    A49T, V89L and TA repeat polymorphisms of steroid 5α-reductase type II and breast cancer risk in Japanese women

    BACKGROUND: Breast cancer is hormone related, as are cancers of the endometrium, ovary, and prostate. Several studies have suggested that higher extracellular levels of androgens are associated with breast cancer risk, while biological evidence indicates that androgens are protective. The codon 49 alanine to threonine substitution (A49T), codon 89 valine to leucine substitution (V89L) and TA repeat polymorphisms of the steroid 5α-reductase type II (SRD5A2) gene are considered functional with respect to enzyme activity converting testosterone into dihydrotestosterone. To test the hypothesis that these three polymorphisms are associated with risk of breast cancer, a case–control study was conducted with patients of Aichi Cancer Center Hospital. METHODS: The cases were 237 patients histologically diagnosed with breast cancer, and the controls were 185 noncancer outpatients. DNA from peripheral blood was genotyped by PCR methods. RESULTS: The threonine allele of A49T was not found in our subjects. Compared with the V/V genotype of V89L, the L/L genotype was associated with a decreased risk (crude odds ratio [OR] = 0.61, 95% confidence interval [CI] = 0.36–1.05). This was also the case for the TA(9/9) genotype, with an OR of 0.58 (95% CI = 0.13–2.63) relative to TA(0/0). Among women with the TA(0/0) genotype, however, the OR for the L/L genotype was 0.46 (95% CI = 0.24–0.88) compared with the V/V genotype, and those with the V/V and TA(0/0) genotypes had the highest risk. The haplotype with the L and TA(9) repeat alleles was not found. CONCLUSION: This study is the first to our knowledge focusing on Japanese women, suggesting that SRD5A2 polymorphisms might have an association with breast cancer risk. Further large-sample studies will be required to confirm the association and to assess any interactions with environmental factors

    Expression of leptin receptors in hepatic sinusoidal cells

    Emerging evidence has suggested a critical role of leptin in hepatic inflammation and fibrogenesis, however, the precise mechanisms underlying the profibrogenic action of leptin in the liver has not been well elucidated. Therefore, the present study was designed to investigate the expression and functions of leptin receptors (Ob-R) in hepatic sinusoidal cells. Hepatic stellate cells (HSCs), Kupffer cells and sinusoidal endothelial cells (SECs) were isolated from rat livers by in situ collagenase perfusion followed by differential centrifugation technique, and expression of Ob-Ra and Ob-Rb, short and long Ob-R isoforms, respectively, were analyzed by RT-PCR. Ob-Ra mRNA was detected ubiquitously in HSCs and SECs. In contrast, Ob-Rb was detected clearly only in SECs and Kupffer cells, but not in 7-day cultured HSCs. Indeed, tyrosine-phosphorylation of STAT-3, a downstream event of Ob-Rb signaling, was observed in SECs, but not in HSCs, 1 hr after incubation with leptin. Further, leptin increased AP-1 DNA binding activity and TGF-beta 1 mRNA levels in Kupffer cells and SECs, whereas leptin failed to increase TGF-beta 1 mRNA in HSCs. These findings indicated that SECs and Kupffer cells, but not HSCs, express functional leptin receptors, through which leptin elicits production of TGF-beta 1. It is hypothesized therefore that leptin, produced systemically from adipocytes and locally from HSCs, up-regulates TGF-beta 1 thereby facilitate tissue repairing and fibrogenesis in the sinusoidal microenvironment

    Useful Extend-release Chitosan Tablets with High Antioxidant Activity.

    The antioxidant properties of different low molecular weight (LMW) chitosans (CS1; 22 kDa, CS2; 38 kDa, CS3; 52 kDa, CS4; 81 kDa) were examined for possible use in extended-release tablets. The criteria used were the ability of the chitosans to reduce Cu2+, and hydroxyl and superoxide radicals and N-centered radicals derived from 1,1\u27-diphenyl-2-picrylhydrazyl, via the use of ESR spectrometry. CS2 showed the highest scavenging activity. CS1 and CS3, however, were much less effective and CS4 was not a viable antioxidant. The results suggest that CS2 could be useful in combating the development of oxidative stress. A series of chitosan tablets were prepared using a spray drying method and evaluated as an extended-release matrix tablet using theophylline (TPH) as a model drug. The release of TPH from the different MW chitosan tablets increased with increasing MW of the chitosan used. CS2, CS3 and CS4 showed a reasonable release activity, but CS1 showed the shortest release activity. Moreover, the CS2-TPH tablet showed the highest scavenging activity of the three chitosan tablets (CS2-CS4) using 2,2\u27-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) radicals.     死別体験における悲嘆の回復過程に関する要因の分析

    スマトラ沖大地震・インド洋津波によって,突然肉親と死別体験をもった2遺族に面接調査を行った.死別による悲嘆から回復する過程に,どのような要因が関連しているのかについて分析した.その結果,この地域が敬虔な仏教徒であったことから,対象となった2遺族は,信仰が悲しみを緩和する要因として働いていた.さらに近隣の人びとの精神的な支援や,新たな生命の誕生は将来への希望につながり,悲嘆から回復する要因の一つとなっていることが明らかとなった.Two families that experienced the sudden death of relatives in the Sumatra earthquake and Indian Ocean tsunami were interviewed, and the kinds of factors in the process of healing that sadness were analysed. As a result, it was discovered that, given the devoutly Buddhist nature of the area, the two families’ faith worked to relieve their sadness. There was also psychological support from neighbours and hope for the future embodied in the birth of children, which were also factors in healing their sadness

    昨年12月26日に,インドネシアのスマトラ島北端沖で発生したスマトラ沖大地震によるインド洋津波被害で,スマトラについで津波による被害が多かったスリランカに,被災6ヵ月後の復興状況,感染症発生状況,被災者の健康調査などを目的に被災地の調査に入った.スリランカ東南部地域の被災状況を報告し,看護職としての援助のあり方や,物資援助について考察した.We visited Sri Lanka, which received the most damage after Sumatra six months after the Sumatra Earthquake struck the northern coast of the Indonesian island last year on December 26 and caused a tsunami in the Indian Ocean. We investigated reconstruction efforts, the spread of infection and the health conditions of survivors in the southeast portion of the island country, made reports on the devastated areas and made observations on health care and aid supplies

    The approach described in this paper uses an array of Field Programmable Gate Array (FPGA) devices to implement a fault tolerant hardware system that can be compared to the running of fault tolerant software on a traditional processor. Fault tolerance is achieved is achieved by using FPGA with on the fly partial programmability feature. Major considerations while mapping to the FPGA includes the size of the area to be mapped and communication issues related to their communication. Area size selection is compared to the page size selection in Operating System Design. Communication issues between modules are compared to the software engineering paradigms dealing with module coupling, fan-in, fan-out and cohesiveness. Finally, the overhead associated with the downloading of the reconfiguration files is discussed

    Research and development for accuracy improvement of neutron nuclear data on minor actinides

    To improve accuracy of neutron nuclear data on minor actinides, a Japanese nuclear data project entitled “Research and development for Accuracy Improvement of neutron nuclear data on Minor ACtinides (AIMAC)” has been implemented. Several independent measurement techniques were developed for improving measurement precision at J-PARC/MLF/ANNRI and KURRI/LINAC facilities. Effectiveness of combining the independent techniques has been demonstrated for identifying bias effects and improving accuracy, especially in characterization of samples used for nuclear data measurements. Capture cross sections and/or total cross sections have been measured for Am-241, Am-243, Np-237, Tc-99, Gd-155, and Gd-157. Systematic nuclear data evaluation has also been performed by taking into account the identified bias effect. Highlights of the AIMAC project are outlined
