    Expression analysis of a mouse orthologue of HSFY, a candidate for the azoospermic factor on the human Y chromosome

    Heat shock transcription factor on Y (HSFY) is located in one of three candidate regions for azoospermic factor (AZF), AZFb on the Y chromosome. We and others have already revealed that some azoospermic males are missing the regions of the Y chromosome including HSFY. Previously, we showed that murine HSFY-like sequence〔mHSFYL(RikencDNA4933413G11Rik)〕, which is the mouse orthologue of HSFY, is exclusively expressed in testis. The sequences encoding the presumed DNA-binding domain in HSFY and mHSFYL were found in other mammals such as dogs, cows and chickens. To elucidate mHSFYL expression in the testes in detail, we carried out in situ hybridization. mHSFYL was predominantly expressed in round spermatids. Furthermore, we clarified the intracellular distribution of mHSFYL inCOS1 cells with HA- or GFP-tagged proteins. Both HA-mHSFYL and GFP-mHSFYL were located in the nucleus. Our results suggest that HSFY/mHSFYL may have evolutionarily conserved functions for spermatogenesis

    Use of Cap Analysis Gene Expression to detect human papillomavirus promoter activity patterns at different disease stages

    Transcription of human papillomavirus (HPV) genes proceeds unidirectionally from multiple promoters. Direct profiling of transcription start sites (TSSs) by Cap Analysis Gene Expression (CAGE) is a powerful strategy for examining individual HPV promoter activity. The objective of this study was to evaluate alterations of viral promoter activity during infection using CAGE technology. We used CAGE-based sequencing of 46 primary cervical samples, and quantitatively evaluated TSS patterns in the HPV transcriptome at a single-nucleotide resolution. TSS patterns were classified into two types: early promoter-dominant type (Type A) and late promoter-dominant type (Type B). The Type B pattern was more frequently found in CIN1 and CIN2 lesions than in CIN3 and cancer samples. We detected transcriptomes from multiple HPV types in five samples. Interestingly, in each sample, the TSS patterns of both HPV types were the same. The viral gene expression pattern was determined by the differentiation status of the epithelial cells, regardless of HPV type. We performed unbiased analyses of TSSs across the HPV genome in clinical samples.     An 83-year-old female developed diffuse large B-cell lymphoma(DLBL) of the left nasal cavity. Complete remission was achieved after two courses of Rituximab and CHOP(R-CHOP) . During the fourth course of R-CHOP, sensory disturbance and palsy of the left face developed. Left trigeminal nerve swelling was observed in magnetic resonance imaging(MRI) followed by double vision in the left eye, and MRI revealed swelling of both trigeminal nerves but not of the abducens nerve. Although the swelling of the trigeminal nerves and the double vision subside after administration of prednisolone, the palsy of the left face persisted. Two months after the fourth course of R-CHOP, symptoms of the palsy of the left face progressed and palsy of the right face, double vision, and palsy of the left facialis nerve developed. Then,blepharoptosis of the right eye developed and palsy of the right oculomotorius nerve was observed. MRI showed the presence of trigeminal nerve and oculomotorius nerve swelling but no swelling of the other cranial nerves. Furthermore, skin eruption developed around the left eye.Cytology of this lesion revealed the invasion of lymphoma cells

    Cognitive behavioral therapy for depression among adults in Japanese clinical settings: a single-group study

    <p>Abstract</p> <p>Background</p> <p>Empirical support for cognitive behavioral therapy (CBT) for treating Japanese patients with major depression is lacking, therefore, a feasibility study of CBT for depression in Japanese clinical settings is urgently required.</p> <p>Findings</p> <p>A culturally adapted, 16-week manualized individual CBT program for Japanese patients with major depressive disorder was developed. A total of 27 patients with major depression were enrolled in a single-group study with the purpose of testing the feasibility of the program. Twenty six patients (96%) completed the study. The mean total score on the Beck Depression Inventory-II (BDI-II) for all patients (Intention-to-treat sample) improved from 32.6 to 11.7, with a mean change of 20.8 (95% confidence interval: 17.0 to 24.8). Within-group effect size at the endpoint assessment was 2.64 (Cohen's d). Twenty-one patients (77.7%) showed treatment response and 17 patients (63.0%) achieved remission at the end of the program. Significant improvement was observed in measurement of subjective and objective depression severity (assessed by BDI-II, Quick Inventory of Depressive Symptomatology-Self Rated, and Hamilton Depression Rating Scale), dysfunctional attitude (assessed by Dysfunctional Attitude Scale), global functioning (assessed by Global Assessment of Functioning of DSM-IV) and subjective well-being (assessed by WHO Subjective Well-being Inventory) (all p values < 0.001).</p> <p>Conclusions</p> <p>Our manualized treatment comprised of a 16-week individual CBT program for major depression appears feasible and may achieve favorable treatment outcomes among Japanese patients with major depression.     Water Column Profiles of Dissolved Inorganic Radiocarbon for the Kuroshio Region, Offshore of the Southern Japanese Coast

    We present the water column profiles (surface to 2000 m depth) for dissolved inorganic radiocarbon ([delta]14CDIC) from 2 stations in the Kuroshio region including the Kuroshio large meander (LM) of 2004–2005. Surprisingly, the [delta]14CDIC value varied up to 125‰ in the intermediate layer, especially near 600 m depth. In addition, the [delta]14CDIC value was approximately -150‰ at 200 m depth at the northern station of Kuroshio in August 2005. This value is ~100‰ less than other [delta]14CDIC values for the same depth. In comparison, the [delta]14CDIC water column profiles for the southern station of Kuroshio and GEOSECS station 224 decrease down to 600 m depth and were similar below 600 m depth. Our results suggest that strong upwelling associated with the Kuroshio LM has a powerful influence on the [delta]14CDIC water column profiles in the study region.The Radiocarbon archives are made available by Radiocarbon and the University of Arizona Libraries.     Cytogenetic analysis in a multiple myeloma patient who developed myelomatous pleural effusion and plasma cell leukemia

    We report on a patient with multiple myeloma who developed myelomatous pleural effusion and plasma cell leukemia. After the fourth course of chemotherapy, pleural effusion developed, and plasma cells were observed in peripheral blood after the sixth course of chemotherapy. Homozygous loss of chromosome 16 and monosomy X developed before the fourth course of chemotherapy. In this case, such cytogenetic abnormalities may be related to the myelomatous effusion and leukemic changes in multiple myelomas
