    Mobilization of colloids in groundwater due to infiltration of water at a coal ash disposal site

    We investigated groundwaters in the vicinity of a coal ash site near an electric generating station in the western U.S.A. The purpose of the study was to ascertain why fine particles or colloids appear in some subsurface water samples there. If such fine particles are merely introduced during bailing or pumping operations which suspend otherwise immobile soil colloids, we should exclude these particulate materials from the water samples before analysis intended to quantify what is moving through the aquifer. However, if the colloids were truly suspended and moving with the groundwater flow in situ, then we should include their contribution to our assessment of the mobile loads. Application of very careful sampling techniques (slow pumping rates, no atmospheric exposure) did not cause the large quantities of colloids observed previously to disappear from well water in which they occurred. Additionally, the same sampling procedures did not cause similar abun- dances of colloids to appear in waters collected from neighboring wells installed and' developed in the same manner and in the same geologic strata. Thus we believe sampling artifacts do not explain the colloids' presence in the groundwater samples. On the other hand, the groundwater chemistry and the nature of the suspended colloids (size, composition) strongly suggest these fine particles were suspended and therefore moving with the groundwater flow. At wells exhibiting large amounts of suspended colloids (- 10-100mg L-), the water was enriched in CO2 and depleted in 02 relative to nearby locations. The colloids were typically between 0.1 and 2 gm in size and were primarily silicates. These results suggest to us that, where infiltrating water is percolating through a site that has been mixed with coal ash, the secondary carbonate minerals in the soils are being dissolved; removal of this cementing carbonate phase may consequently release soil silicate colloids to be carried in the flowing water. Such processes may enhance contaminant transport in groundwater by augmenting the pollutant load moving in the groundwater, and increasing the permeability of the porous medium to pollutant infiltration with waste water and/or rainwater

    An economically viable ionic liquid for the fractionation of lignocellulosic biomass

    Cost-effective fractionation (pretreatment) of lignocellulosic biomass is necessary to enable its large-scale use as a source of liquid fuels, bio-based materials and bio-derived chemicals. While a number of ionic liquids (ILs) have proven capable of highly effective pretreatment, their high cost presents a barrier to commercial viability. In this study, we investigate in detail the application of the low-cost (ca. $1 kg−1) ionic liquid triethylammonium hydrogen sulfate for the fractionation of the grass Miscanthus x giganteus into a cellulose rich pulp, a lignin and a distillate. We found that up to 85% of the lignin and up to 100% of the hemicellulose were solubilized into the IL solution. The hemicellulose dissolved mainly in monomeric form, and pentoses were partially converted into furfural. Up to 77% of the glucose contained in the biomass could be released by enzymatic saccharification of the pulp. The IL was successfully recovered and reused four times. A 99% IL recovery was achieved each time. Effective lignin removal and high saccharification yields were maintained during recycling, representing the first demonstration that repeated IL use is feasible due to the self-cleaning properties of the non-distillable solvent. We further demonstrate that furfural and acetic acid can be separated quantitatively from the non-volatile IL by simple distillation, providing an easily recoverable, valuable co-product stream, while IL degradation products were not detected. We further include detailed mass balances for glucose, hemicellulose and lignin, and a preliminary techno-economic estimate for the fractionation process. This is the first demonstration of an efficient and repeated lignocellulose fractionation with a truly low-cost IL, and opens a path to an economically viable IL-based pretreatment process

    Deconstruction with low-cost ionic liquids (ionoSolv) is a promising method to pre-condition lignocellulosic biomass for the production of renewable fuels, materials and chemicals. This study investigated process intensification strategies for the ionoSolv pretreatment of Miscanthus X giganteus using the low-cost ionic liquid triethylammonium hydrogen sulfate ([TEA][HSO4]) in the presence of 20 wt% water, using high temperatures and a high solid to solvent loading of 1:5 g/g. The temperatures investigated were 150, 160, 170 and 180°C. We discuss the effect of pretreatment temperature on lignin and hemicellulose removal, cellulose degradation and enzymatic saccharification yields. We report that very good fractionation can be achieved across all investigated temperatures, including an enzymatic saccharification yield exceeding 75% of the theoretical maximum after only 15 min of treatment at 180°C. We further characterised the recovered lignins which established some tunability of the hydroxyl group content, subunit composition, connectivity and molecular weight distribution in the isolated lignin while maintaining maximum saccharification yield. This drastic reduction of pretreatment time at increased biomass loading without a yield penalty is promising for the development of a commercial ionoSolv pretreatment process

    Pretreatment of Lignocellulosic Biomass with Low-cost Ionic Liquids.

    A number of ionic liquids (ILs) with economically attractive production costs have recently received growing interest as media for the delignification of a variety of lignocellulosic feedstocks. Here we demonstrate the use of these low-cost protic ILs in the deconstruction of lignocellulosic biomass (Ionosolv pretreatment), yielding cellulose and a purified lignin. In the most generic process, the protic ionic liquid is synthesized by accurate combination of aqueous acid and amine base. The water content is adjusted subsequently. For the delignification, the biomass is placed into a vessel with IL solution at elevated temperatures to dissolve the lignin and hemicellulose, leaving a cellulose-rich pulp ready for saccharification (hydrolysis to fermentable sugars). The lignin is later precipitated from the IL by the addition of water and recovered as a solid. The removal of the added water regenerates the ionic liquid, which can be reused multiple times. This protocol is useful to investigate the significant potential of protic ILs for use in commercial biomass pretreatment/lignin fractionation for producing biofuels or renewable chemicals and materials

    La stratégie d’élimination de la rougeole 2011-2015 élaborée et mise en oeuvre par l’OFSP a été évaluée en utilisant diverses approches méthodologiques complémentaires. Les principaux résultats montrent qu’un large consensus sur la stratégie a été obtenu sous l’impulsion de l’OFSP et que tous les cantons ont mis en oeuvre des mesures visant à augmenter la couverture vaccinale. Une augmentation de la couverture vaccinale, du rattrapage vaccinal et une réduction des disparités cantonales de couverture vaccinale ont été observées. Cependant l’objectif stratégique de couverture vaccinale 2 doses à l’âge de 2 ans n’a pas été obtenu dans tous les cantons pour lesquels on dispose d’une mesure en 2014-2015. Il est probable que la stratégie n’a pas encore déployé tous ses effets. Les mesures mises en oeuvre pour prévenir et contenir les flambées épidémiques et qui visent à augmenter la couverture vaccinale devraient être poursuivies

    Exploring the effect of water content and anion on the pretreatment of poplar with three 1-Ethyl-3-methylimidazolium ionic liquids

    We report on the pretreatment of poplar wood with three different 1-ethyl-3-methylimidazolium ionic liquids, [EMim][OAc], [EMim][MeSO3], and [EMim][HSO4], at varying water contents from 0–40 wt% at 100 °C. The performance was evaluated by observing the lignin and hemicellulose removal, as well as enzymatic saccharification and lignin yield. The mechanism of pretreatment varied between the ionic liquids studied, with the hydrogen sulfate ionic liquid performing delignification and hemicellulose hydrolysis more effectively than the other solvents across the investigated water content range. The acetate ionic liquid produced superior glucose yield at low water contents, while the hydrogen sulfate ionic liquid performed better at higher water contents and produced a recoverable lignin. The methanesulfonate ionic liquid did not introduce significant fractionation or enhancement of saccharification yield under the conditions used. These findings help distinguish the roles of anion hydrogen bonding, solvent acidity, and water content on ionic liquid pretreatment and can aid with anion and water content selections for different applications

    S6K1 and 4E-BP1 Are Independent Regulated and Control Cellular Growth in Bladder Cancer

    Aberrant activation and mutation status of proteins in the phosphatidylinositol-3-kinase (PI3K)/Akt/mammalian target of rapamycin (mTOR) and the mitogen activated protein kinase (MAPK) signaling pathways have been linked to tumorigenesis in various tumors including urothelial carcinoma (UC). However, anti-tumor therapy with small molecule inhibitors against mTOR turned out to be less successful than expected. We characterized the molecular mechanism of this pathway in urothelial carcinoma by interfering with different molecular components using small chemical inhibitors and siRNA technology and analyzed effects on the molecular activation status, cell growth, proliferation and apoptosis. In a majority of tested cell lines constitutive activation of the PI3K was observed. Manipulation of mTOR or Akt expression or activity only regulated phosphorylation of S6K1 but not 4E-BP1. Instead, we provide evidence for an alternative mTOR independent but PI3K dependent regulation of 4E-BP1. Only the simultaneous inhibition of both S6K1 and 4E-BP1 suppressed cell growth efficiently. Crosstalk between PI3K and the MAPK signaling pathway is mediated via PI3K and indirect by S6K1 activity. Inhibition of MEK1/2 results in activation of Akt but not mTOR/S6K1 or 4E-BP1. Our data suggest that 4E-BP1 is a potential new target molecule and stratification marker for anti cancer therapy in UC and support the consideration of a multi-targeting approach against PI3K, mTORC1/2 and MAPK

    Rapid divergence and expansion of the X chromosome in papaya

    X chromosomes have long been thought to conserve the structure and gene content of the ancestral autosome from which the sex chromosomes evolved. We compared the recently evolved papaya sex chromosomes with a homologous autosome of a close relative, the monoecious Vasconcellea monoica, to infer changes since recombination stopped between the papaya sex chromosomes. We sequenced 12 V. monoica bacterial artificial chromosomes, 11 corresponding to the papaya X-specific region, and 1 to a papaya autosomal region. The combined V. monoica X-orthologous sequences are much shorter (1.10 Mb) than the corresponding papaya region (2.56 Mb). Given that the V. monoica genome is 41% larger than that of papaya, this finding suggests considerable expansion of the papaya X; expansion is supported by a higher repetitive sequence content of the X compared with the papaya autosomal sequence. The alignable regions include 27 transcript-encoding sequences, only 6 of which are functional X/V. monoica gene pairs. Sequence divergence from the V. monoica orthologs is almost identical for papaya X and Y alleles; the Carica-Vasconcellea split therefore occurred before the papaya sex chromosomes stopped recombining, making V. monoica a suitable outgroup for inferring changes in papaya sex chromosomes. The papaya X and the hermaphrodite-specific region of the Y(h) chromosome and V. monoica have all gained and lost genes, including a surprising amount of changes in the X

    Hat sich die Unabhängigkeitsstrategie der FDP bei der letzten Bundestagswahl ausgezahlt? Wäre die FDP erfolgreicher gewesen, wenn sie im Vorfeld klar signalisiert hätte, dass man eine Koalition mit der Union anstrebt? Wie war das bei den Grünen, die ja im Gegensatz zur FDP keine Zweifel aufkommen ließen? Natürlich können wir nicht wie in einer Simulation oder einem Experiment einfach den Wahlkampf wiederholen und noch einmal wählen lassen. Um eine befriedigende Antwort auf diese Frage zu finden, vergleichen wir den Kontext der Bundestagswahl 2002 mit den zurückliegenden Bundestagswahlen. Aus dem Längsschnittvergleich versuchen wir Rückschlüsse auf den substanziellen Einfluss von strategischem Stimmensplitting im Sinne einer Koalitionswahl auf das Wahlergebnis gerade der kleinen Parteien zu ziehen. Um unsere Forschungsfrage zu beantworten und substanzielle Schlüsse ziehen zu können, muss zuerst klar sein, in welcher Form und warum Stimmensplitting relevant sein kann, welche Rolle dabei Koalitionsabsprachen vor einer jeden Wahl spielen und, schließlich, welche alternativen Erklärungsmöglichkeiten die Literatur zum Thema Stimmensplitting und strategischem Wählen anzubieten hat. Nur wenn wir auch die Wirkung alternativer und zum Teil konkurrierender Hypothesen zulassen, können wir unserer Schlußfolgerungen sicher sein

    Brown dwarf census with the Dark Energy Survey year 3 data and the thin disc scale height of early L types

    27 pages, 18 figuresIn this paper we present a catalogue of 11 745 brown dwarfs with spectral types ranging from L0 to T9, photometrically classified using data from the Dark Energy Survey (DES) year 3 release matched to the Vista Hemisphere Survey (VHS) DR3 and Wide-field Infrared Survey Explorer (WISE) data, covering ≈2400 deg2 up to iAB = 22. The classification method follows the same phototype method previously applied to SDSS-UKIDSS-WISE data. The most significant difference comes from the use of DES data instead of SDSS, which allow us to classify almost an order of magnitude more brown dwarfs than any previous search and reaching distances beyond 400 pc for the earliest types. Next, we also present and validate the GalmodBD simulation, which produces brown dwarf number counts as a function of structural parameters with realistic photometric properties of a given survey. We use this simulation to estimate the completeness and purity of our photometric LT catalogue down to iAB = 22, as well as to compare to the observed number of LT types. We put constraints on the thin disc scale height for the early L (L0–L3) population to be around 450 pc, in agreement with previous findings. For completeness, we also publish in a separate table a catalogue of 20 863 M dwarfs that passed our colour cut with spectral types greater than M6.
