Genetic Regulatory Perturbation of Gene Expression Impacted by Genomic Introgression in Fiber Development of Allotetraploid Cotton
Interspecific genomic introgression is an important evolutionary process with respect to the generation of novel phenotypic diversity and adaptation. A key question is how gene flow perturbs gene expression networks and regulatory interactions. Here, an introgression population of two species of allopolyploid cotton (Gossypium) to delineate the regulatory perturbations of gene expression regarding fiber development accompanying fiber quality change is utilized. De novo assembly of the recipient parent (G. hirsutum Emian22) genome allowed the identification of genomic variation and introgression segments (ISs) in 323 introgression lines (ILs) from the donor parent (G. barbadense 3–79). It documented gene expression dynamics by sequencing 1,284 transcriptomes of developing fibers and characterized genetic regulatory perturbations mediated by genomic introgression using a multi-locus model. Introgression of individual homoeologous genes exhibiting extreme low or high expression bias can lead to a parallel expression bias in their non-introgressed duplicates, implying a shared yet divergent regulatory fate of duplicated genes following allopolyploidy. Additionally, the IL N182 with improved fiber quality is characterized, and the candidate gene GhFLAP1 related to fiber length is validated. This study outlines a framework for understanding introgression-mediated regulatory perturbations in polyploids, and provides insights for targeted breeding of superior upland cotton fiber.This article is published as Chen, Xinyuan, Xiubao Hu, Guo Li, Corrinne E. Cell transcriptomic atlas of the non-human primate Macaca fascicularis.
Studying tissue composition and function in non-human primates (NHPs) is crucial to understand the nature of our own species. Here we present a large-scale cell transcriptomic atlas that encompasses over 1 million cells from 45 tissues of the adult NHP Macaca fascicularis. This dataset provides a vast annotated resource to study a species phylogenetically close to humans. To demonstrate the utility of the atlas, we have reconstructed the cell-cell interaction networks that drive Wnt signalling across the body, mapped the distribution of receptors and co-receptors for viruses causing human infectious diseases, and intersected our data with human genetic disease orthologues to establish potential clinical associations. The Perception of the National Traceability Platform among Small-Scale Tea Farmers in Typical Agricultural Areas in Central China
As one of the key technologies to ensure the safety of agricultural products, the national traceability platform is being widely promoted in China. However, it has not yet been widely adopted among farmers, especially small-scale farmers. Farmers are both producers and direct participants in the traceability of agricultural products. Their perception directly affects the effectiveness of the promotion of the national traceability platform. This study explores the perception of the national traceability platform among small-scale tea farmers in typical agricultural areas in central China. This research employed Q methodology, an approach that integrates both qualitative and quantitative data allowing individuals’ subjective understandings of a specific topic to be studied. The Q-sort procedure was performed in the field with 16 small-scale tea farmers. Next, Q-factor analyses were conducted using the Ken-Q analysis. The results show that small-scale tea farmers have different perceptions of the national traceability platform. Their main characteristics are active participation, resistant participation, risk aversion, and being driven by pressure. These four categories covered 52% of the perceived variance. Meanwhile, there is also a degree of internal consistency in the perception of small-scale tea farmers. Specifically, they are all concerned that participating in the national traceability platform may increase the cost and risk of cultivation and that it is difficult to obtain support from agricultural technicians. Therefore, understanding the perceptions of tea farmers of the national traceability platform is the premise for formulating effective promotion policies. Our research sheds light on the decision-making mechanisms for small-scale tea farmers to participate in national traceability platforms, further expanding the scope of current research on farmer behavior. This research has reference significance for promoting national traceability platforms in China and other countries around the world
Sulfolobus Replication Factor C stimulates the activity of DNA Polymerase B1
Replication factor C (RFC) is known to function in loading proliferating cell nuclear antigen (PCNA) onto primed DNA, allowing PCNA to tether DNA polymerase for highly processive DNA synthesis in eukaryotic and archaeal replication. In this report, we show that an RFC complex from the hyperthermophilic archaea of the genus Sulfolobus physically interacts with DNA polymerase B1 (PolB1) and enhances both the polymerase and 3′-5′ exonuclease activities of PolB1 in an ATP-independent manner. Stimulation of the PolB1 activity by RFC is independent of the ability of RFC to bind DNA but is consistent with the ability of RFC to facilitate DNA binding by PolB1 through protein-protein interaction. These results suggest that Sulfolobus RFC may play a role in recruiting DNA polymerase for efficient primer extension, in addition to clamp loading, during DNA replication
Study on Spraying Construction Method of a Non-Water Reacting Polymer Layer in the Tunnel
Non-water reacting double-component foamed polyurethane has been increasingly used in the field of transportation. Particularly, it is recognized that a polymer damping layer between tunnel linings and surrounding rocks can improve the seismic performance of tunnels. To facilitate the application of this polymer in tunnels, a spraying construction method of polymer damping layers was proposed. The polymer damping layer was prepared with hydraulic spraying equipment, and the construction process included the pre-treatment of the tunnel base surface, the pressure control of the spraying equipment, the adjustment of the spray gun working parameters and spraying quality control. In this paper, the effects of material ratio, material temperature, environmental factors (i.e., temperature, humidity and wind speed), spraying pressure and spray gun parameters (i.e., speed, distance from the sprayed surface and spray angle) on the layer formation were investigated. Thus, spraying parameters for better spraying performance were recommended. This study will provide technical support for polymer damping layer construction in the seismic design of tunnels
Assembly of CaIn2S4 on Defect-Rich BiOCl for Acceleration of Interfacial Charge Separation and Photocatalytic Phenol Degradation via S-Scheme Electron Transfer Mechanism
The novel 2D/2D S-scheme heterostructure of BiOCl nanosheets coupled with CaIn2S4 nanosheets (CaIn2S4/BiOCl-SOVs), which contains surface oxygen vacancies (SOVs), has been successfully prepared by high-temperature calcination combined with a solvothermal synthetic strategy. Under visible-light irradiation, the apparent rate constant (Kapp/mim−1) for phenol degradation on the 1 wt% CaIn2S4/BiOCl-SOVs photocatalyst is about 32.8 times higher than that of pure BiOCl. The superior performance was attributed to the synergistic effect between the SOVs, CaIn2S4, and BiOCl, which can effectively narrow the bandgap and accelerate the interfacial charge separation of CaIn2S4/BiOCl-SOVs heterojunctions. Subsequently, it significantly promotes the generation of superoxide radicals (O2−), hydroxyl radicals, and h+, which participate in the photodegradation process of phenol. The catalyst still maintained a relatively high activity after repeated tests as a demonstration of its photostability. This work successfully proposed an efficient method to design a new 2D/2D S-scheme heterostructure with SOVs as possible photocatalysts in the field of environmental remediation
SiO_2@Fe_3O_4@C胶体纳米颗粒的制备及其光学性质
Highly dispersed SiO_2@Fe_3O_4@C core-shell nanoparticles with controllable particle sizes were prepared.Under electric field,the optical properties of SiO_2@Fe_3O_4@C nanoparticle suspensions of different size,different concentrations,different the solvent was studied,and the results show that the reflection spectrum of 150 nm SiO_2@Fe_3O_4@C nanoparticles at a concentration of 10%(mass fraction) obtain the widest tunable range from 735 nm to 540 nm.With increasing concentration of nanoparticle suspension,the overall reflectance spectra blue shift.The solvent dependence of the electric fieldresponsive photonic crystals by dispersing 150 nm particles into different solvents was investigated,and the results show that the greater refractive index,the greater the reflection wavelength.The response time of the suspensions is 150 ms,and the suspensions have recovery after electric field disappeared
The deubiquitinase EIF3H promotes hepatocellular carcinoma progression by stabilizing OGT and inhibiting ferroptosis
Abstract Hepatocellular carcinoma (HCC) is one of the most prevalent and lethal human malignancies, and with quite limited treatment alternatives. The proteasome is responsible for most of the protein degradation in eukaryotic cells and required for the maintenance of intracellular homeostasis. However, its potential role in HCC is largely unknown. In the current study, we identified eukaryotic translation initiation factor 3 subunit H (EIF3H), belonging to the JAB1/MPN/MOV34 (JAMM) superfamily, as a bona fide deubiquitylase of O-GlcNAc transferase (OGT) in HCC. We explored that EIF3H was positively associated with OGT in HCC and was related to the unfavorable prognosis. EIF3H could interact with, deubiquitylate, and stabilize OGT in a deubiquitylase-dependent manner. Specifically, EIF3H was associated with the GT domain of ERα via its JAB/MP domain, thus inhibiting the K48-linked ubiquitin chain on OGT. Besides, we demonstrated that the knockdown of EIF3H significantly reduced OGT protein expression, cell proliferation and invasion, and caused G1/S arrest of HCC. We also found that the deletion of EIF3H prompted ferroptosis in HCC cells. Finally, the effects of EIF3H depletion could be reversed by further OGT overexpression, implying that the OGT status is indispensable for EIF3H function in HCC carcinogenesis. In summary, our study described the oncogenic function of EIF3H and revealed an interesting post-translational mechanism between EIF3H, OGT, and ferroptosis in HCC.
