    Prevalence and severity of depression among undergraduate students in Karachi, Pakistan: a cross sectional study

    Purpose: To highlight the prevalence and severity of depression among undergraduate students in public and private universities in Karachi, Pakistan.Methods: In this cross-sectional study a total of 408 undergraduate students from both public and private universities in Karachi, Pakistan completed Zung Self-Rating Depression Scale (SDS) questionnaire to appraise the presence and extent of depression among the participants. Their sociodemographic characteristics such as age, gender and course of study as well as drug use data were also collected and analyzed.Results: Depression prevalence was 53.43 % (38.07 % for males and 61.00 % for females). A significant disparity in the prevalence of depression across ethnicity was observed. Less than 50 % (n = 163) of the students were satisfied with their duration of degree course work while 111 (27.20 %) participants were not sure about this. The intensity of depression increased with declining satisfaction level (Gamma = 0.264, p = 0.001) which affected the performance and results of students during their studies. A majority of the participants (89 %, n = 358) of our study have never used medicines to alter their mood.Conclusion: The findings of this study highlight the importance of understanding the unique strains and mental health effect of university education on undergraduate students, especially female students.Keywords: Depression, Undergraduate students, Mood alteration, Zung self-rating depression scale (SDS), Mental health, Risk assessmen

    BET Bromodomain Inhibition as a Therapeutic Strategy to Target c-Myc

    SummaryMYC contributes to the pathogenesis of a majority of human cancers, yet strategies to modulate the function of the c-Myc oncoprotein do not exist. Toward this objective, we have targeted MYC transcription by interfering with chromatin-dependent signal transduction to RNA polymerase, specifically by inhibiting the acetyl-lysine recognition domains (bromodomains) of putative coactivator proteins implicated in transcriptional initiation and elongation. Using a selective small-molecule bromodomain inhibitor, JQ1, we identify BET bromodomain proteins as regulatory factors for c-Myc. BET inhibition by JQ1 downregulates MYC transcription, followed by genome-wide downregulation of Myc-dependent target genes. In experimental models of multiple myeloma, a Myc-dependent hematologic malignancy, JQ1 produces a potent antiproliferative effect associated with cell-cycle arrest and cellular senescence. Efficacy of JQ1 in three murine models of multiple myeloma establishes the therapeutic rationale for BET bromodomain inhibition in this disease and other malignancies characterized by pathologic activation of c-Myc.PaperFlic

    Cereal Domestication and Evolution of Branching: Evidence for Soft Selection in the Tb1 Orthologue of Pearl Millet (Pennisetum glaucum [L.] R. Br.)

    BACKGROUND: During the Neolithic revolution, early farmers altered plant development to domesticate crops. Similar traits were often selected independently in different wild species; yet the genetic basis of this parallel phenotypic evolution remains elusive. Plant architecture ranks among these target traits composing the domestication syndrome. We focused on the reduction of branching which occurred in several cereals, an adaptation known to rely on the major gene Teosinte-branched1 (Tb1) in maize. We investigate the role of the Tb1 orthologue (Pgtb1) in the domestication of pearl millet (Pennisetum glaucum), an African outcrossing cereal. METHODOLOGY/PRINCIPAL FINDINGS: Gene cloning, expression profiling, QTL mapping and molecular evolution analysis were combined in a comparative approach between pearl millet and maize. Our results in pearl millet support a role for PgTb1 in domestication despite important differences in the genetic basis of branching adaptation in that species compared to maize (e.g. weaker effects of PgTb1). Genetic maps suggest this pattern to be consistent in other cereals with reduced branching (e.g. sorghum, foxtail millet). Moreover, although the adaptive sites underlying domestication were not formerly identified, signatures of selection pointed to putative regulatory regions upstream of both Tb1 orthologues in maize and pearl millet. However, the signature of human selection in the pearl millet Tb1 is much weaker in pearl millet than in maize. CONCLUSIONS/SIGNIFICANCE: Our results suggest that some level of parallel evolution involved at least regions directly upstream of Tb1 for the domestication of pearl millet and maize. This was unanticipated given the multigenic basis of domestication traits and the divergence of wild progenitor species for over 30 million years prior to human selection. We also hypothesized that regular introgression of domestic pearl millet phenotypes by genes from the wild gene pool could explain why the selective sweep in pearl millet is softer than in maize

    Evaluation of a real-time virtual intervention to empower persons living with HIV to use therapy self-management: study protocol for an online randomized controlled trial

    Background: Living with HIV makes considerable demands on a person in terms of self-management, especially as regards adherence to treatment and coping with adverse side-effects. The online HIV Treatment, Virtual Nursing Assistance and Education (Virus de I'immunodeficience Humaine-Traitement Assistance Virtuelle Infirmiere et Enseignement; VIH-TAVIE (TM)) intervention was developed to provide persons living with HIV (PLHIV) with personalized follow-up and real-time support in managing their medication intake on a daily basis. An online randomized controlled trial (RCT) will be conducted to evaluate the efficacy of this intervention primarily in optimizing adherence to combination anti-retroviral therapy (ART) among PLHIV.Methods/design: A convenience sample of 232 PLHIV will be split evenly and randomly between an experimental group that will use the web application, and a control group that will be handed a list of websites of interest. Participants must be aged 18 years or older, have been on ART for at least 6 months, and have internet access. The intervention is composed of four interactive computer sessions of 20 to 30 minutes hosted by a virtual nurse who engages the PLHIV in a skills-learning process aimed at improving self-management of medication intake. Adherence constitutes the principal outcome, and is defined as the intake of at least 95% of the prescribed tablets. The following intermediary measures will be assessed: self-efficacy and attitude towards antiretroviral medication, symptom-related discomfort, and emotional support. There will be three measurement times: baseline (T0), after 3 months (T3) and 6 months (T6) of baseline measurement. The principal analyses will focus on comparing the two groups in terms of treatment adherence at the end of follow-up at T6. An intention-to-treat (ITT) analysis will be carried out to evaluate the true value of the intervention in a real context.Discussion: Carrying out this online RCT poses various challenges in terms of recruitment, ethics, and data collection, including participant follow-up over an extended period. Collaboration between researchers from clinical disciplines (nursing, medicine), and experts in behavioral sciences information technology and media will be crucial to the development of innovative solutions to supplying and delivering health services

    Construction and Validation of the Research Misconduct Scale for Social Science University Students

    The current study aims to construct and validate a measure of research misconduct for social science university students. The research is comprised of three studies; Study I presents the scale construction in three phases. In Phase I, the initial pool of items was generated by reviewing the literature and considering the results of semi-structured interviews. Phase II involved a psychometric cleaning of items, after which 38 items were retained. In Phase III, those 38 items were proposed to 652 university students, and data were exposed to exploratory factor analysis, which extracted a one-factor structure with 15 items and 55.73% variance. Study II confirmed the factorial structure of the scale using an independent sample (N = 200) of university students. Confirmatory factor analysis of the scale demonstrates a good model fit to the data with the one-factor structure established through the exploratory factor analysis. The scale exhibits good internal consistency, with a Cronbach’s alpha of 0.95. Study III involves validation of the scale, with evidence for convergent validity collected from a sample of university students (N = 200). The results reveal that the research misconduct scale has significant positive correlations with academic stress and procrastination and a significant negative correlation with academic achievement. The obtained convergent validity testifies that the scale can be considered a psychometrically sound instrument to measure research misconduct among social science university students

    Subsequent Effect of Lysine Regimens on Egg Characteristics of Native Aseel Chicken

    ABSTRACT This study was planned to evaluate subsequent effects of rearing under various lysine regimens on egg quality traits in Peshawari, Mushki, Mianwali and Lakha varieties of native Aseel. Ninety six pullets and 12 cockerels (24♀ and 3♂) from each variety were randomly chosen, placed in three-tiered cage units of equal space and standard conditions were followed for their maintenance. These birds were reared on three lysine regimens (L1, L2 and L3), wherein L1 containing 1.3% lysine was given in one phase from week 1-6 of age, L2 regimens entailing 1.4 and 1.2% lysine was smeared in two phases from week 1-3 and 4-6 of age, while in L3 regimen, lysine @ 1.5, 1.3 and 1.1% in diet was fed in three phases from week 1-2, 3-4 and 5-6 of age, respectively. Data were analyzed using two-way factorial ANOVA technique in SAS 9.1under randomized complete block design and significant means were compared through Duncan’s Multiple Range Test. Statistical analysis showed that weight of egg and shell, albumen weight, diameter and index, yolk weight and Haugh units (p≀0.05) improved significantly in L3 in Peshawari Aseel depicting the better egg quality traits. It was concluded that L3 regimen has positive impact on egg characteristics of Aseel chicken

    Different Selection Strategies for the Improvement of the Growth Performance and Carcass Traits of Japanese Quails

    ABSTRACT The present study evaluated different selection strategies to improve the growth performance and carcass traits of Japanese quails. To this end, 540 Japanese quails previously selected for high body weight at three ages (10, 12 and 14 weeks) from four close-bred flocks (Major, Kaleem, Saadat and Zahid) were subjected to three selection strategies: pedigree-based, mass selection or random-bred (controls). For pedigree-based selection, 108 birds (4 close-bred flocks × 3 age groups × 9 replicates × 1 bird each) with full pedigree and selected for high body weight were used, whereas in mass selection 324 birds (4 close-bred flocks × 3 age groups × 9 replicates × 3 birds each) with high body weight were selected to be the parents of next generation. Random-bred controls included 108 birds (4 close-bred flocks × 3 age groups × 9 replicates × 1bird each) which were not previously submitted to selection. The effects of selection strategies of Japanese quail parents from four close-bred flocks (CBF) at three ages on the performance growth and carcass traits of their progenies were measured. The parent flocks were selected at three ages (10, 12 and 14 weeks) and reared for 20 weeks, and their progenies were reared for four weeks. The progeny of pedigree-based selected parents presented better feed intake (g), body weight gain (g), feed conversion ratio, live and carcass weights (g), and carcass yield. The progeny of 10- and 14-week-old parents showed better growth performance and carcass traits, respectively. Moreover, there was no influence of CBF on growth and carcass traits. It was concluded that pedigree-based selection had pronounced effect on the growth performance and carcass traits of the progeny compared with mass selection and random breeding. It is suggested that parental ages of 10 and 14 weeks of age promote better progeny growth performance and carcass traits
