    Seasonal Variations in the Organization and Structure of Apis cerana cerana Swarm Queen Cells

    This paper describes the organization and structure of the swarm queen cells of Apis cerana cerana in spring, summer, and autumn in Kunming, Yunnan Province, China. We measured the following indices to reveal the organization rule of swarm cells: number of swarm cells built by each colony during different seasons; the shortest distance between two adjacent swarm cells on the comb; distance between swarm cell base and bottom bar of movable frame. We revealed the swarm cells structural characteristics using the following indicators: maximum diameter of swarm cell, the length between mouth and bottom of swarm cell, depth between maximum diameter and bottom of swarm cell, and the ratio of maximum diameter to depth between maximum diameter and bottom of swarm cell. Regarding seasonal differences, results indicated a significant variation in the distance between the swarm cell base and the bottom bar of the movable frame. Still, no such effect was observed in the shortest distance between two adjacent swarm cells. The maximum swarm cell diameter was not considerably influenced either, while the distance between the maximum diameter and the bottom of the swarm cell had substantial variation. The detected ratio of the maximum diameter to the depth between the maximum diameter and the bottom of theswarm cell indicated seasonal changes in the bottom shape of the swarm cell. This study clarifies the temporal and spatial distribution and structure of swarm cells of A. c. cerana. It establishes the basis for predicting the time and position of appearing swarm cells, thus allowing for a more precise determination of the shape and size of queen-cell punch and the ideal position of a cell cup on the bar of queen cup frames in artificial queen rearing

    Diagnosis of covert coarctation of the aorta in adolescents

    ObjectivesBy reviewing the diagnostic process for adolescents with coarctation of the aorta (CoA) in our institution, we analyzed the reasons for delayed diagnosis of CoA. We also proposed a diagnostic protocol to improve the detection rate of CoA.MethodsIn this retrospective study, we included 48 patients aged 12–18 years who were diagnosed with CoA in our hospital from January 2000 to November 2022. Clinical data from involved cases in local hospitals and our institution were collected.ResultsAll patients had blood pressure (BP) measurements in upper and lower extremities in our institution. They all had hypertension, 29 (60.4%) of whom had known histories of the same. BP in the upper limbs of 47 (97.9%) patients was ≥20 mmHg higher than that in the lower limbs, and BP in the upper limb of 1 (2.1%) patient was greater than 0 and less than 20 mmHg than that in the lower limb. Echocardiography (ECHO) was performed in all patients, computed tomography (CT) or magnetic resonance imaging (MRI) was performed in 44 patients (91.7%). There were 38 (79.2%) patients who visited local hospitals. Among them, a total of 20 (52.6%) patients had their right upper extremity BP measured, 18 (47.4%) only had their left upper extremity BP measured, and 16 (42.1%) had their lower extremity BP measured. ECHO was performed in 27 (56.2%) patients and CT/MRI was performed in 18 (37.5%) patients. The detection rate for CT/MRI was 100%, and those of ECHO were 72.9% and 18.5% at our institution and a local hospital, respectively. Forty-eight (100%) and 23 (60.5%) patients were detected in our institution and local hospitals (P < 0.0001).ConclusionWe recommend measuring BP in the bilateral upper extremities. Measurement of BP in the lower extremities is recommended if hypertension is diagnosed. MRI/CT is recommended when BP in the upper extremity is greater than that in the lower extremity

    Melittin promotes dexamethasone in the treatment of adjuvant rheumatoid arthritis in rats

    Background: Rheumatoid arthritis (RA) is an erosive-destructive inflammation of the joints, and the chronic, long-term stiffness and deformation induced by RA are some of the symptoms of arthritis that are difficult to treat. Dexamethasone (DEX) and melittin (MLT) are two interesting anti-inflammatory substances, both of which possess anti-inflammatory effects exerted through the suppression of the immune system. The purpose of this study was to explore the role of MLT in the treatment of RA by DEX as well as to clarify the influence of MLT on the efficacy and side effects of DEX.Method: The rats were injected with Complete Freund’s Adjuvant (CFA) to induce arthritis, followed by treatment with different doses of DEX and/or MLT. The relevant indexes of paw inflammation were determined, and the appetite, growth status, arthritis status, cytokine levels, and organ coefficient of the rats were evaluated. In addition, the paraffin sections of the joint tissues were prepared to analyze the pathological changes.Result: DEX exhibited side effects, notably hindering feed intake and growth, and inducing immune organ lesions in the rats. MLT significantly reduced the side effects of DEX and promoted its efficacy. DEX in combination with MLT demonstrated a synergistic efficacy in RA treatment, showing advantages in detumescence reduction, pro-inflammatory cytokine inhibition, and joint internal pathological improvement.Conclusion: Thus, MLT promoted the efficacy of DEX in adjuvant RA treatment in rats, offering an approach to reduce the use dosage and side effects of DEX

    Phylogeography of Apis cerana populations on Hainan island and southern mainland China revealed by microsatellite polymorphism and mitochondrial DNA

    International audienceAbstractIn this study, we reexamined the patterns of divergence and phylogeographic structure among Apis cerana populations on Hainan island and southern mainland China. Analysis of 10 microsatellite loci from 12 geographically distinct A. cerana populations identified a total of 151 alleles, with a range of 7 to 24 at each locus. The level of genetic variation (observed heterozygosity) within each population ranged from 0.603 to 0.661, while variation within populations contributed most (93.78–98.84 %) of the molecular variance. Microsatellite data revealed three differentiated groups, one including all the mainland populations while the other two containing one of and the other three island populations, respectively. Severe genetic introgressions from mainland China into Hainan island were apparent, and these were characterized at both population and individual levels using combined analysis of microsatellites and mitochondrial DNA

    Dihydrofolate reductase-like protein inactivates hemiaminal pharmacophore for self-resistance in safracin biosynthesis

    Dihydrofolate reductase (DHFR), a housekeeping enzyme in primary metabolism, has been extensively studied as a model of acid-base catalysis and a clinic drug target. Herein, we investigated the enzymology of a DHFR-like protein SacH in safracin (SAC) biosynthesis, which reductively inactivates hemiaminal pharmacophore-containing biosynthetic intermediates and antibiotics for self-resistance. Furthermore, based on the crystal structure of SacH−NADPH−SAC-A ternary complexes and mutagenesis, we proposed a catalytic mechanism that is distinct from the previously characterized short-chain dehydrogenases/reductases-mediated inactivation of hemiaminal pharmacophore. These findings expand the functions of DHFR family proteins, reveal that the common reaction can be catalyzed by distinct family of enzymes, and imply the possibility for the discovery of novel antibiotics with hemiaminal pharmacophore

    Impact of Brood Cell Cocoons on Metal Accumulation and CYP450 Detoxification Gene Expression in <i>Apis cerana cerana</i>

    Honey bees play a critical role as pollinators. However, their reproduction success and survival face severe threats due to the deterioration of their living environment. Notably, environmental conditions during their preimaginal stage inside brood cells can influence their immune capabilities and overall health after emergence. During the in-cell developmental stage, workers are in close contact with cocoons, which can become a source of stress due to accumulated metals. To investigate this potential threat, experiments were conducted to examine the impact of cocoons in brood cells used to rear different generations on the metal content and detoxification gene expression levels in Apis cerana cerana. Our findings indicated significant differences in the layers, weight, base thickness, and metal contents like Cr, Cd, Pb, Mn, Ni, and As of cocoons in multi-generation brood cells compared to single-generation brood cells. These increases led to significant elevations in metal levels and upregulations of the four CYP450 detoxification genes in both six-day-old larvae and newly emerged workers. In conclusion, this study highlights the negative impact of cocoons in multi-generation brood cells on bee health and provides evidence supporting the development of rational apiculture management strategies for ecosystem stability

    A Mouse Model of Enterovirus D68 Infection for Assessment of the Efficacy of Inactivated Vaccine

    In recent years, enterovirus D68 (EVD68) has been reported increasingly to be associated with severe respiratory tract infections and acute flaccid myelitis (AFM) in children all over the world. Yet, no effective vaccines or antiviral drugs are currently available for EVD68. Although several experimental animal models have been developed, immunogenicity and protective efficacy of inactivated EVD68 vaccines has not been fully evaluated. To promote the development of vaccines, we established an Institute of Cancer Research (ICR) suckling mouse model of EVD68 infection in this study. The results showed that ICR neonatal mice up to about nine days of age were susceptible to infection with EVD68 clinical strain US/MO/14-18947 by intraperitoneal injection. The infected mice exhibited progressive limb paralysis prior to death and the mortality of mice was age- and virus dose-dependent. Tissue viral load analysis showed that limb muscle and spinal cord were the major sites of viral replication. Moreover, histopathologic examination revealed the severe necrosis of the limb and juxtaspinal muscles, suggesting that US/MO/14-18947 has a strong tropism toward muscle tissues. Additionally, β-propiolactone-inactivated EVD68 vaccine showed high purity and quality and induced robust EVD68-specific neutralizing antibody responses in adult mice. Importantly, results from both antisera transfer and maternal immunization experiments clearly showed that inactivated EVD68 vaccine was able to protect against lethal viral infection in the mouse model. In short, these results demonstrate the successful establishment of the mouse model of EVD68 infection for evaluating candidate vaccines against EVD68 and also provide important information for the development of inactivated virus-based EVD68 vaccines
