    Kulathadi Kwatha and Talisapatradi Churna In Tamaka Shwasa (Bronchial Asthma) : An Open Label, Single Arm Clinical Study

    Background: Tamaka Shwasa is a Pranavaha Sroto Vikara, with predominant Dosha, Kapha and Vata and the site of origin is Pittasthana. The condition has similarity in clinical presentation with Bronchial Asthma. It leads to recurrent episodes of breathlessness, wheezing, cough and tightness of chest. Shamshodhana and Shamshamana are the two treatment modalities explained in Tamaka Shwasa. Objectives: To clinically assess the combined effectiveness of Kulathadi Kwatha and Talisapatradi Churna in Tamaka Shwasa. Materials and methods: This study was carried out by Shamanaushadhis viz; Kulathadi Kwatha and Talisapatradi Churna in 30 subjects of either sex in between the age 18-40 years, for 30 days, followed by follow up on 45th day. Collected data were tabulated and analysed using SPSS (Statistical package for social sciences) version 20 by using appropriate statistical test. Results: There was statistically significant improvement observed in the signs and symptoms of Tamaka Shwasa and Peakflow Meter with the P value <0.05. Conclusion: The trial drug Kulathadi Kwatha and Talisapatradi Churna was found beneficial in symptoms of Tamaka Shwasa

    SARAS 2 Constraints on Global 21 cm Signals from the Epoch of Reionization

    Spectral distortions in the cosmic microwave background over the 40--200~MHz band are imprinted by neutral hydrogen in the intergalactic medium prior to the end of reionization. This signal, produced in the redshift range z=634z = 6-34 at the rest frame wavelength of 21 cm, has not been detected yet; and poor understanding of high redshift astrophysics results in a large uncertainty in the expected spectrum. The SARAS~2 radiometer was purposely designed to detect the sky-averaged 21-cm signal. The instrument, deployed at the Timbaktu Collective (Southern India) in April--June 2017, collected 63~hr of science data, which were examined for the presence of the cosmological 21-cm signal. In our previous work the first-light data from SARAS~2 radiometer were analyzed with Bayesian likelihood-ratio tests using 264264 plausible astrophysical scenarios. In this paper we re-examine the data using an improved analysis based on the frequentist approach and forward modeling. We show that SARAS~2 data rejects 27 models, out of which 25 are rejected at a significance >5σ>5\sigma. All the rejected models share the scenario of inefficient heating of the primordial gas by the first population of X-ray sources along with rapid reionization

    First Results on the Epoch of Reionization from First Light with SARAS 2

    Long wavelength spectral distortions in the Cosmic Microwave Background arising from the 21-cm transition in neutral Hydrogen are a key probe of Cosmic Dawn and the Epoch of Reionization. These features may reveal the nature of the first stars and ultra-faint galaxies that transformed the spin temperature and ionization state of the primordial gas. SARAS~2 is a spectral radiometer purposely designed for precision measurement of these monopole or all-sky global 21-cm spectral distortions. We use 63~hr night time observing of the radio background in the frequency band 110-200~MHz with the radiometer deployed at the Timbaktu Collective in Southern India to derive likelihoods for plausible redshifted 21-cm signals predicted by theoretical models. First light with SARAS 2 disfavors the class of models that feature weak X-ray heating (with fX0.1f_X \leq 0.1) and rapid reionization (with peak dTbdz120 mK per unit redshift interval\frac{dT_b}{dz} \geq 120~\textrm{mK per unit redshift interval} )

    Home advantage in the Winter Paralympic Games 1976–2014

    Purpose: There is a limited amount of home advantage research concerned with winter sports. There is also a distinct lack of studies that investigate home advantage in the context of para-sport events. This paper addresses this gap in the knowledge by examining home advantage in the Winter Paralympic Games. Methods: Using a standardised measure of success, we compared the performances of host nations at home with their own performances away from home between 1976 and 2014. Both country level and individual sport level analysis is conducted for this time period. Comparisons are also drawn with the Winter Olympic Games since 1992, the point from which both the Winter Olympic Games and the Winter Paralympic Games have been hosted by the same nations and in the same years. Results: Clear evidence of a home advantage effect in the Winter Paralympic Games was found at country level. When examining individual sports, only alpine skiing and cross country skiing returned a significant home advantage effect. When comparing home advantage in the Winter Paralympic Games with the Winter Olympic Games for the last seven host nations (1992–2014), we found that home advantage was generally more pronounced (although not a statistically significant difference) in the case of the former. Conclusion: The causes of home advantage in the Winter Paralympic Games are unclear and should be investigated further

    An investigation of home advantage in the Summer Paralympic Games

    Purpose: There is a paucity of home advantage research set in the context of para-sport events. It is this gap in the knowledge that this paper addresses by investigating the prevalence and size of home advantage in the Summer Paralympic Games. Methods: Using a standardised measure of success, we compared the performances of nations when competing at home with their own performances away from home in the competition between 1960 and 2016. Both country level and individual sport level analysis was conducted for this time frame. A Wilcoxon signed rank test was used to determine whether there was a genuine difference in nations' performance under host and non-host conditions. Spearman's rank-order correlation was run to assess the relationship between nation quality and home advantage. Results: Strong evidence of a home advantage effect in the Summer Paralympic Games was found at country level (p 0.10). Conclusion: While our results confirm that home advantage is prevalent in the Summer Paralympic Games at an overall country level and within specific sports, they do not explain fully why such an effect does exist. Future studies should investigate the causes of home advantage in the competition and also draw comparisons with the Summer Olympic Games in order to explore any differences between para-sport events and able-bodied events

    Premolis semirufa (Walker, 1856) Envenomation, Disease Affecting Rubber Tappers of the Amazon: Searching for Caterpillar-Bristles Toxic Components

    Pararama, the popular name of the larval form of the moth Premolis semirufa inhabits rubber plantations in the Amazon region and the accidental contact of the skin with the caterpillar's bristles or cocoons results in immediate and intense heat, pain, edema, and itching. In many cases a chronic inflammatory reaction with immobilization of the joints occurs. The current study has evaluated the biological and immunochemical characteristics of the Pararama caterpillar bristles extract. Electrophoretic analysis showed the presence of several components, including a very intense 82 kDa band. This latter component was endowed with intense gelatinolytic activity, as observed in zymography assays. Further analysis revealed that the extract also contained hyaluronidase activity but is devoid of phospholipase A2 activity. In vivo assays, using mice, showed that the extract was not lethal, but caused significant edema and induced intense infiltration of inflammatory cells to the envenomation site. The extract also induced high specific antibody titers, but no autoantibodies were detected. The data obtained, so far, demonstrate the existence of a mixture of different enzymes in the bristles of Premolis semirufa caterpillar, which can act together in the generation and development of the clinical manifestations of the Pararama envenomation

    Mycobacteria Exploit Host Hyaluronan for Efficient Extracellular Replication

    In spite of the importance of hyaluronan in host protection against infectious organisms in the alveolar spaces, its role in mycobacterial infection is unknown. In a previous study, we found that mycobacteria interact with hyaluronan on lung epithelial cells. Here, we have analyzed the role of hyaluronan after mycobacterial infection was established and found that pathogenic mycobacteria can grow by utilizing hyaluronan as a carbon source. Both mouse and human possess 3 kinds of hyaluronan synthases (HAS), designated HAS1, HAS2, and HAS3. Utilizing individual HAS-transfected cells, we show that HAS1 and HAS3 but not HAS2 support growth of mycobacteria. We found that the major hyaluronan synthase expressed in the lung is HAS1, and that its expression was increased after infection with Mycobacterium tuberculosis. Histochemical analysis demonstrated that hyaluronan profoundly accumulated in the granulomatous legion of the lungs in M. tuberculosis-infected mice and rhesus monkeys that died from tuberculosis. We detected hyaluronidase activity in the lysate of mycobacteria and showed that it was critical for hyaluronan-dependent extracellular growth. Finally, we showed that L-Ascorbic acid 6-hexadecanoate, a hyaluronidase inhibitor, suppressed growth of mycobacteria in vivo. Taken together, our data show that pathogenic mycobacteria exploit an intrinsic host-protective molecule, hyaluronan, to grow in the respiratory tract and demonstrate the potential usefulness of hyaluronidase inhibitors against mycobacterial diseases

    High and Low Molecular Weight Hyaluronic Acid Differentially Regulate Human Fibrocyte Differentiation

    Following tissue injury, monocytes can enter the tissue and differentiate into fibroblast-like cells called fibrocytes, but little is known about what regulates this differentiation. Extracellular matrix contains high molecular weight hyaluronic acid (HMWHA; ∼2×10(6) Da). During injury, HMWHA breaks down to low molecular weight hyaluronic acid (LMWHA; ∼0.8-8×10(5) Da).In this report, we show that HMWHA potentiates the differentiation of human monocytes into fibrocytes, while LMWHA inhibits fibrocyte differentiation. Digestion of HMWHA with hyaluronidase produces small hyaluronic acid fragments, and these fragments inhibit fibrocyte differentiation. Monocytes internalize HMWHA and LMWHA equally well, suggesting that the opposing effects on fibrocyte differentiation are not due to differential internalization of HMWHA or LMWHA. Adding HMWHA to PBMC does not appear to affect the levels of the hyaluronic acid receptor CD44, whereas adding LMWHA decreases CD44 levels. The addition of anti-CD44 antibodies potentiates fibrocyte differentiation, suggesting that CD44 mediates at least some of the effect of hyaluronic acid on fibrocyte differentiation. The fibrocyte differentiation-inhibiting factor serum amyloid P (SAP) inhibits HMWHA-induced fibrocyte differentiation and potentiates LMWHA-induced inhibition. Conversely, LMWHA inhibits the ability of HMWHA, interleukin-4 (IL-4), or interleukin-13 (IL-13) to promote fibrocyte differentiation.We hypothesize that hyaluronic acid signals at least in part through CD44 to regulate fibrocyte differentiation, with a dominance hierarchy of SAP>LMWHA≥HMWHA>IL-4 or IL-13
