    The homozygosity verification for doubled haploid Japanese flounder, Paralichthys olivaceus, by microsatellite DNA markers

    A total of 845 doubled haploids Japanese flounder, Paralichthys olivaceus from three spawns were produced by utilizing hydrostatic pressure treatments on eggs fertilized with ultraviolet (UV)-irradiated sperm of red sea bream (Pagrus major). 481 polymorphic microsatellite markers were used to identify the homozygosity of these doubled haploids, of which, only 31% (265 out of 845) individuals were fully homozygous at all loci tested, while, the rest 580 offspring were homozygous at 183 to 320 loci. Additionally, the embryonic development and external morphology of doubled haploids were also observed and recorded. The whole process of embryonic development was divided into 19 stages based on the morphological characteristics of the developing embryo. Hatching took place 73 to 74 h after fertilization. The means of body weight, body length and body depth in 360 days old fishes were always small in doubled haploids and large in normal diploids, but, the standard deviations and coefficients of variation of doubled haploids were significantly higher than those of normal diploids.Keywords: Doubled haploids, gynogenesis, Japanese flounder, Paralichthys olivaceu

    Analysis of cognitive function and its related factors after treatment in Meniere’s disease

    A growing body of research recently suggested the association between vestibular dysfunction and cognitive impairment. Meniere’s disease (MD), a common clinical vestibular disorder, is usually accompanied by hearing loss and emotional stress, both of which may mediate the relationship between vestibule dysfunction and cognition. It is currently unknown whether the cognitive decline in MD patients could improve through treatment and how it relates to multiple clinical characteristics, particularly the severity of vertigo. Therefore, in the present study, the MD patients were followed up for 3, 6, and 12 months after treatment, and the cognitive functions, vertigo symptoms, and related physical, functional, and emotional effects of the patients were assessed using the Montreal Cognitive Assessment (MoCA) and Dizziness Handicap Inventory (DHI), aiming to explore the change in cognition before and after therapy and the correlation with various clinical features. It was found that cognitive decline in MD patients compared to healthy controls before therapy. Importantly, this cognitive impairment could improve after effective therapy, which was related to the severity of vertigo, especially in functional and physical impacts. Our results support the view that vestibular dysfunction is a potentially modifiable risk factor for cognitive decline

    Assimilation of Typhoon Wind Field Retrieved from Scatterometer and SAR Based on the Huber Norm Quality Control

    Observations of sea surface wind field are critical for typhoon prediction. The scatterometer observation is one of the most important sources of sea surface winds, which provides both wind speed and wind direction information. However, the spatial resolution of scatterometer wind is low. Synthetic Aperture Radar (SAR) can provide a more detailed wind structure of the tropical cyclone. In addition, the cross-polarization observation of SAR can provide more detailed information of high speed wind (>25 m·s − 1 ) than the scatterometer. Nevertheless, due to the narrow swath of SAR, the number of retrieved sea surface wind data used in the data assimilation is limited, and another limitation of SAR wind observation is that it does not provide true wind direction information. In this paper, the joint assimilation of the Advanced Scatterometer (ASCAT) wind and Sentinel-1 SAR wind was investigated. Another limitation in the current operational typhoon prediction is the inefficient quality control (QC) method used in the data assimilation since a large number of high speed wind observations was rejected by the traditional Gaussian distribution QC. We introduce the Huber norm distribution quality control (QC) into the data assimilation successfully. A numerical simulation experiment of typhoon by Lionrock (2016) is conducted to test the proposed method. The experimental results showed that the new quality control scheme not only greatly increases the availability of wind data in the area of the typhoon center, but also improves the typhoon track prediction, as well as the intensity prediction. The joint assimilation of scatterometer and SAR winds does have a positive impact on the typhoon prediction

    Abstract Background Circular RNAs (CircRNAs) have been associated with acute lung injury (ALI), but their molecular mechanisms remain unclear. Methods This study developed a rat model of lipopolysaccharide (LPS)-induced ALI and evaluated the modeling effect by hematoxylin and eosin staining, Masson’s trichrome staining, lung wet-to-dry weight ratio, terminal deoxynucleotidyl transferase UTP nick end labeling (TUNEL), and enzyme-linked immunosorbent assay (ELISA) detection of inflammatory factors (interleukin-1β, tumor necrosis factor alpha, and interleukin-6). Using lung tissues from a rat model of LPS-induced ALI, we then conducted circRNA sequencing, mRNA sequencing, and bioinformatics analysis to obtain differential circRNA and mRNA expression profiles as well as potential ceRNA networks. Furthermore, we performed quantitative real-time polymerase chain reaction (qRT-PCR) assays to screen for circ-Phkb in ALI rat lung tissues, alveolar macrophages, and LPS-induced NR8383 cells. We conducted induction with or without LPS with circ-Phkb siRNA and overexpression lentivirus in NR8383. Cell Counting Kit-8, C5-Ethynyl-2′-deoxyuridine (Edu), TUNEL, and cytometry were used to identify proliferation and apoptosis, respectively. We detected inflammatory factors using ELISA. Finally, we used Western blot to detect the apoptosis-related proteins and TLR4/MyD88/NF-kB/CCL2 pathway activation. Results Our results revealed that both circRNA and mRNA profiles are different from those of the Sham group. We observed a significant circ-Phkb upregulation in NR8383 cells and LPS-exposed rats. Apoptosis and inflammation were greatly reduced when circ-Phkb expression was reduced in NR8383 cells, cell proliferation was increased, and circ-Phkb overexpression was decreased. Conclusions In terms of mechanism, circ-Phkb suppression inhibits CCL2 expression via the TLR4/MyD88/NF-kB pathway in LPS-induced alveolar macrophages

    A Fault-Tolerant Strategy for Three-Level Flying-Capacitor DC/DC Converter in Spacecraft Power System

    With the development of space exploration, high-power and high-voltage power systems are essential for future spacecraft applications. Because of the effects of space radiation such as single event burnout (SEB), the rated voltage of power devices in converters for a spacecraft power system is limited to a level much lower than that for traditional ground applications. Thus, multi-level DC/DC converters are good choices for high-voltage applications in spacecraft. In this paper, a fault-tolerant strategy is proposed for a three-level flying capacitor DC/DC converter to increase the reliability with minimal cost. There is no extra hardware needed for the proposed strategy; the fault tolerance of the converter is only achieved by changing the software control strategy. A stage analysis of the proposed strategy is provided in detail for different fault locations and ratios between the input and output voltage. Finally, a simulation model and prototype are built to verify the effectiveness of the proposed strategy

    Tropospheric NO2 Pollution Monitoring with the GF-5 Satellite Environmental Trace Gases Monitoring Instrument over the North China Plain during Winter 2018–2019

    The Environmental Trace Gases Monitoring Instrument (EMI) is a high-spectral-resolution payload onboard the latest pathfinder mission GaoFen-5, designed specifically for the monitoring of global atmospheric trace gas compositions and trends. This study describes a comparative analysis of the tropospheric nitrogen dioxide (NO2) columns over the North China Plain (NCP) from November 2018 to April 2019 based on EMI products. Validation of satellite products based on a cross-correlation analysis with data from four ground-based multi-axis differential optical absorption spectroscopy sites provided good correlation coefficients (r) ranging from 0.78 to 0.88. The distribution and monthly averaged tropospheric NO2 columns revealed high pollution exposure levels during winter (November–January) and a decrease from February onward in the NCP. Moreover, a typical pollution event was analyzed in detail in combination with wind field statistics. The results indicated that variations of NO2 concentrations in Beijing and Tianjin were highly correlated with the wind direction from 22.5–45.0 degrees west of south, especially during times of high NO2 amounts. These findings highlight that the EMI payload on the GaoFen-5 (GF-5) satellite is useful for remote sensing of regional and global NO2 detection

    Modified intratympanic steroid therapy for sudden sensorineural hearing loss via tympanic tube and gelfoam as a salvage treatment

    Background: Sudden sensorineural hearing loss (SSNHL) is a prevalent emergency in ear, nose, and throat practice. Previous studies have demonstrated that intratympanic steroid therapy (IST) can serve as a salvage treatment for SSNHL after the failure of systemic steroid therapy (SST). Objective: This study aimed to analyze the efficacy of modified IST involving the insertion of a tympanic tube and gelfoam as a salvage treatment for patients with SSNHL, and to explore its associated factors. Methods: Totally, 74 patients who were aged 22–81 years with SSNHL were enrolled and allocated to either the control group (n = 25) or the treatment group (n = 49) based on their treatment modalities. All patients received SST lasting for at least 7 days. Subsequently, patients in the treatment group, after SST failure, underwent IST twice a week for 2–6 weeks, while the control group did not. Efficacy was assessed by the improvement in pure tone average at the affected frequency at the beginning and end of IST. Results: Hearing improvement in all patients after IST in the treatment group was 9.71 ± 14.84 dB, with significant improvement at affected frequencies (250-8000 Hz) compared with the control group (P  0.05). Conclusion: The modified IST was demonstrated to be a safe and effective method as a salvage treatment for SSNHL. This study explored the efficacy of a modified IST approach, incorporating the utilization of tympanic tubes and gelfoam as key components. The findings underscore the advantages of gelfoam as a strategic drug carrier placed in the round window niche. By minimizing drug loss, extending action time, and increasing perilymph concentration, gelfoam enhances the therapeutic impact of IST, contributing to improved hearing outcomes in patients with SSNHL
