    Transcriptional profiling avian beta-defensins in chicken oviduct epithelial cells before and after infection with Salmonella enterica serovar Enteritidis

    <p>Abstract</p> <p>Background</p> <p><it>Salmonella enterica </it>serovar Enteritidis (SE) colonizes the ovary and oviduct of chickens without causing overt clinical signs which can lead to SE-contamination of the content and membrane of shell-eggs as well as hatchery eggs. The organism utilizes the <it>Salmonella </it>Pathogenicity Island-2 encoded type III secretion system (T3SS-2) to promote persistence in the oviduct of laying hens. In this study, reverse transcriptase-polymerase chain reaction (RT-PCR) was carried out to determine the expression profiles of 14 known avian beta defensins (AvBDs) in primary chicken oviduct epithelial cells (COEC) before and after infections with a wild type SE strain and T3SS mutant SE strains carrying an inactivated <it>sipA </it>or <it>pipB </it>gene.</p> <p>Results</p> <p>Based on the expression levels in uninfected COEC, AvBDs can be loosely grouped into three categories with AvBD4-5 and AvBD9-12 being constitutively expressed at high levels; AvBD1, AvBD3, and AvBD13-14 at moderate levels; and AvBD2 and AvBD6-8 at minimal levels. Infection with the wild type SE strain temporarily repressed certain highly expressed AvBDs and induced the expression of minimally expressed AvBDs. The <it>pipB </it>mutant, compared to the wild type strain, had reduced suppressive effect on the expression of highly expressed AvBDs. Moreover, the <it>pipB </it>mutant elicited significantly higher levels of the minimally expressed AvBDs than the wild type SE or the <it>sipA </it>mutant did.</p> <p>Conclusion</p> <p>Chicken oviduct epithelial cells express most of the known AvBD genes in response to SE infection. PipB, a T3SS-2 effector protein, plays a role in dampening the β-defensin arm of innate immunity during SE invasion of chicken oviduct epithelium.</p

    Burden of illness of trigeminal neuralgia among patients managed in a specialist center in England

    BACKGROUND: Trigeminal neuralgia (TN) causes severe episodic, unilateral facial pain and is initially treated with antiepileptic medications. For patients not responding or intolerant to medications, surgery is an option. METHODS: In order to expand understanding of the pain-related burden of illness associated with TN, a cross-sectional survey was conducted of patients at a specialist center that utilizes a multidisciplinary care pathway. Participants provided information regarding their pain experience and treatment history, and completed several patient-reported outcome (PRO) measures. RESULTS: Of 129 respondents, 69/128 (54%; 1 missing) reported no pain in the past 4 weeks. However, 84 (65%) respondents were on medications, including 49 (38%) on monotherapy and 35 (27%) on polytherapy. A proportion of patients had discontinued at least one medication in the past, mostly due to lack of efficacy (n = 62, 48%) and side effects (n = 51, 40%). A total of 52 (40%) patients had undergone surgery, of whom 30 had microvascular decompression (MVD). Although surgery, especially MVD, provided satisfactory pain control in many patients, 29% of post-surgical patients reported complications, 19% had pain worsen or stay the same, 48% were still taking pain medications for TN, and 33% reported new and different facial pain. CONCLUSIONS: In most PRO measures, respondents with current pain interference had poorer scores than those without pain interference. In the Patient Global Impression of Change, 79% expressed improvement since beginning of treatment at this clinic. These results indicate that while the multidisciplinary approach can substantially alleviate the impact of TN, there remains an unmet medical need for additional treatment options

    Origin of the ϕ ∼ ±9° peaks in YBa2Cu3O7−δ films grown on cubic zirconia substrates

    The c-axis oriented YBa2Cu3O7−δ films grown on (001) yttria-stabilized cubic zirconia (YSZ) substrates often contain domains whose in-plane alignment is rotated approximately 9° from the cube-on-cube epitaxial relationship, in addition to the more commonly observed 0° and 45° in-plane rotations. We have investigated the origin of this ∼9° orientation using in situ electron diffraction during growth and ex situ 4-circle x-ray diffraction. Our results indicate that the ∼9° orientation provides the most favorable lattice match between the interfacial (110)-oriented BaZrO3 epitaxial reaction layer, which forms between YBa2Cu3O7−δ and the YSZ substrate. If epitaxy occurs directly between YBa2Cu3O7−δ and the YSZ substrate, i.e., before the BaZrO3 epitaxial reaction layer is formed, the 0° and 45° domains have the most favorable lattice match. However, growth conditions that favor the formation of the BaZrO3 reaction layer prior to the nucleation of YBa2Cu3O7−δ lead to an increase in ∼9° domains. The observed phenomenon, which results from epitaxial alignment between the diagonal of a square surface net and the diagonal of a rectangular surface net, is a general method for producing in-plane misorientations, and has also been observed for the heteroepitaxial growth of other materials, including (Ba, K)BiO3/LaAlO3. The YBa2Cu3O7−δ/YSZ case involves epitaxial alignment between [111]BaZrO3 and [110]YSZ, resulting in an expected in-plane rotation of 11.3° to 9.7° for fully commensurate and for fully relaxed (110)BaZrO3 on (001)YSZ, respectivel

    Are there gender differences in the geography of alcohol-related mortality in Scotland? An ecological study

    &lt;b&gt;Background&lt;/b&gt; There is growing concern about alcohol-related harm, particularly within Scotland which has some of the highest rates of alcohol-related death in western Europe. There are large gender differences in alcohol-related mortality rates in Scotland and in other countries, but the reasons for these differences are not clearly understood. In this paper, we aimed to address calls in the literature for further research on gender differences in the causes, contexts and consequences of alcohol-related harm. Our primary research question was whether the kind of social environment which tends to produce higher or lower rates of alcohol-related mortality is the same for both men and women across Scotland. &lt;b&gt;Methods&lt;/b&gt; Cross-sectional, ecological design. A comparison was made between spatial variation in men's and women's age-standardised alcohol-related mortality rates in Scotland using maps, Moran's Index, linear regression and spatial analyses of residuals. Directly standardised mortality rates were derived from individual level records of death registration, 2000–2005 (n = 8685). &lt;b&gt;Results&lt;/b&gt; As expected, men's alcohol-related mortality rate substantially exceeded women's and there was substantial spatial variation in these rates for both men and women within Scotland. However, there was little spatial variation in the relationship between men's and women's alcohol-mortality rates (r2 = 0.73); areas with relatively high rates of alcohol-related mortality for men tended also to have relatively high rates for women. In a small number of areas (8 out of 144) the relationship between men's and women's alcohol-related mortality rates was significantly different. &lt;b&gt;Conclusion&lt;/b&gt; In as far as geographic location captures exposure to social and economic environment, our results suggest that the relationship between social and economic environment and alcohol-related harm is very similar for men and women. The existence of a small number of areas in which men's and women's alcohol-related mortality had an different relationship suggests that some places may have unusual drinking cultures. These might prove useful for further investigations into the factors which influence drinking behaviour in men and women

    Phase I Study of the Safety and Pharmacokinetics of Plerixafor in Children Undergoing a Second Allogeneic Hematopoietic Stem Cell Transplantation for Relapsed or Refractory Leukemia

    AbstractThe safety, pharmacokinetics, and biological effect of plerixafor in children as part of a conditioning regimen for chemo-sensitization in allogeneic hematopoietic stem cell transplantation (HSCT) have not been studied. This is a phase I study of plerixafor designed to evaluate its tolerability at dose of .24 mg/kg given intravenously on day −4 (level 1); day −4 and day −3 (level 2); or day −4, day −3, and day −2 (level 3) in combination with fludarabine, thiotepa, melphalan, and rabbit antithymocytic globulin for a second allogeneic HSCT in children with refractory or relapsed leukemia. Immunophenotype analysis was performed on blood and bone marrow before and after plerixafor administration. Twelve patients were enrolled. Plerixafor at all 3 levels was well tolerated without dose-limiting toxicity. Transient gastrointestinal side effects of National Cancer Institute–grade 1 or 2 in severity were the most common adverse events. The area under the concentration-time curve increased proportionally to the dose level. Plerixafor clearance was higher in males and increased linearly with body weight and glomerular filtration rate. The clearance decreased and the elimination half-life increased significantly from dose level 1 to 3 (P < .001). Biologically, the proportion of CXCR4+ blasts and lymphocytes both in the bone marrow and peripheral blood increased after plerixafor administration

    Short-Range Ising Spin Glass: Multifractal Properties

    The multifractal properties of the Edwards-Anderson order parameter of the short-range Ising spin glass model on d=3 diamond hierarchical lattices is studied via an exact recursion procedure. The profiles of the local order parameter are calculated and analysed within a range of temperatures close to the critical point with four symmetric distributions of the coupling constants (Gaussian, Bimodal, Uniform and Exponential). Unlike the pure case, the multifractal analysis of these profiles reveals that a large spectrum of the α\alpha -H\"older exponent is required to describe the singularities of the measure defined by the normalized local order parameter, at and below the critical point. Minor changes in these spectra are observed for distinct initial distributions of coupling constants, suggesting an universal spectra behavior. For temperatures slightly above T_{c}, a dramatic change in the F(α)F(\alpha) function is found, signalizing the transition.Comment: 8 pages, LaTex, PostScript-figures included but also available upon request.     ABSTRACT A dry-low-NOx, high-aimow-capacity fuel injection system for a leanpremixed combustor has been developed for a moderate pressure ratio (20:l) aeroderivative gas turbine engine. Engine requirements for cornbustor pressure drop, emissions, and operability have been met. Combustion performance was evaluated at high power conditions in a high-pressure, single-nozzle test facility which operates at full baseload conditions. Single digit NOx levels and high combustion efficiency were achieved A wide operability range with no signs of flashback, autoignition. or thermal problems was demonsuated.
