The direct association of the multiple PDZ domain containing proteins (MUPP-1) with the human c-Kit C-terminus is regulated by tyrosine kinase activity
AbstractWe have identified the multiple PDZ domain containing protein (MUPP-1 or MPDZ) as a novel binding partner of the human c-Kit. c-Kit binds specifically to the 10th PDZ domain of MUPP-1 via its C-terminal sequence. Furthermore, a kinase negative-mutant receptor interacted more strongly with MUPP-1 than the wild-type c-Kit. Strikingly, a constitutively activated c-Kit (D816V-Kit) did not bind to MUPP-1, although this oncogenic form retains the PDZ binding motif ‘HDDV’ at the C-terminal end. Deletion of V967 of c-Kit abolished binding to MUPP-1 and drastically reduced its tyrosine kinase activity, suggesting that the structure of the C-terminal tail of c-Kit influences its enzymatic activity
The cytotoxin of Pseudomonas aeruginosa:Cytotoxicity requires proteolytic activation
The primary structure of a cytotoxin from Pseudomonas aeruginosa was determined by sequencing of the structural gene. The cytotoxin (31,700 Mr) lacks an N-terminal signal sequence for bacterial secretion but contains a pentapeptide consensus sequence commonly found in prokaryotic proteins which function in a TonB-dependent manner. The cytotoxin gene has a [G + C]-content of 53.8% which is considerably lower than generally observed for genes from Pseudomonas aeruginosa. The cytotoxin gene was exclusively detected in strain 158 but not in three other clinical isolates, as determined by Southern and Northern hybridization. The latter technique revealed that the toxin is translated from monocistronic mRNA. The promoter of the cytotoxin is inactive in Escherichia coli. Upon site-directed modification of the 5'-noncoding region by the polymerase chain reaction the gene was expressed under control of the trc-promoter. The gene product obtained in Escherichia coli was nontoxic. Toxicity was induced by subsequent treatment with trypsin. [35S]methionine-labeled cytotoxin with high specific radioactivity was obtained by in vitro transcription/translation. Like [125I] labeled material from Pseudomonas aeruginosa this polypeptide bound to membrane preparations from Ehrlich ascites cells, as evidenced by sedimentation through a sucrose gradient at neutral pH
Discovery and quantification of a widespread methane ebullition event in a coastal inlet (Baltic Sea) using a novel sonar strategy
How much of the greenhouse gas methane is transported from the seafloor to the atmosphere is unclear. Here, we present data describing an extensive ebullition event that occurred in Eckernförde Bay, a shallow gas-hosting coastal inlet in the Baltic Sea, in the fall of 2014. A weak storm induced hydrostatic pressure fluctuations that in turn stimulated gas ebullition from the seabed. In a finely tuned sonar survey of the bay, we obtained a hydroacoustic dataset with exceptionally high sensitivity for bubble detection. This allowed us to identify 2849 bubble seeps rising within 28 h from the seafloor across the 90 km² study site. Based on our calculations, the estimated bubble-driven episodic methane flux from the seafloor across the bay is 1,900 μMol m-2 d-1. Our study demonstrates that storm-associated fluctuations of hydrostatic pressure induce bulk gas-driven ebullitions. Given the extensive occurrence of shallow gas-hosting sediments in coastal seas, similar ebullition events probably take place in many parts of the Western Baltic Sea. However, these are likely to be missed during field investigations, due to the lack of high-quality data acquisition during storms, such that atmospheric inputs of marine-derived methane will be highly underestimated
Evidence for conserved DNA and histone H3 methylation reprogramming in mouse, bovine and rabbit zygotes
<p>Abstract</p> <p>Background</p> <p>In mammals the parental genomes are epigenetically reprogrammed after fertilization. This reprogramming includes a rapid demethylation of the paternal (sperm-derived) chromosomes prior to DNA replication in zygotes. Such active DNA demethylation in the zygote has been documented for several mammalian species, including mouse, rat, pig, human and cow, but questioned to occur in rabbit.</p> <p>Results</p> <p>When comparing immunohistochemical patterns of antibodies against 5-methyl-cytosine, H3K4me3 and H3K9me2 modifications we observe similar pronuclear distribution and dynamics in mouse, bovine and rabbit zygotes. In rabbit DNA demethylation of the paternal chromosomes occurs at slightly advanced pronuclear stages. We also show that the rabbit oocyte rapidly demethylates DNA of donor fibroblast after nuclear transfer.</p> <p>Conclusion</p> <p>Our data reveal that major events of epigenetic reprogramming during pronuclear maturation, including mechanisms of active DNA demethylation, are apparently conserved among mammalian species.</p
