Protein transduction: A novel tool for tissue regeneration

    Get PDF
Tissue regeneration in humans is limited and excludes vitals organs like heart and brain. Transformation experiments with oncogenes like T antigen have shown that retrodifferentiation of the respective cells is possible but hard to control. To bypass the risk of cancer formation a protein therapy approach has been developed. The transient delivery of proteins rather than genes could still induce terminallydifferentiated cells to reenter the cell cycle. This approach takes advantage of proteintransducing domains that mediate the transfer of cargo proteins into cells. The goal of this brief review is to outline the basics of protein transduction and to discuss potential applications for tissue regeneration

Supersymmetric reduced models with a symmetry based on Filippov algebra

Generalizations of the reduced model of super Yang-Mills theory obtained by replacing the Lie algebra structure to Filippov nn-algebra structures are studied. Conditions for the reduced model actions to be supersymmetric are examined. These models are related with what we call \{cal N}_{min}=2 super pp-brane actions.Comment: v3: In the previous versions we overlooked that Eq.(3.9) holds more generally, and missed some supersymmetric actions. Those are now included and modifications including a slight change in the title were made accordingly. 1+18 page

Immunogene therapy with fusogenic nanoparticles modulates macrophage response to Staphylococcus aureus.

The incidence of adverse effects and pathogen resistance encountered with small molecule antibiotics is increasing. As such, there is mounting focus on immunogene therapy to augment the immune system's response to infection and accelerate healing. A major obstacle to in vivo gene delivery is that the primary uptake pathway, cellular endocytosis, results in extracellular excretion and lysosomal degradation of genetic material. Here we show a nanosystem that bypasses endocytosis and achieves potent gene knockdown efficacy. Porous silicon nanoparticles containing an outer sheath of homing peptides and fusogenic liposome selectively target macrophages and directly introduce an oligonucleotide payload into the cytosol. Highly effective knockdown of the proinflammatory macrophage marker IRF5 enhances the clearance capability of macrophages and improves survival in a mouse model of Staphyloccocus aureus pneumonia

Duality of Quasilocal Black Hole Thermodynamics

    Get PDF
We consider T-duality of the quasilocal black hole thermodynamics for the three-dimensional low energy effective string theory. Quasilocal thermodynamic variables in the first law are explicitly calculated on a general axisymmetric three-dimensional black hole solution and corresponding dual one. Physical meaning of the dual invariance of the black hole entropy is considered in terms of the Euclidean path integral formulation.Comment: 19 pages, Latex, no figures, to be published in Class. Geodesic Motions in 2+1 Dimensional Charged Black Holes

    Get PDF
We study the geodesic motions of a test particle around 2+1 dimensional charged black holes. We obtain a class of exact geodesic motions for the massless test particle when the ratio of its energy and angular momentum is given by square root of cosmological constant. The other geodesic motions for both massless and massive test particles are analyzed by use of numerical method.Comment: 13page

Three-algebra for supermembrane and two-algebra for superstring

While string or Yang-Mills theories are based on Lie algebra or two-algebra structure, recent studies indicate that M-theory may require a one higher, three-algebra structure. Here we construct a covariant action for a supermembrane in eleven dimensions, which is invariant under global supersymmetry, local fermionic symmetry and worldvolume diffeomorphism. Our action is classically on-shell equivalent to the celebrated Bergshoeff-Sezgin-Townsend action. However, the novelty is that we spell the action genuinely in terms of Nambu three-brackets: All the derivatives appear through Nambu brackets and hence it manifests the three-algebra structure. Further the double dimensional reduction of our action gives straightforwardly to a type IIA string action featuring two-algebra. Applying the same method, we also construct a covariant action for type IIB superstring, leading directly to the IKKT matrix model.Comment: 1+15 pages, no figure; Refs added, Accepted for publication in JHE

    Lineal Trails of D2-D2bar Superstrings

We study the superstrings suspended between a D2- and an anti-D2-brane. We quantize the string in the presence of some general configuration of gauge fields over the (anti-)D-brane world volumes. The interstring can move only in a specific direction that is normal to the difference of the electric fields of each (anti-)D-branes. Especially when the electric fields are the same, the interstring cannot move. We obtain the condition for the tachyons to disappear from the spectrum.Comment: 15 pages with 4 figures, referenced added, Sec. 5 on the spectrum made cleare

Bucillamine prevents cisplatin-induced ototoxicity through induction of glutathione and antioxidant genes.

Bucillamine is used for the treatment of rheumatoid arthritis. This study investigated the protective effects of bucillamine against cisplatin-induced damage in auditory cells, the organ of Corti from postnatal rats (P2) and adult Balb/C mice. Cisplatin increases the catalytic activity of caspase-3 and caspase-8 proteases and the production of free radicals, which were significantly suppressed by pretreatment with bucillamine. Bucillamine induces the intranuclear translocation of Nrf2 and thereby increases the expression of Îł-glutamylcysteine synthetase (Îł-GCS) and glutathione synthetase (GSS), which further induces intracellular antioxidant glutathione (GSH), heme oxygenase 1 (HO-1) and superoxide dismutase 2 (SOD2). However, knockdown studies of HO-1 and SOD2 suggest that the protective effect of bucillamine against cisplatin is independent of the enzymatic activity of HO-1 and SOD. Furthermore, pretreatment with bucillamine protects sensory hair cells on organ of Corti explants from cisplatin-induced cytotoxicity concomitantly with inhibition of caspase-3 activation. The auditory-brainstem-evoked response of cisplatin-injected mice shows marked increases in hearing threshold shifts, which was markedly suppressed by pretreatment with bucillamine in vivo. Taken together, bucillamine protects sensory hair cells from cisplatin through a scavenging effect on itself, as well as the induction of intracellular GSH
