    Direction Detector on an Excitable Field: Field Computation with Coincidence Detection

    Living organisms process information without any central control unit and without any ruling clock. We have been studying a novel computational strategy that uses a geometrically arranged excitable field, i.e., "field computation." As an extension of this research, in the present article we report the construction of a "direction detector" on an excitable field. Using a numerical simulation, we show that the direction of a input source signal can be detected by applying the characteristic as a "coincidence detector" embedded on an excitable field. In addition, we show that this direction detection actually works in an experiment using an excitable chemical system. These results are discussed in relation to the future development of "field computation."Comment: 6 pages, 3 figure

    Ultradian oscillations of Stat, Smad, and Hes1 expression in response to serum

    Serum response has been used as a model for studying signaling transduction for many biological events such as cell proliferation and survival. Although expression of many genes is up- or down-regulated after serum stimulation, the Notch effector Hes1 displays oscillatory response. However, the precise mechanism and biological significance of this oscillation remain to be determined. Here, we identified serum-induced ultradian oscillators, including molecules in Stat and Smad signaling. Stat and Smad oscillations involve activation of Stat3 and Smad1 and delayed negative feedback by their inhibitors Socs3 and Smad6, respectively. Moreover, Stat oscillations induce oscillatory expression of Hes1 by regulating its half-life, and loss of Hes1 oscillations leads to G1 phase retardation of the cell cycle. These results indicate that coupled Stat and Hes1 oscillations are important for efficient cell proliferation and provide evidence that expression modes of signaling molecules affect downstream cellular events

    RELATIONSHIP OF BASE-RUNNING PERFORMANCE WITH RUNNING DIRECTION AND ITS CHANGE

    The purpose of this study was to clarify whether the running direction and its change affect the base-running performance. Thirty-five male baseball players performed 54.86 m (180 ft.) straight-line sprinting and distance-matched base-running, during which two dimensional positions of players were recorded with B Y Sports Tracking System. The results of this study revealed that the total base-running performance was probably affected by the running performance of the later running phase. In addition, smaller direction change before the first base and larger direction change after the first base maybe responsible for better performance in the entire base-running through better performance in the later section. These results would be useful for players and coaches who attempt to improve base-running performance

    Short Stat5-Interacting Peptide Derived from Phospholipase C-β3 Inhibits Hematopoietic Cell Proliferation and Myeloid Differentiation

    Constitutive activation of the transcription factor Stat5 in hematopoietic stem/progenitor cells leads to various hematopoietic malignancies including myeloproliferative neoplasm (MPN). Our recent study found that phospholipase C (PLC)-β3 is a novel tumor suppressor involved in MPN, lymphoma and other tumors. Stat5 activity is negatively regulated by the SH2 domain-containing protein phosphatase SHP-1 in a PLC-β3-dependent manner. PLC-β3 can form the multimolecular SPS complex together with SHP-1 and Stat5. The close physical proximity of SHP-1 and Stat5 brought about by interacting with the C-terminal segment of PLC-β3 (PLC-β3-CT) accelerates SHP-1-mediated dephosphorylation of Stat5. Here we identify the minimal sequences within PLC-β3-CT required for its tumor suppressor function. Two of the three Stat5-binding noncontiguous regions, one of which also binds SHP-1, substantially inhibited in vitro proliferation of Ba/F3 cells. Surprisingly, an 11-residue Stat5-binding peptide (residues 988-998) suppressed Stat5 activity in Ba/F3 cells and in vivo proliferation and myeloid differentiation of hematopoietic stem/progenitor cells. Therefore, this study further defines PLC-β3-CT as the Stat5- and SHP-1-binding domain by identifying minimal functional sequences of PLC-β3 for its tumor suppressor function and implies their potential utility in the control of hematopoietic malignancies

    Survival versus collapse: Abrupt drop of excitability kills the traveling pulse, while gradual change results in adaptation

    Excitable media show changes in their basic characteristics that reflect temporal changes in the environment. In the photosensitive Belousov-Zhabotinsky (BZ) reaction, excitability is decreased by illumination. We found that a traveling pulse failed to propagate when a certain level of light intensity was switched on abruptly, but the pulse continued propagating when the light intensity reached the same level gradually. We investigated the mechanism of adaptation of pulse propagation to the change in light intensity using two mathematical models, the Oregonator model (a specific model for the photosensitive BZ reaction), and the FitzHugh-Nagumo model (a generic model for excitable media). The appearance of a characteristic such as adaptation is shown to be a general feature for a traveling pulse in excitable media. © 2007 The American Physical Society
