    Explaining Individual Differences in Executive Functions Performance in Multilinguals: The Impact of Code-Switching and Alternating Between Multicultural Identity Styles

    This study sheds new light on the relative impact of switching between languages and switching between cultures on Executive Functions (EFs) in bilinguals. Several studies have suggested that bilingualism has a measurable impact on executive functioning, presumably due to bilinguals’ constant practice in dealing with two languages, or two cultures. Yet, the evidence on the relative contribution of culture and bilingualism to EFs is not well understood, because disentangling language, culture and immigration status is very difficult. The novelty of our approach was to keep the language pair and immigration status constant, whilst the cultural identity of participants was systematically varied, and measured at the individual level (not just at group level). Two groups of Turkish–English bilinguals, all adult immigrants to the United Kingdom, took part in the study, but one group (n = 29) originated from mainland Turkey and the other (n = 28) from Cyprus. We found that the bilinguals experienced smaller Conflict Effects on a Flanker task measuring inhibition, by comparison with monolingual British participants (n = 30). The key variable explaining EF performance variance at the individual level turned out to be bilinguals’ Multicultural Identity Style. In particular those who indicated that they attempted to alternate between different British and Turkish (Cypriot) identity styles were found to have shorter RTs on incongruent trials of the Flanker task. The two multicultural identity variables, Alternating and Hybrid Identity Styles, together explained 32% in RTs over and above Education, Working Memory and Nonverbal reasoning (overall explained variance 49%). Thus, the data provide strong evidence for the impact of culture on EFs. We suggest that, as a result of their daily practice in recognizing cultural cues which highlight the need to switch to a different cultural frame, multicultural bilinguals develop a heightened context-sensitivity, and this gives them an advantage over monolinguals in a Flankers task. Our approach, which draws on models from cross-cultural psychology, bilingualism and executive functioning, illustrates the importance of theory building in which sociolinguistic and cultural variables are integrated into models of EFs

    Defusing easy arguments for numbers

    Pairs of sentences like the following pose a problem for ontology: (1) Jupiter has four moons. (2) The number of moons of Jupiter is four. (2) is intuitively a trivial paraphrase of (1). And yet while (1) seems ontologically innocent, (2) appears to imply the existence of numbers. Thomas Hofweber proposes that we can resolve the puzzle by recognizing that sentence (2) is syntactically derived from, and has the same meaning as, sentence (1). Despite appearances, the expressions ‘the number of moons of Jupiter’ and ‘four’ do not function semantically as singular terms in (2). Hofweber’s primary evidence for this proposal concerns differences in the focus-related communicative functions of (1) and (2). In this paper I raise several serious problems for Hofweber’s proposal, and for his attempt to support it by appeal to focus-related phenomena. I conclude by offering independent evidence for an alternative, purely pragmatic resolution of the ontological puzzle

    Nuclear import receptors directly bind to arginine-rich dipeptide repeat proteins and suppress their pathological interactions

    Nuclear import receptors, also called importins, mediate nuclear import of proteins and chaperone aggregation-prone cargoes (e.g., neurodegeneration-linked RNA-binding proteins [RBPs]) in the cytoplasm. Importins were identified as modulators of cellular toxicity elicited by arginine-rich dipeptide repeat proteins (DPRs), an aberrant protein species found in C9orf72-linked amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Mechanistically, the link between importins and arginine-rich DPRs remains unclear. Here, we show that arginine-rich DPRs (poly-GR and poly-PR) bind directly to multiple importins and, in excess, promote their insolubility and condensation. In cells, poly-GR impairs Impα/ÎČ-mediated nuclear import, including import of TDP-43, an RBP that aggregates in C9orf72-ALS/FTD patients. Arginine-rich DPRs promote phase separation and insolubility of TDP-43 in vitro and in cells, and this pathological interaction is suppressed by elevating importin concentrations. Our findings suggest that importins can decrease toxicity of arginine-rich DPRs by suppressing their pathological interactions

    The guanine nucleotide binding protein Ras plays a central role as molecular switch in cellular signal transduction. Ras cycles between a GDP-bound "off" state and a GTP-bound "on" state. Specific oncogenic mutations in the Ras protein are found in up to 30% of all human tumors. Previous 31P NMR studies had demonstrated that in liquid solution different conformational states in the GDP-bound as well as in the GTP-bound form coexist. High-field EPR spectroscopy of the GDP complexes in solution displayed differences in the ligand sphere of the wild-type complex as compared to its oncogenic mutant Ras(G12V). Only three water ligands were found in the former with respect to four in the G12V mutant [Rohrer, M. et al. (2001) Biochemistry 40, 1884-1889]. These differences were not detected in previous X-ray structures in the crystalline state. In this paper, we employ high-frequency electron nuclear double resonance (ENDOR) spectroscopy to probe the ligand sphere of the metal ion in the GDP-bound state. This technique in combination with selective isotope labeling has enabled us to detect the resonances of nuclei in the first ligand sphere of the ion with high spectral resolution. We have observed the 17O ENDOR spectra of the water ligands, and we have accurately determined the 17O hyperfine coupling with aiso = -0.276 mT, supporting the results of previous line shape analysis in solution. Further, the distinct resonances of the -, -, and -phosphorus of the bound nucleotides are illustrated in the 31P ENDOR spectra, and their hyperfine tensors lead to distances in agreement with the X-ray structures. Finally, 13C ENDOR spectra of uniformly 13C-labeled Ras(wt)·GDP and Ras(G12V)·GDP complexes as well as of the Ras(wt)·GppNHp and the selectively 1,4-13C-Asp labeled Ras(wt)·GDP complexes have revealed that in frozen solution only one amino acid is ligated to the ion in the GDP state, whereas two are bound in the GppNHp complex. Our results suggest that a second conformational state of the protein, if correlated with a different ligand sphere of the Mn2+ ion, is not populated in the GDP form of Ras at low temperatures in frozen solution
