    A difluorinated dienophile underwent cycloaddition reactions with a range of furans to afford cycloadducts whichcould be processed regio- and stereoselectively via episulfonium ions, generated by the reaction between their alkenyl groups and phenylsulfenyl chloride. The oxabicyclic products were oxidised to the phenylsulfonyl level and ring opened via E1CB or reductive desulfonative pathways to afford, ultimately, difluorinated cyclohexene or cyclohexane polyols

    Towards novel difluorinated sugar mimetrics; syntheses and conformational analyses of highly-functionalised difluorinated cyclooctenones

    Highly-functionalised difluorinated cyclooctenones were synthesised from trifluoroethanol using either metallated difluoroenol acetal or carbamate chemistry, followed by a [2,3]-Wittig rearrangement or aldol reaction. Efficient RCM reactions afforded the title compounds which showed rather restricted fluxional behaviour by VT 19F NMR. Topological characterisation by molecular modelling and NOESY/ROESY experiments offered a number of challenges, but allowed the identification of two favoured boat-chair conformers which interconverted by pseudorotation with relatively large activation barriers

    TCCON Philippines: First Measurement Results, Satellite Data and Model Comparisons in Southeast Asia

    The Total Carbon Column Observing Network (TCCON) is a global network dedicated to the precise and accurate measurements of greenhouse gases (GHG) in the atmosphere. The TCCON station in Burgos, Ilocos Norte, Philippines was established with the primary purpose of validating the upcoming Greenhouse gases Observing SATellite-2 (GOSAT-2) mission and in general, to respond to the need for reliable ground-based validation data for satellite GHG observations in the region. Here, we present the first 4 months of data from the new TCCON site in Burgos, initial comparisons with satellite measurements of CO_2 and model simulations of CO . A nearest sounding from Japan’s GOSAT as well as target mode observations from NASA’s Orbiting Carbon Observatory 2 (OCO-2) showed very good consistency in the retrieved column-averaged dry air mole fractions of CO_2 , yielding TCCON - satellite differences of 0.86 ± 1.06 ppm for GOSAT and 0.83 ± 1.22 ppm for OCO-2. We also show measurements of enhanced CO , probably from East Asia. GEOS-Chem model simulations were used to study the observed CO variability. However, despite the model capturing the pattern of the CO variability, there is an obvious underestimation in the CO magnitude in the model. We conclude that more measurements and modeling are necessary to adequately sample the variability over different seasons and to determine the suitability of current inventories

    The Mechanism of Formation of N-Formylkynurenine by Heme Dioxygenases

    [Image: see text] Heme dioxygenases catalyze the oxidation of l-tryptophan to N-formylkynurenine (NFK), the first and rate-limiting step in tryptophan catabolism. Although recent progress has been made on early stages in the mechanism, there is currently no experimental data on the mechanism of product (NFK) formation. In this work, we have used mass spectrometry to examine product formation in a number of dioxygenases. In addition to NFK formation (m/z = 237), the data identify a species (m/z = 221) that is consistent with insertion of a single atom of oxygen into the substrate during O(2)-driven turnover. The fragmentation pattern for this m/z = 221 species is consistent with a cyclic amino acetal structure; independent chemical synthesis of the 3a-hydroxypyrroloindole-2-carboxylic acid compound is in agreement with this assignment. Labeling experiments with (18)O(2) confirm the origin of the oxygen atom as arising from O(2)-dependent turnover. These data suggest that the dioxygenases use a ring-opening mechanism during NFK formation, rather than Criegee or dioxetane mechanisms as previously proposed

    Evaluating the Effects of SARS-CoV-2 Spike Mutation D614G on Transmissibility and Pathogenicity.

    Global dispersal and increasing frequency of the SARS-CoV-2 spike protein variant D614G are suggestive of a selective advantage but may also be due to a random founder effect. We investigate the hypothesis for positive selection of spike D614G in the United Kingdom using more than 25,000 whole genome SARS-CoV-2 sequences. Despite the availability of a large dataset, well represented by both spike 614 variants, not all approaches showed a conclusive signal of positive selection. Population genetic analysis indicates that 614G increases in frequency relative to 614D in a manner consistent with a selective advantage. We do not find any indication that patients infected with the spike 614G variant have higher COVID-19 mortality or clinical severity, but 614G is associated with higher viral load and younger age of patients. Significant differences in growth and size of 614G phylogenetic clusters indicate a need for continued study of this variant
