    Occurrence of LINE, gypsy-like, and copia-like retrotransposons in the clonally propagated sweet potato (Ipomoea batatas L.)

    Retrotransposons are a class of transposable elements that represent a major fraction of the repetitive DNA of most eukaryotes. Their abundance stems from their expansive replication strategies. We screened and isolated sequence fragments of long terminal repeat (LTR), gypsy-like reverse transcriptase (rt) and gypsy-like envelope (env) domains, and two partial sequences of non-LTR retrotransposons, long interspersed element (LINE), in the clonally propagated allohexaploid sweet potato (Ipomoea batatas (L.) Lam.) genome. Using dot-blot hybridization, these elements were found to be present in the ~1597 Mb haploid sweet potato genome with copy numbers ranging from ~50 to ~4100 as observed in the partial LTR (IbLtr-1) and LINE (IbLi-1) sequences, respectively. The continuous clonal propagation of sweet potato may have contributed to such a multitude of copies of some of these genomic elements. Interestingly, the isolated gypsy-like env and gypsy-like rt sequence fragments, IbGy-1 (~2100 copies) and IbGy-2 (~540 copies), respectively, were found to be homologous to the Bagy-2 cDNA sequences of barley (Hordeum vulgare L.). Although the isolated partial sequences were found to be homologous to other transcriptionally active elements, future studies are required to determine whether they represent elements that are transcriptionally active under normal and (or) stressful conditions

    Accuracy of multiparametric magnetic resonance imaging to detect significant prostate cancer and index lesion location

    Background: Multiparametric magnetic resonance imaging (mpMRI) of the prostate appears to improve prostate cancer detection, but studies comparing mpMRI to histopathology at the time of radical prostatectomy (RP) are lacking. This retrospective study determined the accuracy of mpMRI predicting Gleason score and index lesion location at the time of RP, the current gold standard for diagnosis. Methods: Between April 2013 and April 2016, a database of all men aged more than 40 years who underwent RP after positive transrectal ultrasound biopsy by an experienced urological surgeon was collated at a single regional centre. This was cross‐referenced with a database of all men who had mpMRIs performed at a single centre and reported according to Prostate Imaging Reporting and Data System (PI‐RADS version 1) during this period to generate a sample size of 64 men. A Spearman\u27s rho test was utilized to calculate correlation. Results: Median age of patients was 64 years, the median prostate‐specific antigen at RP was 6.22 ng/mL. mpMRI was positive (≥PI‐RADS 3) in 85.9% of patients who underwent RP. More than 92% of participants had Gleason ≥7 disease. A positive relationship between mpMRI prostate PI‐RADS score and RP cancer volume was demonstrated. An anatomical location correlation calculated in octants was found to be 89.1% accurate. Conclusion: mpMRI accurately detects prostate cancer location and severity when compared with gold standard histopathology at the time of RP. It thus has an important role in planning for future prostate biopsy and cancer treatment

    This study advances a refutation of Physicalism. It demonstrates that it cannot, coherently, be maintained. An alternative approach based on Husserl’s ‘transcendental ego’ is developed. This is an account where the physical world is constituted by a freely acting self from a phenomenology that is ontologically neutral. By doing so, the, so-called, ‘hard problem’ of consciousness is dissolved. It will be shown that the self is compelled to attribute moral and aesthetic value to the world that it has constituted. The determinism that Physicalism engenders is shown to lead to a nihilism that makes all knowledge impossible. Genuine personal responsibility for action, is asserted within strict limits. A position is outlined that gives a role to the self in causation and one that entails the possibility of panpsychism

    The spatial clustering of ultraluminous infrared galaxies over 1.5 < z < 3

    We present measurements of the spatial clustering of galaxies with stellar masses 1011 M, infrared luminosities 1012 L, and star formation rates 200 M yr-1 in two redshift intervals: 1.5 &lt; z &lt; 2.0 and 2 &lt; z &lt; 3. Both samples cluster moderately strongly, with spatial correlation lengths of r0 = 6.14 B1 0.84 h-1 Mpc for the 2 &lt; z &lt; 3 sample and r0 = 5.36 B1 1.28 h-1 Mpc for the 1.5 &lt; z &lt; 2.0 sample. These clustering amplitudes are consistent with both populations residing in dark matter halos with masses of 7 C 1012 M, which is comparable to that seen for optical QSOs at the same epochs. We infer that a minimum dark matter halo mass is an important factor for all forms of luminous, obscured activity in galaxies at z &gt; 1, both starbursts and active galactic nuclei. Adopting plausible models for the growth of dark matter halos with redshift, the halos hosting the 2 &lt; z &lt; 3 sample will likely host poor to rich clusters of galaxies at z = 0, whereas the halos hosting the 1.5 &lt; z &lt; 2.0 sample will likely host L* elliptical galaxies or poor clusters at z = 0. We conclude that ultraluminous infrared galaxies (ULIRGs) at z 2.5 likely signpost stellar buildup in galaxies that will reside in clusters at z = 0 and that ULIRGs at z 1.7 signpost stellar buildup in sources that will either become L* elliptical galaxies or reside in poor clusters at z = 0

    Evidence for variable selective pressures at MC1R

    It is widely assumed that genes that influence variation in skin and hair pigmentation are under selection. To date,the melanocortin 1 receptor (MC1R) is the only gene identified that explains substantial phenotypic variance inhuman pigmentation. Here we investigate MC1R polymorphism in several populations, for evidence of selection.We conclude that MC1R is under strong functional constraint in Africa, where any diversion from eumelanin production (black pigmentation) appears to be evolutionarily deleterious. Although many of the MC1R amino acid variants observed in non-African populations do affect MC1R function and contribute to high levels of MC1R diversity in Europeans, we found no evidence, in either the magnitude or the patterns of diversity, for its enhancement by selection; rather, our analyses show that levels of MC1R polymorphism simply reflect neutral expectations underrelaxation of strong functional constraint outside Africa

    Automated Detection of Vessel Abnormalities on Fluorescein Angiogram in Malarial Retinopathy

    The detection and assessment of intravascular filling defects is important, because they may represent a process central to cerebral malaria pathogenesis: neurovascular sequestration. We have developed and validated a framework that can automatically detect intravascular filling defects in fluorescein angiogram images. It first employs a state-of-the-art segmentation approach to extract the vessels from images and then divide them into individual segments by geometrical analysis. A feature vector based on the intensity and shape of saliency maps is generated to represent the level of abnormality of each vessel segment. An AdaBoost classifier with weighted cost coefficient is trained to classify the vessel segments into normal and abnormal categories. To demonstrate its effectiveness, we apply this framework to 6,358 vessel segments in images from 10 patients with malarial retinopathy. The test sensitivity, specificity, accuracy, and area under curve (AUC) are 74.7%, 73.5%, 74.1% and 74.2% respectively when compared to the reference standard of human expert manual annotations. This performance is comparable to the agreement that we find between human observers of intravascular filling defects. Our method will be a powerful new tool for studying malarial retinopathy

    Teicoplanin in the treatment of infections caused by coagulase-negative staphylococci

    A total of 88 patients were treated with teicoplanin for infections caused by coagulase-negative staphylococci in open clinical studies in France and the UK. Teicoplanin was administered once daily, with a mean dose of 323 mg, for a mean duration of 16 days. Thirty-nine patients received teicoplanin alone while 49 received combination treatment. Clinical cure or improvement occurred in 79 of 82 evaluable cases (96-3%) and bacteriological elimination in 82 cases (95-1%). Treatment with teicoplanin alone was clinically successful in all cases, including 20 septicaemias, and the elimination rate was 93%. There was no apparent correlation between clinical and bacteriological outcomes and results of in-vitro testing. The variation in MICs between France and the UK was attributed to differences in the methods used
