    Stallion Sperm Viability, as Measured by the Nucleocounter SP-100, Is Affected by Extender and Enhanced by Single Layer Centrifugation

    On-stud assessment of stallion sperm quality can be problematic. A new instrument, the Nucleocounter SP-100, was validated for measuring stallion sperm concentration and viability. It was subsequently used to evaluate sperm viability in Kenney's extender and INRA96. There was a strong correlation between sperm concentrations measured by the Nucleocounter SP-100 and by the Bürker counting chamber (r = 0.84; P < .001). Similarly, there was a good correlation between sperm viability results from the Nucleocounter SP-100 and flow cytometric results (r = 0.73; P < .001). Sperm viability at 24 hours was significantly better for samples extended in INRA96 than in Kenney's extender (P < .001). Furthermore, sperm kinematics were better for stored samples in INRA96 than in Kenney's extender. Single Layer Centrifugation selected spermatozoa that maintained their viability better during storage for 24 hours than the uncentrifuged samples. In conclusion, the type of semen extender used and Single Layer Centrifugation were found to influence both the kinematics and viability of stallion spermatozoa.     Some stallions yield ejaculates that do not tolerate conservation by refrigeration prior to artificial insemination (AI), showing improvement after removal of most of the seminal plasma (SP) by centrifugation. In this study, the SP-proteome of 10 different stallions was defined through high-performance liquid chromatography with tandem mass spectrometry and bioinformatic analysis in relation to the ability of the ejaculates to maintain semen quality when cooled and stored at 5°C. Stallions were classified into three groups, depending on this ability: those maintaining good quality after direct extension in a commercial extender (good), stallions requiring removal of seminal plasma (RSP) to maintain seminal quality (good-RSP), and stallions, unable to maintain good semen quality even after RSP (poor). Pathway enrichment analysis of the proteins identified in whole equine SP using human orthologs was performed using g: profiler showing enriched Reactome and the Kyoto Encyclopedia of Genes and Genomes pathways related to hexose metabolism, vesicle mediated transport, post translational modification of proteins and immune response. Specific proteins overrepresented in stallions tolerating conservation by refrigeration included a peroxiredoxin-6 like protein, and transcobalamin-2, a primary vitamin B12-binding, and transport protein. Also, the protein involved in protein glycosylation, ST3 beta-galactoside alpha-2,3-sialyltransferase 1 was present in good stallions. These proteins were nearly absent in poor stallions. Particularly, annexinA2 appeared as to be the most powerful discriminant variable for identification of stallions needing RSP prior to refrigeration, with a P = 0.002 and a q value = 0.005. Overall this is the first detailed study of the equine SP-proteome, showing the potential value of specific proteins as discriminant bio-markers for clinical classification of stallions for AI

    Structural Disorder Induced Polaron Formation and Magnetic Scattering in the Disordered Holstein-Double Exchange Model

    In this paper we present results on the disordered Holstein-Double Exchange model, explicitly in three dimension and `metallic' densities, obtained by using a recently developed Monte Carlo approach. Following up on our earlier paper, cond-mat/0406085, here we provide a detailed microscopic picture of the thermally driven metal-insulator transition (MIT) that arises close to the ferromagnet to paramagnet transition in this problem. This paper is focused mainly on the `diagnostics', clarifying the origin of the effective disorder that drives the MIT in this system. To that effect, we provide results on the thermal evolution of the distributions of (i) lattice distortions, (ii) the net `structural disorder' and (iii) the `hopping disorder' arising from spin randomness feeding back through the Hunds coupling. We suggest a phenomenology for the thermally driven MIT, viewing it as an `Anderson-Holstein' transition.Comment: 6 pages, latex, JPSJ style, 7 eps figs. Style files included.     Suppression of ferromagnetic ordering in doped manganites: Effects of the superexchange interaction

    From a Monte Carlo study of the ferromagnetic Kondo lattice model for doped manganites, including the antiferromagnetic superexchange interaction (JAFJ_{AF}), we found that the ferromagnetic ordering was suppressed as JAFJ_{AF} increased. The ferromagnetic transition temperature TcT_c, as obtained from a mean field fit to the calculated susceptibilities, was found to decrease monotonically with increasing JAFJ_{AF}. Further, the suppression in TcT_c scales with the bandwidth narrowing induced by the antiferromagnetic frustration originating from JAFJ_{AF}. From these results, we propose that the change in the superexchange interaction strength between the t2gt_{2g} electrons of the Mn ions is one of the mechanisms responsible for the suppression in TcT_c observed in manganites of the type (La0.7y_{0.7-y}Pry_{y})Ca0.3_{0.3}MnO3_3.Comment: 5 pages, 6 figures.     The Effect of ff-dd Magnetic Coupling in Multiferroic RRMnO3_3 Crystals

    We have established detailed magnetoelectric phase diagrams of (Eu0.595_{0.595}Y0.405_{0.405})1x_{1-x}Tbx_xMnO3_3 (0x10 \le x \le 1) and (Eu,Y)1x_{1-x}Gdx_xMnO3_3 (0x0.690 \le x \le 0.69), whose average ionic radii of RR-site (RR: rare earth) cations are equal to that of Tb3+^{3+}, in order to reveal the effect of rare earth 4ff magnetic moments on the magnetoelectric properties. In spite of the same RR-site ionic radii, the magnetoelectric properties of the two systems are remarkably different from each other. A small amount of Tb substitution on RR sites (x0.2x \sim 0.2) totally destroys ferroelectric polarization along the a axis (PaP_a), and an increase in Tb concentration stabilizes the PcP_c phase. On the other hand, Gd substitution (x0.2x \sim 0.2) extinguishes the PcP_c phase, and slightly suppresses the PaP_a phase. These results demonstrate that the magnetoelectric properties of RRMnO3_3 strongly depend on the characteristics of the rare earth 4ff moments.Comment: 10 pages, 5 figures Submitted to Journal of the Physical Society of Japa
