    Effect of inoculum age on kinetic of biomass formation and phenolic accumulations in Larrea divaricata Cav. cell suspension culture

    This work focuses on the analysis of the effect of inoculum age on the kinetic of biomass formation and phenolic accumulation in Larrea divaricata Cav. cell suspension culture. L. divaricata plant cell cultures (callus and cell suspension culture) successfully produce interesting phenolic compounds, being the main the lignan Nordihydroguaiaretic acid. The findings revealed that the inoculum age is an important factor on the kinetic biomass formation and phenolic compounds production in L. divaricata cell suspension culture. Four to seven month - inoculum age were suitable to establishment cell suspension culture, not only for biomass but also for the phenolic compounds production. On the contrary, when the age of the callus was over eighth month, no stimulation of phenolic compounds biosynthesis was observed.Fil: Palacio, Lorena. Provincia de Córdoba.     In vitro propagation and stoloniferous stem formation of Thelesperma megapotamicum

    An efficient “in vitro” propagation protocol for Thelesperma megapotamicum (Spreng.) Kuntnze was established. Nodal segments were cultured in vitro with different combination and concentration of growth regulators. Microshoots (60-days) cultured in Murashige and Skoog (MS) with the addition of 4 mg/l 6-benzyl adenine (BA) were used as explants and incubated on a half salt concentrations MS/2 medium supplemented with different combination of indol butyric acid (IBA) and sucrose for 120 days. Explants cultured on a half of Murashige and Skoog salts (MS/2) with the addition of 1,0 mg/L-40 g/L and 1,5mg/L - 30g/L of IBA and sucrose gave shoot number and principal stem length as the control medium with higher stoloniferous stem percentage or development of shoots in stoloniferous stem respectively.Fil: Figueroa, A.     “In vitro” Hedeoma multiflorum Benth propagation in response to different nutritional conditions

    An efficient protocol for in vitro propagation of Hedeoma multiflorum Benth (“Tomillo de las Sierras”) was established. To determine the effect of different medium and plant growth regulators on the growth rates and essential oil production, nodal segments from in vitro 90-days old plantlets were incubated on the following media: 1) SH (Schenk and Hildebrandt); 2) WP (Lloyd and McCown); 3) B5 (Gamborg). All the media studied were supplemented with different combinations and consentrations of growth regulators: naphtaleneacetic acid (NAA) and 6-benzyl adenine (BA). The optimal shoot length and number of nodes of the plantlets, showing no statistical difference, were obtained in WP with the addition of 2.69:0.5 μM, 0.5:0.5 and 2.69:0.05 μM of NAA:BA and in absence of these growth regulators (control). The best multiple shoot medium with no statistical difference, corresponded to SH with the addition of 0.5:2.25 μM and B5 with 0.05:2.25, 0.5:2.25 and 2.69:2.25 μM of NAA:BA. The major essential oil determined by GC/MS, were common in all the plantlets grown in all the media being pulegone (62-64%) , menthone (25-28 %) and isomenthone (2-2,3%).Fil: Diaz S..     In Vitro Propagation of Muña-Muña ( Clinopodium odorum (Griseb.) Harley)

    A micropropagation protocol was developed which may assist in the safeguarding and augmentation of dwindling natural populations of Clinopodium odorum (Griseb.) Harley, a critically and endangered medicinal plant. Factors affecting culture initiation bud sprouting and growth, rooting, and acclimatization were studied, using nodal segments of in vitro germinated seedling as primary explants on six media supplemented with different concentrations and combinations of 6-benzylaminopurine (BAP) (0.5?1.5 and 2-Naphthalene acetic acid (NAA) (0.5?1.5). Best results for culture initiation with sustainable multiplication rates (100%) were obtained onWP medium without any growth regulator.WP with the addition of 0.5 : 1 or 0.5 : 1.5) of BAP and NAA promoted a higher elongation; however, the optimum number of nodes were obtained in plantlets grown on 1/2 MS with the addition of 1 : 1.5 of BAP and NAA. Culture of sectioned individual nodes transferred to the media with different rates of BAP and NAA 1/2 MS-9 (1.5 : 1.5), SH-8 (1.5 : 1.0), and 1/2 B5-4 (1.0 : 0.5) media resulted in no proliferated shoots. The in vitro plants were successfully acclimatized garden soil and sand (2 : 1) in the greenhouse, with over 90% survival rate. The in vitro-grown plants could be transferred to ex vitro conditions and the efficacy in supporting ex vitro growth was assessed, with a view to develope longer-term strategies for the transfer and reintroduction into natural habitats.Fil: Diaz, María Soledad.
