    Real-time and interactive tools for vocal training based on an analytic signal with a cosine series envelope

    We introduce real-time and interactive tools for assisting vocal training. In this presentation, we demonstrate mainly a tool based on real-time visualizer of fundamental frequency candidates to provide information-rich feedback to learners. The visualizer uses an efficient algorithm using analytic signals for deriving phase-based attributes. We start using these tools in vocal training for assisting learners to acquire the awareness of appropriate vocalization. The first author made the MATLAB implementation of the tools open-source.     Safety and efficacy of pirfenidone in idiopathic pulmonary fibrosis in clinical practice

    SummaryBackgroudPrevious pirfenidone trials have only involved patients with mild-to-moderate idiopathic pulmonary fibrosis (IPF). The aim of this study was to investigate the safety and efficacy of pirfenidone in patients with mild-to-severe IPF in clinical practice.MethodsThe clinical records of 76 patients who were diagnosed with IPF and received pirfenidone were reviewed.ResultsThe most frequent adverse event was anorexia, although the grade of anorexia in most patients was mild. Dose reduction of pirfenidone improved anorexia in 84% affected patients, which resulted in a high medication compliance rate. The mean forced vital capacity (FVC) at the initiation of pirfenidone therapy in this study was approximately 10% lower than that in previous clinical trials. The mean change in FVC during the 6-month period prior to the therapy initiation was −188 mL, which improved to −19 mL during the 6-month period after therapy. Significant attenuation in percentage predicted diffusion capacity of the lung for carbon monoxide decline was also achieved after pirfenidone therapy initiation. The efficacy of pirfenidone in attenuating the degree of FVC decline was higher in the group with FVC decline of ≥150 mL during the 6-month period prior to therapy initiation. The levels of serum markers, such as KL-6 and SP-D, were also lowered by the therapy.ConclusionsThese results showed that pirfenidone was well-tolerated and had beneficial effects in patients with mild-to-severe and/or progressive IPF. The degree of disease progression prior to the initiation of pirfenidone therapy had an impact on the response to the therapy

    The uncoating of EV71 in mature late endosomes requires CD-M6PR

    Enterovirus 71 (EV71) is one of the causative agents of hand-foot-and-mouth disease, which in some circumstances could lead to severe neurological diseases. Despite of its importance for human health, little is known about the early stages of EV71 infection. EV71 starts uncoating with its receptor, human scavenger receptor B2 (hSCARB2), at low pH. We show that EV71 was not targeted to lysosomes in human rhabdomyosarcoma cells overexpressing hSCARB2 and that the autophagic pathway is not essential for EV71 productive uncoating. Instead, EV71 was efficiently uncoated 30 minutes after infection in late endosomes (LEs) containing hSCARB2, mannose-6-phosphate receptor (M6PR), RAB9, bis(monoacylglycero)phosphate and lysosomal associated membrane protein 2 (LAMP2). Furthering the notion that mature LEs are crucial for EV71 uncoating, cation-dependent (CD)-M6PR knockdown impairs EV71 infection. Since hSCARB2 interacts with cation-independent (CI)-M6PR through M6P-binding sites and CD-M6PR also harbor a M6P-binding site, CD-M6PR is likely to play important roles in EV71 uncoating in LEs

    A humanized mouse model identifies key amino acids for low immunogenicity of H7N9 vaccines

    Influenza vaccines of H7N9 subtype are consistently less immunogenic in humans than vaccines developed for other subtypes. Although prior immunoinformatic analysis identified T-cell epitopes in H7 hemagglutinin (HA) which potentially enhance regulatory T cell response due to conservation with the human genome, the links between the T-cell epitopes and low immunogenicity of H7 HA remains unknown due to the lack of animal models reproducing the response observed in humans. Here, we utilized a humanized mouse model to recapitulate the low immunogenicity of H7 HA. Our analysis demonstrated that modification of a single H7 epitope by changing 3 amino acids so that it is homologous with a known H3 immunogenic epitope sequence significantly improved the immunogenicity of the H7 HA in the humanized mouse model, leading to a greater than 4-fold increase in HA-binding IgG responses. Thus, we provide experimental evidence for the important contribution of this H7-specific T cell epitope in determining the immunogenicity of an influenza vaccine. Furthermore, this study delineates strategies that can be used for screening and selecting vaccine strains using immunoinformatics tools and a humanized mouse model

    Relationship of flow-volume curve pattern on pulmonary function test with clinical and radiological features in idiopathic pulmonary fibrosis.

    Background:The flow-volume (FV) curve pattern in the pulmonary function test (PFT) for obstructive lung diseases is widely recognized. However, there are few reports on FV curve pattern in idiopathic pulmonary fibrosis (IPF). In this study, we investigated the relationship between FV curve pattern and clinical or radiological features in IPF.Methods:The FV curves on PFTs and chest high-resolution computed tomography (HRCT) images of 130 patients with IPF were retrospectively evaluated. The FV curves were divided into four groups based on the presence or absence of the convex and concave patterns: convex/concave, non-convex/concave, convex/non-concave, and non-convex/non-concave. Using a computer-aided system, CT honeycombing area (%HA) and subtracted low attenuation area (%sLAA) were quantitatively measured. To assess the distribution of CT findings, the lung area was divided into upper, lower, central, and peripheral areas. The relationships of FV curve patterns with patient characteristics, spirometry results, and quantitative CT findings were evaluated.Results:The patients with convex pattern was identified in 93 (71.5%) and concave pattern in 72 (55.4%). Among the four groups, patients with the convex/non-concave pattern had significantly lower forced vital capacity (FVC) and higher %HA of the upper/peripheral lung area (p = 0.018, and p = 0.005, respectively). The convex/non-concave pattern was a significant predictor of mortality for IPF (hazard ratio, 2.19; p = 0.032).Conclusions:Patients with convex/non-concave pattern in FV curve have lower FVC and poorer prognosis with distinct distribution of fibrosis. Hence, FV curve pattern might be a useful predictor of mortality in IPF

    Probing the MSSM Higgs Boson Sector with Explicit CP Violation through Third Generation Fermion Pair Production at Muon Colliders

    We perform a systematic study of the production of a third-generation fermion-pair, μ+μffˉ\mu^+\mu^-\to f\bar{f} for f=τ,bf=\tau^-,b, and t in the minimal supersymmetric standard model (MSSM) with explicit CP violation, which is induced radiatively by soft trilinear interactions related to squarks of the third generation. We classify all the observables for probing the CP property of the Higgs bosons constructed by the initial muon beam polarization along with the unpolarized final fermions and with the final-fermion polarization configuration of equal helicity, respectively. The observables allow for complete determination of CP property of the neutral Higgs bosons. The interference between the Higgs boson and gauge boson contributions also could provide a powerful method for the determination of the CP property of two heavy Higgs bosons in the top-quark pair production near the energy region of the Higgs-boson resonances. For the lightest Higgs-boson mass there is no sizable interference between scalar and vector contributions for the determination of the CP property of the lightest Higgs boson. We give a detailed numerical analysis to show how the radiatively-induced CP violation in the Higgs sector of the MSSM can be measured.Comment: 30 pages, 7 figures including 5 eps ones.
