    The role of glucagon-like peptide-1 on food intake, glucokinase expression and fatty acid metabolism

    The increase in blood levels of GLP-1 with dietary resistant starch is thought to be associated with the activities of hypothalamic NPY/AgRP neurons and the gene expression of glucokinase in the ARC of the hypothalamus. Exendin-4 has been shown to be associated with fatty acid synthesis and oxidation. In this project, it was proposed that 1) food intake will be decreased in GLP-1R KO mice, the glucokinase (GK) mRNA expression in the liver and in the hypothalamus, the hypothalamic NPY mRNA expression will be up-regulated by exendin-4 treatment; and 2) exendin-4 will decrease fatty acid synthesis and increase fatty acid oxidation. In order to investigate these hypotheses, animal experiments were performed. Exendin-4 treated WT and GLP-1R KO mice were injected with exendin-4 at 100”g/Kg body weight or vehicle for 7 days. The exendin-4 treated mice and the vehicle mice were subdivided into fed and fasted groups. There were 8 groups of mice (n=8). Food intake after refeeding at 1h, 2h and 4h was measured. At end of the study, the blood glucose level, the GK, CPT1, AOX1, SCD1, FAS, SREBP1c and PPARã mRNA expressions in liver, and the GK and NPY mRNA expressions in hypothalamus were measured. In this study, the food intake of GLP-1R KO mice at 1h, 2h and 4h was significantly less than in WT mice, and the FAS expression in liver was significantly higher in GLP-1R KO mice than WT mice. Exendin-4 treatment decreased blood glucose, and increased the GK gene expression in liver, but not in the hypothalamus. The food intake and the NPY gene expression were not altered by exendin-4. Only the AOX mRNA expression was increased by exendin-4. CPT1, SCD1, FAS, SREBP1c, and PPARã mRNA were not changed. GLP-1 might have beneficial effects on improving the impairment of nutrient sensing system induced by aging. The up-regulated FAS gene expression might be one possible mechanism for higher body fat in GLP1R KO mice.     The Short Text Matching Model Enhanced with Knowledge via Contrastive Learning

    In recent years, short Text Matching tasks have been widely applied in the fields ofadvertising search and recommendation. The difficulty lies in the lack of semantic information and word ambiguity caused by the short length of the text. Previous works have introduced complement sentences or knowledge bases to provide additional feature information. However, these methods have not fully interacted between the original sentence and the complement sentence, and have not considered the noise issue that may arise from the introduction of external knowledge bases. Therefore, this paper proposes a short Text Matching model that combines contrastive learning and external knowledge. The model uses a generative model to generate corresponding complement sentences and uses the contrastive learning method to guide the model to obtain more semantically meaningful encoding of the original sentence. In addition, to avoid noise, we use keywords as the main semantics of the original sentence to retrieve corresponding knowledge words in the knowledge base, and construct a knowledge graph. The graph encoding model is used to integrate the knowledge base information into the model. Our designed model achieves state-of-the-art performance on two publicly available Chinese Text Matching datasets, demonstrating the effectiveness of our model.Comment: 11 pages,2 figure

    Synthesis and physical properties of Ce2_2Rh3+ÎŽ_{3+\delta}Sb4_4 single crystals

    Millimeter-sized Ce2_2Rh3+ÎŽ_{3+\delta}Sb4_4 (ή≈1/8\delta\approx 1/8) single crystals were synthesized by a Bi-flux method and their physical properties were studied by a combination of electrical transport, magnetic and thermodynamic measurements. The resistivity anisotropy ρa,b/ρc∌2\rho_{a,b}/\rho_{c}\sim2, manifesting a quasi-one-dimensional electronic character. Magnetic susceptibility measurements confirm ab\mathbf{ab} as the magnetic easy plane. A long-range antiferromagnetic transition occurs at TN=1.4T_N=1.4 K, while clear short-range ordering can be detected well above TNT_N. The low ordering temperature is ascribed to the large Ce-Ce distance as well as the geometric frustration. Kondo scale is estimated to be about 2.4 K, comparable to the strength of magnetic exchange. Ce2_2Rh3+ÎŽ_{3+\delta}Sb4_4, therefore, represents a rare example of dense Kondo lattice whose Ruderman-Kittel-Kasuya-Yosida exchange and Kondo coupling are both weak but competing.Comment: 7 pages, 4 figures, 2 table

    Does Incident Solar Ultraviolet Radiation Lower Blood Pressure?

    Background Hypertension remains a leading global cause for premature death and disease. Most treatment guidelines emphasize the importance of risk factors, but not all are known, modifiable, or easily avoided. Population blood pressure correlates with latitude and is lower in summer than winter. Seasonal variations in sunlight exposure account for these differences, with temperature believed to be the main contributor. Recent research indicates that UV light enhances nitric oxide availability by mobilizing storage forms in the skin, suggesting incident solar UV radiation may lower blood pressure. We tested this hypothesis by exploring the association between environmental UV exposure and systolic blood pressure (SBP) in a large cohort of chronic hemodialysis patients in whom SBP is determined regularly. Methods and Results We studied 342 457 patients (36% black, 64% white) at 2178 US dialysis centers over 3 years. Incident UV radiation and temperature data for each clinic location were retrieved from the National Oceanic and Atmospheric Administration database. Linear mixed effects models with adjustment for ambient temperature, sex/age, body mass index, serum Na+/K+ and other covariates were fitted to each location and combined estimates of associations calculated using the DerSimonian and Laird procedure. Pre-dialysis SBP varied by season and was ≈4 mm Hg higher in black patients. Temperature, UVA and UVB were all linearly and inversely associated with SBP. This relationship remained statistically significant after correcting for temperature. Conclusions In hemodialysis patients, in addition to environmental temperature, incident solar UV radiation is associated with lower SBP. This raises the possibility that insufficient sunlight is a new risk factor for hypertension, perhaps even in the general population.</p

    Logistic Regression Analysis of Factors Influencing the Effectiveness of Intensive Sound Masking Therapy in Patients with Tinnitus

    Objectives: To investigate factors influencing the effectiveness intensive sound masking therapy on tinnitus using Logistic Regression Analysis. Design: The study used a retrospective cross-section analysis. Participants: 102 patients with tinnitus were recruited at the Sun Yat-sen Memorial Hospital of Sun Yat-sen University, China. Intervention: Intensive sound masking therapy was used as an intervention approach for patients with tinnitus. Primary and secondary outcome measures: participants underwent audiological investigations and tinnitus pitch and loudness matching measurements, followed by intensive sound masking therapy. The Tinnitus Handicap Inventory (THI) was used as the outcome measure pre- and post-treatment. Multivariate logistic regression was performed to investigate the association of demographic and audiological factors with effective therapy. Results: According to the THI score changes pre-and post-sound masking intervention, fifty-one participants were categorised into an effective group, the remaining 51 participants were placed in a non-effective group. Those in the effective group were significantly younger than those in the non-effective group (p=0.012). Significantly more participants had flat audiogram configurations in the effective group (p=0.04). Multivariable logistic regression analysis showed that age (OR=0.96, 95% CI: 0.93, 0.99, p=0.007), audiometric configuration (p=0.027) and THI score pre-treatment (OR=1.04, 95% CI: 1.02, 1.07, p<0.001) were significantly associated with therapeutic effectiveness. Further analysis showed that patients with flat audiometric configurations were 5.45 times more likely to respond to intervention than those with high-frequency steeply sloping audiograms (OR=5.45, 95% CI: 1.67, 17.86, p=0.005). Conclusion: Audiometric configuration, age and THI scores appear to be predictive for the effectiveness of sound masking treatment. Gender, tinnitus characteristics and hearing threshold measures seem not to be related to treatment effectiveness. Further randomized control study is needed to provide further evidence of the effectiveness of prognostic factors in tinnitus interventions

    Association of recent exposure to ambient metals on fractional exhaled nitric oxide in 9–11 year old inner-city children

    Exposure to ambient metals in urban environments has been associated with wheeze, and emergency room visits and hospitalizations due to respiratory illness. However, the effect of ambient metals exposure on airway inflammation, and how these associations may be modified by seroatopy, has not been determined. Fractional exhaled nitric oxide (FENO) is a reliable proxy marker of airway inflammation. We hypothesized that recent ambient concentrations of Ni, V, Zn and Fe would be associated differentially with proximal and distal fractions of exhaled NO, and that these associations would be modified by seroatopy. As part of the Columbia Center for Children’s Environmental Health (CCCEH) birth cohort study, 9–11 year old children (n = 192) were evaluated. Ambient measures of Ni, V, Zn and Fe were obtained from a local central monitoring site and averaged over 9 days based on three 24 h measures every third day. Fractional exhaled nitric oxide (FENO) samples were obtained at constant flows of 50 (FENO50), 83 and 100 mL/s, and used to determine surrogate measures for proximal (JNO) and alveolar (Calv) inflammation. Seroatopy was determined by specific IgE at age 7. Data were analyzed using multivariable linear regression. Ambient V and Fe concentrations were associated positively with FENO50 (p = 0.018, p = 0.027). Ambient Fe was associated positively with JNO (p = 0.017). Ambient Ni and V concentrations were associated positively with Calv (p = 0.004, p = 0.018, respectively). A stronger association of Ni concentrations with Calv was observed among the children with seroatopy. These results suggest that ambient metals are associated differentially with different fractions of FENO production, and this relationship may be modified by seroatopy
