The origin of pine pollen grains captured from air at Calypsobyen, Svalbard
Spitsbergen is the largest island in the Svalbard Archipelago (Norway) that has been permanently populated. The harsh Arctic climate prevents development of large vascular plants such as trees. A two-year aerobiological survey was conducted within the framework of two consecutive polar expeditions (2014 and 2015) in Spitsbergen (Calypsobyen, Bellsund).
The air quality was measured continuously from June/July to August using a 7-day volumetric air sampler, Tauber trap and moss specimens. Collected air samples and gravimetric pollen
deposits were processed following transfer to sterile laboratory conditions and analyzed with the aid of light microscopy. Days when pine pollen grains were detected in the air were selected for further analysis. Clusters of back-trajectories, computed using the Hybrid Single Particle Lagrangian Integrated Trajectory model in combination with ArcGIS software as well as the Flextra trajectory model, showed the movement of air masses to the sampling location
at Hornsund, and thus indicated the likely origin of pollen grains. The GlobCover 2009 and CORINE Land Cover 2012 datasets were employed to establish the distribution of coniferous forests in the areas of interest. Conclusions were drawn based on the analyses of the circulation of air masses, using visualization of global weather conditions forecast to
supercomputers. For the first time, we have demonstrated that pine pollen grains occurring in pine-free Spitsbergen, could originate from numerous locations, including Scandinavia,
Iceland, Siberia and northern Canada. Pollen grains were transported via air masses for distances exceeding ~2000 km. Both air samples and gravimetric pollen deposits revealed the
same pattern of Pinus pollen distributio

Histochemical investigation of trichomes and chemical composition of essential oil from Euphrasia stricta D. Wolff ex J.F. Lehm. (Orobanchaceae)
Micromorphological and anatomical investigations of trichomes on vegetative and generative organs of the drug eyebright (Euphrasia stricta D. Wolff ex J.F. Lehm.) were carried out. Additionally, identification of the main components of the secreted products was conducted. The following types of glandular structures were found: capitate trichomes (i) with a unicellular head, a neck cell, and a 2-celled stalk, (ii) with a unicellular head, a neck cell, and 3-celled stalk, (iii) with a bicellular head and sporadically with a 3- or 4-celled head; moreover, on the surface of corolla unicellular and 2–3-celled clavate trichomes, ribbon-like trichomes, and papillae were present. Using fluorescence microscopy and histochemical tests, various substances such as lipids, phenolic compounds, terpenes containing steroids, terpenoids, sesquiterpenes, tannins, and flavonoids were determined but only in the capitate trichomes. The analysis of the essential oil showed that its content in the dry herb of E. stricta was on average 0.257%. Gas chromatography revealed the presence of 28 compounds
Mikromorfologia liści Aesculus hippocastanum L. i uszkodzenia wywołane przez minujące larwy Cameraria ohridella Deschka i Dimić
The present study is a continuation of our research on
the structure of healthy leaves of Aesculus hippocastanum and
leaves damaged by larvae of Cameraria ohridella. In this study,
the epidermal micromorphology of both leaf surfaces in A.
hippocastanum was investigated by scanning electron microscopy
(SEM). Light microscopy was used to examine on which
side of the leaf blades eggs of Cameraria ohridella were laid.
The characteristic features of the adaxial and abaxial epidermis
were shown. In the hypostomatic leaves of the studied species,
the stomata occurred at a density of 173 × mm-2 and they represented
the cyclocytic type. A striated layer of the cuticle
was observed only in the adaxial epidermis in which glandular
hairs were also present along the midribs. Non-glandular trichomes
grew only on the surface of the midribs or in their axils
in the abaxial epidermis. We found eggs of C. ohridella only
on the adaxial surface of the epidermis. Using SEM, we also
observed in the mines leaf tissues damaged by C. ohridella.
We found palisade parenchyma to be absent, whereas in the
spongy parenchyma there could be seen large intercellular spaces
and at places a dense mass of organic matter, formed from
damaged cells and larval excrement. The vascular bundles and
calcium oxalate crystals remained in the feeding places. We
found numerous bacteria, fungal spores and hyphae as well as
cleistothecia of Erysiphales in the mines on the surface of the
damaged mesophyll.
The glandular trichomes occurring only locally on the
leaves, the relatively thin cell walls of the epidermis and a not
well-developed cuticle layer on their surface do not protect sufficiently
these leaves against the invasion of C. ohridella. On
the other hand, damaged tissue areas are a convenient place for
the growth of bacteria and fungi.Obecna praca stanowi kontynuację naszych
badań dotyczących struktury zdrowych liści Aesculus
hippocastanum i uszkodzonych przez larwy Cameraria
ohridella. Analizowano mikromorfologię epidermy obu
powierzchni liści A. hippocastanum w skaningowym
mikroskopie elektronowym. W mikroskopie świetlnym
oceniano, po której stronie blaszek liściowych składane
są jaja Cameraria ohridella. Wykazano charakterystyczne
cechy epidermy doosiowej i odosiowej. W hypostomatycznych
liściach badanego gatunku aparaty szparkowe
występowały w zagęszczeniu 173 × mm-2 i reprezentowały
cyklocytyczny typ.
odosiowej epidermy. Jaja C. ohridella znaleźliśmy
jedynie na powierzchni doosiowej epidermy. W mikroskopie
skaningowym obserwowaliśmy również
uszkodzone przez C. ohridella tkanki liści w obrębie
min. Stwierdziliśmy brak miękiszu palisadowego, zaś
w obrębie miękiszu gąbczastego widoczne były duże
przestwory międzykomórkowe, a miejscami zwarta
masa materii organicznej, która powstała z uszkodzonych
komórek i ekskrementów larw. W żerowiskach
pozostały wiązki przewodzące i kryształy szczawianu
wapnia. W obrębie min, na powierzchni zniszczonego
mezofilu znaleźliśmy liczne bakterie, zarodniki
i strzępki grzybów oraz kleistotecja Erysiphales.
Występujące na liściach tylko lokalnie włoski
gruczołowe, stosunkowo cienkie ściany komórkowe
epidermy oraz niezbyt dobrze rozwinięta warstwa kutykuli
na ich powierzchni nie zabezpieczają w dostateczny
sposób tych liści przed inwazją C. ohridella.
Natomiast obszary uszkodzonych tkanek są dogodnym
miejscem do rozwoju bakterii i grzybów
Zmiany w tkankach liści kasztanowca zwyczajnego (Aesculus hippocastanum L.) zasiedlonych przez szrotówka kasztanowcowiaczka (Cameraria ochridella Deschka and Dimic)
The present study, conducted during the period 2010-
2011, involved morphological observations and anatomical
investigations of horse chestnut (Aesculus hippocastanum L.)
leaves with symptoms of damage caused by feeding of larvae
of the horse-chestnut leaf miner (Cameraria ohridella Deschka&
Dimić). Leaves were collected from trees growing in the
city of Lublin (Poland). Microscopic slides were prepared from
fresh and fixed plant material. Leaf anatomical features were
examined by light microscopy in order to determine the mechanical
barrier for feeding pests. Changes were also observed
during the progressive damage of the leaf tissues caused by the
larvae. Selected developmental stages of the pest are presented
in the paper. It has been shown that very thin blades of the mesomorphic
leaves of Aesculus hippocastanum produce a poorly-
-developed mechanical barrier in which the following elements
can be included: the presence of collenchyma and idioblasts
with druses of calcium oxalate, few non-glandular trichomes
found close to the leaf veins as well as relatively thin outer walls
of the epidermal cells. The cells containing tannins and the oil
cells found in the mesophyll may form a physiological barrier.
However, foraging leaf miner larvae feed only on the palisade
and spongy parenchyma cells, leaving undamaged the cells with
tannins as well as the idioblasts with calcium oxalate crystals
and oils. The feeding of the pest in the leaf mesophyll leads to
the death of the epidermis on both sides of the lamina and to
drying of the parts of the leaves in the area of the mines.W latach 2010-2011 przeprowadzono obserwacje
morfologiczne i badania anatomiczne liści kasztanowca
zwyczajnego (Aesculus hippocastanum L.) z oznakami
uszkodzeń spowodowanymi żerowaniem larw szrotówka
kasztanowcowiaczka (Cameraria ohridella Deschka&
Dimić). Liście pobierano z drzew rosnących na
terenie Lublina (Poland). Preparaty mikroskopowe wykonano
ze świeżego i utrwalonego materiału roślinnego.
W mikroskopie świetlnym analizowano cechy anatomiczne
liścia w celu określenia bariery mechanicznej
dla żerujących szkodników. Obserwowano również
zmiany w czasie postępujących uszkodzeń tkanek liści
przez larwy. Przedstawiono wybrane stadia rozwojowe
szkodnika. Wykazano, że bardzo cienkie blaszki mezomorficznych
liści Aesculus hippocastanum wytwarzają
słabo rozwiniętą barierę mechaniczną, do której
możemy zaliczyć obecność kolenchymy, idioblastów
z druzami szczawianu wapnia, nieliczne włoski mechaniczne
występujące przy nerwach oraz stosunkowo
cienkie ściany zewnętrzne komórek epidermy. Barierę
fizjologiczną mogą tworzyć komórki zawierające
garbniki oraz występujące w mezofilu komórki olejkowe.
Jednakże żerujące larwy szrotówka żywią się tylko
komórkami parenchymy palisadowej i gąbczastej,
pozostawiając nieuszkodzone komórki z garbnikami
oraz idioblasty z kryształami szczawianu wapnia oraz
olejkami. Postępujące żerowanie szkodnika w mezofilu
liścia doprowadza do obumierania epidermy po obu
stronach blaszki liściowej i zasychania fragmentów liści
w obrębie min
Morphological differentiation of non-glandular and glandular trichomes on Marrubium vulgare L.
Marrubium vulgare L., commonly known as a white horehound or common horehound, belongs to the plant family Lamiaceae. It is a perennial aromatic herb which grows naturally in Europe, Asia, and America. Since ancient Egypt, this species has been known as a remedy for upper respiratory tract ailments. Nowadays, horehound is used in herbal medicine for treatment of liver diseases, biliary tract disorders, and for increasing the appetite and supporting the function of the stomach. The main biologically active substances in M. vulgare organs are: marrubiin, tannins, essential oils, and ursolic acid. The paper presents micromorphological analyses of non-glandular and glandular trichomes of M. vulgare. The research material was sampled from the plant collection in the Botanical Garden of the Maria Curie-Sklodowska University in Lublin (51°14′ N, 22°34′ E). The above-ground parts of horehound were collected during the flowering period in July 2013. Using light microscopy (LM) and scanning electron microscopy (SEM), the types and sizes of trichomes from the stem, leaf, calyx, and corolla were investigated. The results of the microscopic observations show that the surfaces of M. vulgare vegetative and reproductive organs are densely clothed with glandular and non-glandular trichomes. The glandular trichomes are of two main types: peltate and capitate. Peltate trichomes consist of a short stalk cell and a large head with secretory cells arranged in a circle. The height of a mature trichome is about 31.33 μm and the diameter of the head is 31.47 μm. The substance produced by secretory cells passes through the apical walls and accumulates within a space between the cuticle and the cell wall layer. Capitate long trichomes with a basal cell, long stalk, neck cell, and a unicellular head are 36.65 μm long and the diameter of the head is about 15.6 μm. There are two types of short capitate trichomes: with a bicellular head and a unicellular stalk and with four-celled head and a unicellular stalk. The length of these trichomes is on average 26.32 μm and the diameter of the head is 20.32 μm. Moreover, we observed two types of non-glandular trichomes: multicellular uniserrate and multicellular branched. The length of the non-glandular trichomes is between 60.69 μm and 1130.3 μm
Cechy ekologiczne kwiatów i kwiatostanów dwóch gatunków z rodzaju Petasites Miller (Asteraceae)
The structural features of flowers and inflorescences of
Petasites hybridus and P. albus were compared. Only individuals
producing flower heads with male flowers and few female
flowers were found in the studied populations. Light microscopy
(LM) and scanning electron microscopy (SEM) were used
for examination.
The present study shows that the stems of the above-
-mentioned species differed in height and number of flower heads,
but the number of flowers per head was similar. Larger flowers
were found on the stems of P. albus. The following features
has been found to play an important role in pollination ecology:
the strongly contrasting colours of the floral parts; on the petals,
the occurrence of several types of cells which can increase the
attractiveness of the flowers by refracting sunlight in a different
way; production of odorous oils by the petal cells; production of
significant amounts of pollen offered to insects by the well-developed
pollen presenters; the development of nectaries and nectar
production by the male flowers as well as the development of
colour attractants by the corolla, anthers, and bracts.Porównywano cechy budowy kwiatostanów
i kwiatów Petasites hybridus i P. albus. W badanych
populacjach znaleziono tylko osobniki wytwarzające
koszyczki z kwiatami męskimi i nielicznymi żeńskimi.
Do badań wykorzystano mikroskopię świetlną (LM)
i skaningową elektronową (SEM).
Z badań wynika, że pędy wymienionych gatunków
różniły się wysokością i liczbą koszyczków, ale
liczba kwiatów w koszyczkach była zbliżona. Większe
kwiaty znaleziono na pędach P. albus. Do cech odgrywających
znaczną rolę w ekologii zapylania zaliczono:
silnie skontrastowane barwy elementów kwiatowych;
występowanie na płatkach kilku typów komórek, które
mogą zwiększać atrakcyjność kwiatów przez załamywanie
w różny sposób promieni świetlnych; wytwarzanie
olejków zapachowych przez komórki płatków;
produkcję znacznych ilości pyłku oferowanego owadom
przez dobrze rozwinięte prezentery pyłkowe; wytwarzanie
nektarników i nektaru przez kwiaty męskie
oraz utworzenie barwnych atraktantów sygnalizacyjnych
przez koronę, pylniki i liście przykwiatowe
Zakres wahań stężenia zarodników konidialnych Alternaria w powietrzu z różnych lokalizacji geograficznych w Polsce (2010–2011)
Knowledge of seasonal and diurnal fluctuations of allergenic
and phytopathogenic fungal spores in the air and the
determination of the influence of weather and environmental
parameters on spore concentrations are of great practical use
in control of both human and animal health as well as plant diseases.
The aim of this study was to evaluate the dynamics of the
occurrence of Alternaria spores in the air at three sites located
in different geographical regions of Poland. The study was done
from 15 April to 30 September in 2010–2011 in Rzeszów (Carpathian
Foothills, south-east Poland), Lublin (Lublin Upland,
central east), and Poznań (central west); the distance between
the experimental sites ranged from 138 to 441 km. To assess
the concentration of Alternaria spores, the volumetric method
was used. In both years, the highest concentration of spores was
observed in Poznań and the lowest in Rzeszów. In 2010 the
annual total, monthly total and maximum spore concentration
were higher than in 2011 at all monitoring sites. The greatest
concentration of Alternaria spores was recorded in August, followed
by July. High spore concentrations were also noted at
the beginning of September. The differences in daily concentrations,
average concentrations and cumulative numbers of spores
were statistically significant. The reasons for this can be related
not only to weather parameters, which is often raised in the literature,
but also to the type of landscape. Landscape and geobotanical
conditions of Great Poland and Carpathian Foothills
greatly differ. In both years, the highest number of Alternaria
spores was found in Poznań – the city with the highest urbanization
factor and the capital of the region with large-scale farming,
connected with the intensification of agricultural practices.Znajomość sezonowej i dobowej rytmiki występowania
alergennych i patogennych zarodników
grzybów w powietrzu oraz określenie wpływu elementów
środowiska na wartości stężeń mogą być wykorzystane
w praktyce, w alergologii i w rolnictwie.
Celem badań było określenie dynamiki występowania
zarodników Alternaria w powietrzu trzech stanowisk
zlokalizowanych w różnych regionach geograficznych
Polski. Badania prowadzono od 15 kwietnia do
30 września w latach 2010–2011 na terenie Rzeszowa,
Lublina i Poznania; odległości pomiędzy punktami
badawczymi wynosiły od 138 do 441 km. Do oceny
stężenia zarodników rodzaju Alternaria zastosowano
metodę wolumetryczną. W obu latach najwyższe
stężenia zarodników odnotowano w Poznaniu, a najniższe
w Rzeszowie. Na wszystkich stanowiskach
w 2010 roku miesięczne i roczne sumy zarodników,
jak też ich maksymalne stężenie było wyższe niż
w 2011 roku. Stężenie zarodników Alternaria było
największe w sierpniu i niewiele niższe w lipcu. Wysokie
stężenia odnotowano także na początku września.
Różnice w dobowych stężeniach, średnich stężeniach
i sumach w kolejnych miesiącach w okresie
dwóch lat badań były statystycznie istotne. Przyczyny
lecz bardziej zbliżony do Wielkopolski. W obu latach
największą liczbę zarodników grzybów rodzaju
Alternaria stwierdzono w Poznaniu – mieście o największym
współczynniku urbanizacji oraz stolicy regionu
z rolnictwem wielkoobszarowym, związanym
z intensyfikacją zabiegów agrotechnicznych
