    Some Orchid Species Fungi Isolated by Different Methods

    Due to their very small seeds that do not contain endosperm, many terrestrial orchid species require the presence of fungi in order to germinate and maintain their lives; and symbiotic culture studies are being carried out on this topic. For the purpose of determining the orchid species on which the fungus to be used as inoculants in the symbiotic culture will be effective, fungi isolated through several isolation methods are cultured with orchid species. In this study a total of four different isolation methods were applied as one on the tubers and rhizomes and three on the soil of eleven orchid species from the Anacamptis, Cephalanthera, Dactylorhiza and Orchis genera. Three different culture media were used in the methods. At the end of the study Alternaria, Aspergillus, Fusarium, Macrophomina, Rhizoctonia, Trichoderma and Verticillium fungi were isolated. In the study that was conducted with the aimed to isolate particularly Rhizoctania spp. fungi, the fungi was isolated from the tubers of Dactylorhiza umbrosa and Orchis palustris species and the soil of the Orchis simia species.     Bu çalışmada; Türkiye’de yetiştirilen standart nar çeşitleri (Hicaz narı, Silifke Aşısı, Katırbaşı, 33N23-Çevlik, 01N04-Fellahyemez, 33N34, İzmir-26, İzmir-23, İzmir-1513, 33N24, Kuş narı ) ile Gündoğdu (2006)’nun Pervari (Siirt) yöresinde daha önceden belirlemiş olduğu en iyi 5 nar genotipinde (56PER021, 56PER022, 56PER020, 56PER019, 56PER003) meyvelerin pomolojik özellikleri ve bazı kimyasal içeriklerinin tespit edilmesi amaçlanmıştır. Araştırmada meyve ağırlığını 251.01-530.25 gr, meyve uzunluğunu 60.30-89.97 mm, meyve enin 75.57- 100.68 mm, meyve hacminin 230.00-542.50 cm3, meyve suyu miktarının 106.66-186 ml ve meyve yoğunluğunun 0.92 -1.19 gr cm-3, SÇKM miktarının % 11.50-14.62, pH 3.45-4.71, şekil indeksinin 0.82-0.92 ve toplam asitliğin % 0.19-1.17 arasında değişiklik gösterdiği tespit edilmiştir. Bunun yanında incelenen çeşit ve genotiplerin kabuk alt zemin rengi, üst zemin rengi, çekirdek sertliği, meyve tadı, dane rengi, üst odacık sayısı, alt odacık sayısı, odacıkların görünümü ve daneleme kolaylığı belirlenmiştir

    Early protective effects of iloprost, a stable prostacyclin analog, during spinal cord ischemia in a rabbit model

    Spinal cord ischemia may develop into paraplegia in some cases during operation of the thoracoabdominal aorta. This is attributable to the vulnerability of spinal motor neurons to ischemia. In this study, iloprost was used as an agent to decrease the severity of ischemia and reperfusion injury to the spinal cord motor neurons. Twenty-one rabbits were randomized into three groups of seven animals each: group A (iloprost not administered), group B (25 ng/kg per minute iloprost), and group S (sham-operated). The spinal cord ischemia model was created by a 15-min occlusion of the aorta just caudal to the renal artery with a balloon catheter. Administration of iloprost began 10 min before occlusion of the aorta, and continued thereafter for 60 min. The pre- and postocclusion arterial pressure and heart rate recordings, results of blood gas analyses, and hematocrit and glucose levels were recorded. The spinal cords were removed after 8-h monitoring of neurologic function. Viable and nonviable motor neurons in the anterior horn of the spinal cord were counted under light microscopy. Any significant alteration in hemodynamics, blood gases, and other physiologic parameters could not be detected within the groups. Iloprost had a moderately hypotensive effect. Neurologic function in terms of Johnson scoring was significantly better in the iloprost group (P < 0.05). The number of viable cells was higher, whereas the number of nonviable cells was lower in iloprost group, when compared with the control group ( P < 0.05). Higher numbers of viable motor neurons were consistent with the neurological findings. As a result of this study we concluded that iloprost infused during clamping of the aorta mitigates the spinal cord injury due to ischemia and reperfusion, and has a significant protective effect
