4.º ano

    Relatório de Estágio apresentado à Escola Superior de Educação de Lisboa para obtenção de grau de mestre em Ensino do 1.º e 2.º Ciclos do Ensino BásicoA criação de ambientes de sala de aula onde os alunos são agentes efetivos da sua aprendizagem e têm espaço para participar e expor os seus conhecimentos, sabendo que vão ser ouvidos pelo professor e colegas, é, cada vez mais, uma recomendação curricular da atualidade e objeto de estudo. Este relatório dá conta da intervenção realizada numa turma do 4.º ano de escolaridade, com o propósito de promover a aprendizagem, valorizando, sistematicamente, a comunicação. Em simultâneo, explana-se a investigação realizada nesta turma, que envolveu a avaliação do impacto da construção de um instrumento de apoio aos momentos de Comunicação Matemática, no contexto de uma rotina semanal da turma, a Apresentação do Problema da Semana. Com a pretensão de dar resposta às questões (i) quais os contributos de um instrumento de apoio à organização e estruturação dos momentos de comunicação matemática? e (ii) de que forma esse mesmo instrumento contribui para o desenvolvimento de estratégias de resolução de problemas?, investigou-se a evolução da turma quanto à comunicação das resoluções dos problemas e à variedade das estratégias utilizadas, antes e após a construção coletiva do instrumento de apoio. A análise de resultados, feita através da análise das produções dos alunos e das respostas a um questionário que instigou a reflexão sobre o papel do instrumento de apoio ao longo das várias apresentações do problema da semana, permitiu perceber uma crescente variedade de estratégias utilizadas e uma maior preocupação, por parte dos alunos, em organizar e estruturar os momentos de comunicação matemática, bem como em mobilizar um discurso com correção matemática. Este é um estudo que se limita a uma turma, não sendo as suas conclusões generalizáveis, mas que pode contribuir com uma estratégia para melhorar a comunicação matemática dos alunos, de uma forma que os envolve de forma direta e que prevê a sua participação e papel ativo na aprendizagem.ABSTRACT Creating classroom environments where students are effective agents of their learning process and have space to participate and exhibit their skills, knowing that they will be heard by the teacher and classmates, is an increasingly curricular recomendation and object of stdy nowadays. This report gives an account of the intervention carried out on a 4th grade level class, with the purpose of promoting learning, valuing, systematically, the communication. At the same time, explains research performed in this class, which involved the evaluation of the impact of the construction of an instrument in support of Mathematical Communication moments, in the context of a weekly routine of the class, The Presentation of rhe Problem of the Week. With the pretense of responding to questions (i) what are the contributions of an instrument of support to the organization and structuring of moments of mathematical communication? and (ii) how this same instrument contributes to the development of problem solving strategies?, it was investigated the evolution of the class regarding the communication of the resolutions of the problems and the variety of strategies used in problem solving, before and after the collective construction of the instrument. The analysis of results, through the analysis of students’ productions and the replies to a questionnaire that instigated the reflection about the role of the instrument of support along the various presentations of the problem of the week, allowed to realize a growing variety of strategies used and greater concern, on the part of students, to organise and structure the moments of mathematical communication as well as in mobilizing a discourse with mathematical correction. This is a study that is limited to a class, which makes its conclusions not generalizable, but that can contribute with a strategy to improve students’ math communication, in a way that involves them directly and provides their participation and active role in their own learning process.N/

    Molecular cloning of a hyaluronidase fromBothrops pauloensisvenom gland

    Background Hyaluronate is one of the major components of extracellular matrix from vertebrates whose breakdown is catalyzed by the enzyme hyaluronidase. These enzymes are widely described in snake venoms, in which they facilitate the spreading of the main toxins in the victim’s body during the envenoming. Snake venoms also present some variants (hyaluronidases-like substances) that are probably originated by alternative splicing, even though their relevance in envenomation is still under investigation. Hyaluronidases-like proteins have not yet been purified from any snake venom, but the cDNA that encodes these toxins was already identified in snake venom glands by transcriptomic analysis. Herein, we report the cloning and in silicoanalysis of the first hyaluronidase-like proteins from a Brazilian snake venom.Methods The cDNA sequence of hyaluronidase was cloned from the transcriptome of Bothrops pauloensisvenom glands. This sequence was submitted to multiple alignment with other related sequences by ClustalW. A phylogenetic analysis was performed using MEGA 4 software by the neighbor joining (NJ) method.Results The cDNA from Bothrops pauloensis venom gland that corresponds to hyaluronidase comprises 1175 bp and codifies a protein containing 194 amino acid residues. The sequence, denominated BpHyase, was identified as hyaluronidase-like since it shows high sequence identities (above 83%) with other described snake venom hyaluronidase-like sequences. Hyaluronidases-like proteins are thought to be products of alternative splicing implicated in deletions of central amino acids, including the catalytic residues. Structure-based sequence alignment of BpHyase to human hyaluronidase hHyal-1 demonstrates a loss of some key secondary structures. The phylogenetic analysis indicates an independent evolution of BpHyal when compared to other hyaluronidases. However, these toxins might share a common ancestor, thus suggesting a broad hyaluronidase-like distribution among venomous snakes.Conclusions This work is the first report of a cDNA sequence of hyaluronidase from Brazilian snake venoms. Moreover, the in silico analysis of its deduced amino acid sequence opens new perspectives about the biological function of hyaluronidases-like proteins and may direct further studies comprising their isolation and/or recombinant production, as well as their structural and functional characterization

    BthMP: a new weakly hemorrhagic metalloproteinase from Bothrops moojeni snake venom

    In this work, a new weakly hemorrhagic metalloproteinase (BthMP) was purified from Bothrops moojeni snake venom. This enzyme was homogeneous by native and SDS-PAGE. It showed a polypeptide chain of 23.5 kDa, pI=7.1, and N-terminal blocked. BthMP is comprised of high proteolytic activity on casein, fibrin and bovine fibrinogen, with no coagulating, esterase or phospholipase A(2) activities; it was inhibited by EDTA, EGTA and 1,10-phenanthroline and maintained its activity on pH from 7.0 to 9.0 and temperature from 5-40 degrees C. Assays with metal ions showed that Ca(2+) is an activator, whereas Zn(2+) and Hg(2+) inhibited about 50 and 80% of its activity, respectively. The edema evidenced the important role of the toxin in the inflammatory activity of the venom. BthMP also caused unclotting, and provoked histological alterations in the gastrocnemius muscle of mice inducing hemorrhage, necrosis and leukocytic infiltrate. The molecular mass and the inhibition assays suggest that the metal loproteinase BthMP belongs to class P-I of SVMPs. (c) 2008 Elsevier Ltd. All rights reserved

    Crotalus durissus collilineatus venom gland transcriptome: Analysis of gene expression profile

    Crotalus durissus rattlesnakes are responsible for the most lethal cases of snakebites in Brazil. Crotalus durissus collilineatus subspecies is related to a great number of accidents in Southeast and Central West regions, but few studies on its venom composition have been carried out to date. In an attempt to describe the transcriptional profile of the C. durissus collilineatus venom gland, we generated a cDNA library and the sequences obtained could be identified by similarity searches on existing databases. Out of 673 expressed sequence tags (ESTs) 489 produced readable sequences comprising 201 singletons and 47 clusters of two or more ESTs. One hundred and fifty reads (60.5%) produced significant hits to known sequences. The results showed a predominance of toxin-coding ESTs instead of transcripts coding for proteins involved in all cellular functions. The most frequent toxin was crotoxin, comprising 88% of toxin-coding sequences. Crotoxin B, a basic phospholipase A(2) (PLA(2)) subunit of crotoxin, was represented in more variable forms comparing to the non-enzymatic subunit (crotoxin A), and most sequences coding this molecule were identified as CB1 isoform from Crotalus durissus terrificus venom. Four percent of toxin-related sequences in this study were identified as growth factors, comprising five sequences for vascular endothelial growth factor (VEGF) and one for nerve growth factor (NGF) that showed 100% of identity with C. durissus terrificus NGF. We also identified two clusters for metalloprotease from PII class comprising 3% of the toxins, and two for serine proteases, including gyroxin (2.5%). The remaining 2.5% of toxin-coding ESTs represent singletons identified as homologue sequences to cardiotoxin, convulxin, angiotensin-converting enzyme inhibitor and C-type natriuretic peptide, Ohanin, crotamin and PLA(2) inhibitor. These results allowed the identification of the most common classes of toxins in C. durissus collilineatus snake venom, also showing some unknown classes for this subspecies and even for C. durissus species, such as cardiotoxins and VEGF.     Biochemical and functional properties of a thrombin-like enzyme isolated from Bothrops pauloensis snake venom

    In the present study, a thrombin-like enzyme named BpSP-I was isolated from Bothrops pauloensis snake venom and its biochemical, enzymatic and pharmacological characteristics were determined. BpSP-I is a glycoprotein that contains both N-linked carbohydrates and sialic acid in its structure, with M(r) = 34,000 under reducing conditions and pI similar to 6.4. The N-terminal sequence of the enzyme (VIGGDECDINEHPFL) showed high similarity with other thrombin-like enzymes from snake venoms. BpSP-I showed high clotting activity upon bovine and human plasma and was inhibited by PMSF, benzamidine and leupeptin. Moreover, this enzyme showed stability when examined at different temperatures (-70 to 37 degrees C), pH values (3-9) or in the presence of divalent metal ions (Ca(2+), Mg(2+), Zn(2+) and Mn(2+)). BpSP-I showed high catalytic activity upon substrates, such as fibrinogen, TAME, S-2238 and S-2288. It also showed kallikrein-like activity, but was unable to act upon factor Xa and plasmin substrates. Indeed, the enzyme did not induce hemorrhage, myotoxicity or edema. Taken together, our data showed that BpSP-I is in fact a thrombin-like enzyme isoform isolated from Bothrops pauloensis snake venom. (C) 2009 Elsevier Ltd.     Bothrops alternatus Duméril, Bibron & Duméril, 1854 é uma serpente de importância em saúde pública, com ampla distribuição geográfica, desde o Mato Grosso do Sul até o sudeste do Brasil, chegando até a Argentina e Uruguai, ocupando vários domínios morfoclimáticos. Neste trabalho investigou-se a variação do veneno de adultos de Bothrops alternatus, em função de sua distribuição geográfica no Brasil, comparativamente ao veneno elaborado sob a forma de "pool" desta espécie (veneno referência), que inclui serpentes, em sua maioria, da região do estado de São Paulo. Foram analisadas as atividades letal, coagulante sobre o plasma, proteolítica sobre a caseína e miotóxica, bem como os padrões eletroforéticos de 61 amostras individuais de veneno contrapostas ao "pool". Os resultados mostraram que o veneno de B. alternatus é pouco ativo, comparativamente ao de outros Bothrops Wagler, 1824. A variação individual prevaleceu, não apresentando correlação com as áreas de distribuição geográfica e domínios morfoclimáticos, porém a atividade coagulante das amostras de veneno provenientes do nordeste da distribuição geográfica apresentaram-se menos ativas comparativamente às da porção central da distribuição. Os venenos provenientes das bordas da distribuição apresentaram ações proteolíticas e miotóxicas mais intensas, que estatisticamente não foram significativamente diferentes. As variações individuais prevaleceram.<br>Bothrops alternatus Duméril, Bibron & Duméril, 1854 snakebites are an important public health problem in Brazil. Such snakes are found from Mato Grosso do Sul (central Brazil) to southeastern Brazil, reaching even Argentina and Uruguay and thereby occupying different morphoclimatic domains. This work investigated venom variation occurring in adult specimens of B. alternatus specimens, according to their geographic distribution in Brazil. The standard venom pool (reference venom) produced by Instituto Butantan, which includes mostly venoms from B. alternatus specimens captured in São Paulo State, was also used for comparison. Lethal, myotoxic, proteolytic (on casein), and coagulant (on human plasma) activities, as well as the electrophoretic patterns of 61 individual venom samples, were evaluated and compared to those of the reference venom pool. Results showed that B. alternatus venom activities are less potent than those of other Bothrops Wagler, 1824 species. A great individual variation was noticed, which could not be correlated with either geographic distribution or morphoclimatic domains. Coagulant activity of venom samples obtained from specimens captured in the northeastern area of B. alternatus distribution was less intense if compared to those obtained from the central zone. Venoms from the peripheric area of B. alternatus distribution presented higher proteolytic and myotoxic activities, but no statistically significant difference was observed.
